    Biomarkers, human health

The term “biomarker” comes from the Greek term “βιο-“, meaning life, in combination with “mark”, Old English of “mearcere”, meaning “writer, notary”, or literally “one who marks”. In scientific literature the word first emerged in the 1970s, but the concept has been part of biomedical research for decades. Various definitions were established over the years, the simplest describes it as a “biological endpoint (or change of) measured in the organism” used as an indicator in clinical practice, drug development and risk assessment research. As a result, several subtypes have been defined according to the field/subject of the study in question. This report of the supervised educational practice for the recognition of the Master's degree in Pre-School Education is the culmination of a long process of growth of the master student at a personal and professional level, and proposes to address, critically and reflectively, the process developed during the professional internships carried out in the nursery school and pre-school education. It should be noted that this training included the integration of an articulation between theoretical and legal references, in a continuous perspective of research and reflection. All the experiences in practice, were supported by a dynamic and spiral cycle of observation, planning, action, assessment and reflection processes, which constitute the action research methodology, contributed towards an innovative, reflective and meaningful action for all those involved. The student developed professional knowledge that was transformed into an action supported by participatory pedagogies, involving children as active subjects in the construction of their learning, always based on their interests and needs, in order to build an environment that promotes significant learning, from a global and integrative perspective. In addition to the above, all the actions developed were anchored in a critical and reflective attitude that allowed the student to understand problems emerging from educational practice, investigating them and reflecting on, in and for action (Schön, 1987, cited by Alarcão, 1996), developing a transformation of herself, as a subject in training and providing real opportunities for learning and growth. The sharing of experiences and the collaboration established between the dyad, the cooperating educators and the institutional supervisors contributed to the personal and professional development of the master's student, as a future Early Childhood Educator

Biological effects of thermal water-associated hydrogen sulfide on human airways and associated immune cells: Implications for respiratory diseases

    No full text
Natural mineral (thermal) waters have been used for centuries as treatment for various diseases. However, the scientific background of such therapeutic action is mostly empiric and based on knowledge acquired over time. Among the various types of natural mineral waters, sulfurous thermal waters (STWs) are the most common type in the center of Portugal. STWs are characterized by high pH, poor mineralization, and the presence of several ions and salts, such as bicarbonate, sodium, fluoride, silica, and carbonate. Furthermore, these waters are indicated as a good option for the treatment of various illnesses, namely respiratory diseases (e.g., allergic rhinitis, asthma, and chronic obstructive pulmonary disease). From the sulfide species present in these waters, hydrogen sulfide (H2S) stands out due to its abundance. In healthy conditions, H2S-related enzymes (e.g., cystathionine β-synthase and cystathionine γ-lyase) are expressed in human lungs, where they have mucolytic, antioxidant, anti-inflammatory, and antibacterial roles, thus contributing to airway epithelium homeostasis. These roles occur mainly through S-sulfhydration, a post-translational modification through which H2S is able to change the activity of several targets, such as ion channels, second messengers, proteins, among others. However, in respiratory diseases the metabolism of H2S is altered, which seems to contribute somehow to the respiratory deterioration. Moreover, H2S has been regarded as a good biomarker of airway dysfunction and severity, and can be measured in serum, sputum, and exhaled air. Hence, in this review we will recapitulate the effects of STWs on lung epithelial-immune crosstalk through the action of its main component, H2S

