    A Nitric Oxide-Donor Furoxan Moiety Improves the Efficacy of Edaravone against Early Renal Dysfunction and Injury Evoked by Ischemia/Reperfusion

    Edaravone (5-methyl-2-phenyl-2,4-dihydro-3H-pyrazol-3-one, EDV) is a free-radical scavenger reduces organ ischemic injury. Here we investigated whether the protective effects of EDV in renal ischemia/reperfusion (I/R) injury may be enhanced by an EDV derivative bearing a nitric oxide- (NO-) donor furoxan moiety (NO-EDV). Male Wistar rats were subjected to renal ischemia (45 minutes), followed by reperfusion (6 hours). Administration of either EDV (1.2–6–30 µmol/kg, i.v.) or NO-EDV (0.3–1.2–6 µmol/kg, i.v.) dose-dependently attenuated markers of renal dysfunction (serum urea and creatinine, creatinine clearance, urine flow, urinary N-acetyl-β-D-glucosaminidase, and neutrophil gelatinase-associated lipocalin/lipocalin-2). NO-EDV exerted protective effects in the dose-range 1.2–6 µmol/kg, while a higher dose (30 µmol/kg) was needed to obtain protection by EDV. Both EDV and NO-EDV modulated tissue markers of oxidative stress and lipid peroxidation. NO-EDV, but not EDV, activated endothelial NO synthase (NOS) and blunted I/R-induced upregulation of inducible NOS, secondary to modulation of Akt and NF-κB activation, respectively. Besides NO-EDV administration inhibited I/R-induced IL-1β, IL-18, IL-6, and TNF-α overproduction. Overall, these findings demonstrate that the NO-donor moiety contributes to the protection against early renal I/R injury and suggest that NO-donor EDV codrugs are worthy of additional study as innovative pharmacological tools.Fil: Chiazza, Fausto. O que se quer é identificar e discutir quais são, na concepção do autor, as potencialidades e as limitações da tradução na sua relação com a literatura.The present work intends to discuss the thought of Italo Calvino concerning translation of literary works.     Synthesis of NO-donor bisphosphonates and their in-vitro action on bone resorption

    A new class of bisphosphonates containing nitrooxy NO-donor functions has been developed. The products proved to display affinity for hydroxyapatite. Injection of (99m)Tc-labeled derivatives 11 and 18 into male rats showed a preferential accumulation of the compounds in bone as compared to blood and muscles. The products were found to inhibit the differentiation of pre-osteoclasts to functional osteoclasts induced by receptor activator of NF-kappaB ligand (RANKL), through a prevalent NO-dependent mechanism

    Anti-pseudomonas activity of 3-nitro-4-phenylfuroxan

    Pseudomonas aeruginosa is a microorganism that is well adapted to both clinical and industrial settings, where it can form adherent communities that are difficult to eradicate. New anti-Pseudomonas compounds and strategies are necessary, as the current antimicrobial approaches for the inhibition of biofilm formation and, above all, the eradication of formed biofilms are ineffective. Compounds that belong to the furoxan family, which are well-known NO donors, have recently been shown to display anti-Pseudomonas activity. The present study investigates three furoxan compounds that are substituted at the hetero-ring with electron-withdrawing groups (NO2, CN, CONH2) for their effects on P. aeruginosa PAO1 growth and biofilm formation/dispersal. Of the furoxans tested, only 3-nitro-4-phenylfuroxan (KN455) inhibited the growth of suspended P. aeruginosa PAO1 cultures. Furthermore, KN455 inhibited the formation of both younger and older biofilms with very high yields and thus proved itself to be toxic to planktonic subpopulations. It also displayed moderate eradicating power. The activity of KN455 does not appear to be related to its capacity to release small amounts of NO. Interestingly, the isomer 4-nitro- 3-phenylfuroxan (KN454), included for comparison, displayed a comparable antibiofilm rate, but did not show the same antimicrobial activity against suspended cells and planktonic subpopulations. While hypotheses as to the mechanism of action have been formulated, further investigations are necessary to shed light onto the antimicrobial activity of this furoxan

    By discussing the essays collected in the volume, Roberta Dreon's paper focuses on the reasons that justify the very idea of a pragmatist aesthetic. This is done by considering that the association between the traditional, contemplative, disinterested, and anti-instrumental conception of aesthetic experience seems to preclude the possibility of characterizing it in practical or pragmatic terms. The author argues that this is achieved on the one hand by a rethinking of the very notion of the "aesthetic" found in the philosophies of James and Dewey. This allows for supporting the idea that artistic practices are grounded in ordinary experience, and particularly in their aesthetic-qualitative aspects. On the other hand, the author argues that Dewey's aesthetics was convincingly pragmatist to the extent that it lucidly focused on the consequences of the autonomist conception of art and proposed a continuist, meliorist, and pluralist alternative capable of providing effective contributions to democratic and inclusive development

    Secondo Putnam argomentare sulla natura della razionalità è l’attività per eccellenza dei filosofi. Sulla traccia di Putnam, l’autore esamina le principali teorie della razionalità presenti nel pensiero contemporaneo. Tali concezioni hanno il difetto di essere unilaterali, mentre la nozione di razionalità si rivela complessa, quindi una teoria della razionalità è possibile, benché non possa essere definitiva. In seguito l’autore cerca di individuare le caratteristiche fondamentali che competono alla razionalità, in opposizione tanto alla concezione positivista quanto al relativismo.According to Putnam, arguing about the nature of rationality is the typical task of philosophers. Following Putnam, in this paper the author examines the main theories of rationality to be found in contemporary thought. Whereas such views betray their own one-sidedness, the idea of rationality is very complicated. As a consequence, a theory of rationality is possible, but cannot be definitive. Furthermore, the author tries to highlight the chief features pertaining to rationality, opposing positivsm as well as relativism