Orientação: Ana Filipa Gordino Beato ; coorientação: Renato Emanuel Carvalho Pessoa dos SantosCurrent literature studding Long Distance Relationship (LDR) originated by military deployment is mainly focused on at home partner’s perspective and the associations between geographical separation and relational satisfaction. Little is known about the role that commitment and sexual satisfaction play in the relationship and the perspective of the deployed military members in comparison with those living a relationship based on a daily contact with their partners. Objective: The main purpose of this cross-sectional and comparative study was to compare diverse relational variables, such as commitment, relational satisfaction and sexual satisfaction between Portuguese military members in military deployment, i.e., experiencing a Long Distance Relationship (LDR) and those who maintained a Geographically Close Relationship (GCR). Design and Method: Participants in Long Distance Relationships (n=172) were Portuguese military members deployed in Afghanistan (n=72), Central-African Republic (n=86) and São Tomé and Príncipe (n=15). The ones that were in a GCR (n=256) were also Portuguese military members, but on normal duty in different Military Units and Bases in the Portuguese Territory. All voluntary participants were in a romantic relationship and completed an online survey, that included the Investment Model Scale (Rodrigues & Lopes, 2013), the New Sexual Satisfaction Scale (Pechorro et al., 2014) and items assessing Communication satisfaction, previously used in exploratory studies with Portuguese military members and their partners. LDR and GCR data was collected simultaneously, three months after deployment. Bivariate correlations were used to analyze the associations between variables, independent samples T-test was applied to compare the samples according the variables of interest, and simple and sequential mediations were performed to check for the predicted models. Results: The findings suggest that the military members in LDR had significantly higher levels of commitment, relational and sexual satisfaction than those in GCR. No differences were found in the degree of relational investment. Additionally, communicational satisfaction and relational satisfaction were identified as significant indirect mediators of the relationship between the group (LDR vs GCR) and commitment and the group (LDR vs GCR) and sexual satisfaction. Conclusions: Overall, the results are in line with previous research that evidenced an increased overall satisfaction with various relational factors when partners are living a Long Distance Relationship

The airport is increasingly the link between the region and the world, serving as an economic, social and cultural driver. An airport is so efficient the better their integration into its environment. This premise is an obligation in modern times due to a global economic crisis that tends to drag. As is known, the Northern Region of Portugal is heavily populated by migratory population, which annually returns to Portugal, it's also increasingly a point of interest for tourists from around the globe. The air link through Francisco Sá Carneiro Airport is a success factor in the North, thus contributing to a mutual development. Since the liberalization of airspace in the European Continent and the arrival of Low- Cost airlines to Portugal, was found in the North Region an exponential increase in tourism and therefore commercial. This paper will address the real impact of Francisco Sá Carneiro Airport, also known as Oporto Airport, regarding the development of the Northern Region. The Catchment Area of it, goes beyond the boundaries of Portugal border, and increasingly, as a result of intermodality, extends to the neighboring country, thus providing new opportunities

Transcriptome profiling in Spinocerebellar ataxia type 2: a focus on RNA-binding proteins

    No full text
Spinocerebellar ataxia type 2 (SCA2) is a rare autosomal dominant neurodegenerative disease first described in 1971 and characterized by cerebellar ataxia and slow saccades. It is caused by a gain-of-function mutation of ATXN2, which encodes the ataxin-2 protein. Ataxin 2 is a multi-functional RNA-binding protein (RBP), and many of its related biological pro cesses, such as calcium homeostasis and lipid metabolism, have been implicated in SCA2 path ophysiology. However, ataxin-2’s RNA-binding function, through which it interacts with vari ous facets of the RNA metabolism system, has yet to be directly linked to SCA2. This thesis’ main objective was to analyze the cerebellar transcriptome of the novel transgenic SCA2 mouse model, TG-SCA-Q129, with a focus on genes encoding RBPs, to un cover whether or not SCA2 might be characterized by a dysfunction of RNA metabolism, as it pertains to its presentation in this mouse model. Eighty-four differentially expressed genes were identified between two lines of trans genic mice compared against wild-type mice, such as Alyref and Aptx, and an analysis of the functions of their encoded RBPs indicated an abundance of functions related to known SCA2 mechanisms, but also to mRNA processing and DNA repair. Gene ontology and weighted gene co-expression network analyses of the whole tran scriptome of the transgenic animals were also conducted, identifying not only known mecha nisms of SCA2, but also inflammation in the cerebellum of the transgenic line with a prominent motor phenotype, and alterations in synaptic vesicle endocytosis. In conclusion, the results from this thesis point to RNA metabolism being a core mech anism in SCA2, and identify DNA repair, pre-mRNA processing, neuroinflammation, and syn aptic vesicle endocytosis as warranting further research to verify whether they might also char acterize SCA2.A Ataxia Espinocerebelosa Tipo 2 (SCA2) é uma doença neurodegenerativa autossó mica dominante descrita pela primeira vez em 1971 e caraterizada por uma ataxia cerebelar e sacadas lentas.
