Determination of differentially expressed late responsive genes upon treatments of bth, baba and trichoderma harzianum in wheat
Chemically and biologically induced genes are involved in complex sets of defense responses against a broad spectrum of pathogens. Understanding the cross-link between specific and non-specific responses, systemic acquired resistance (SAR), upstream and downstream signaling molecules that regulate the transduction pathway is essential to solve response mechanisms. The wheat line Avocet-S was treated with benzo (1,2,3) thiadiazole-7-carbothioic acid S-methyl ester (BTH), beta-aminobutyric acid (BABA) and Trichoderma harzianum Raifi KRL-AG2, potentially used as bio-control fungal agent, for identification of candidate genes/gene fragments induced/repressed upon treatments. The differentially expressed wheat genes were detected by Differential Display RTPCR (DDRT-PCR), then cloned and sequenced. GenBank database was searched for significant hits of known genes using blastn algorithm. To verify the differences in the expressions of these gene fragments quantitative RT-PCR (qRT-PCR) was conducted. The confirmed differentially expressed wheat genes are Triticum aestivum Ubiquitin conjugating enzyme (TaUBC2), T. aestivum inducible phenylalanine ammonia-lyase (TaPAL1), T.aestivum autophagy-related protein 8 (TaATG8) and an unknown protein (TR1AE_CS42_4AL_TGACv1_288542_AA0951670.1). These findings are useful for identification of novel mechanisms modulating the defense-signaling network. Autophagy, ubiquitination and phenylpropanoid metabolism are important for immune responses and they can be activated by nonpathogenic chemicals and biological agents.ODTÜ'deki MSA laboratuvar
Genetic relationship of wild einkorn based on geographical distribution in Anatolia and Thrace using AFLP markers
Triticum monococcum L. ssp boeoticum Boiss., is the wild progenitor of domesticated einkorn. High throughput AFLP genetic analysis showed that the domestication of einkorn started in the northern part of the Fertile Crescent, near the Karacadag Mountains, Southeastern Turkey [1]. This study assesses the genetic distribution and the diversity of wild einkorn throughout Turkey, using total of 59 accessions from 22 locations in four different geographical regions. In our study, the four selective combinations of AFLP markers (E+ACC/M+ACT, E+ACC/M+ATA, E+ACT/M+ATA, and E+ATC/M+AAG) resulted in 161 AFLP marker loci. Phylogenetic trees for individual accessions and populations based on geographical regions were obtained using 'PopGen-32' population genetic analysis software. East and Southeast samples were genetically closest to each other among the samples from other regions. The samples from West, Northwest, and Central Anatolia were clustered together
Genetic diversity and relationship analysis among accessions of Aegilops ssp in Turkey using amplified fragment length polymorphism (AFLP) markers
Amplified fragment length polymorphism (AFLP) DNA markers were used to assess the genetic diversity and relationships between 55 accessions of genus Aegilops, including the species Aegilops triuncialis L. (UUCC), Aegilops geniculata Roth (MMUU), Aegilops cylindrica Host (CCDD) and Aegilops umbellulata Zhuk (UU). The samples were collected from Aegean region and East Anatolia, Turkey. 16 AFLP selective primer combinations generated a total of 3200 polymorphic amplification products. 50 Aegilops accessions were analyzed using the data analysis software, unweighted pair-group method arithmetic average (UPGMA) method and numerical taxonomy and multivariate analysis system (NTSYSpc-2.02k). The similarity index coefficients were calculated according to simple matching coefficient. Using 16 AFLP primer combinations, species from Aegean region and east Anatolia were clustered as four major groups. Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Bu topluluklar, kimi zaman AVM inşaatı için ağaçların kesilmesine karşı çıkmakta, kimi zaman toplu taşıma fiyatlarının artışını protesto etmekte, bazen de kapitalist dünyanın hükümdarları olduğu düşünülen %1'lik kaymak kesimin alanlarını işgal etmektedir. Bu hareketlerin önemli bir bölümünü ise tüketici hareketi oluşturmaktadır. İşte bu çalışma; tüketimin, zararlı, yok eden, küçük düşürücü ve ayrıştıran yönüne karşı çıkan özgün bir grubu—tüketim karşıtlarını—inceleme yoluyla toplumsal/sürdürülebilir Pazarlama nosyonuna katkıda bulunmayı amaçlamaktadır. Temellendirilmiş Kuram isimli nitel metodolojiden faydalanılarak yürütülen araştırma, Türkiye'de farklı illerde yaşayan ve farklı jenerasyon ve altyapılara sahip tüketim karşıtlarının, baskın pazarlama ve tüketim sisteminin özelliklerinin farkında olan bireyler olduğunu ve bu bireylerin tüketime benzer şekilde baktıklarını ortaya koymaktadır. Araştırma sonucunda beliren model, tüketim karşıtlarının dönüşüm, amaç ve davranışlarını tüm yönleriyle açığa vurmaktadır. Araştırma sürecinin işletme ve pazarlama yönetimine sağladığı yol haritası, üretilen ürünlerin, fonksiyonalite, rasyonel değer, verimlilik ve etik değer ölçüleri gözetilerek geliştirilmesi ve fiyatlandırılmasına işaret etmektedir. Bu yol haritasına göre etik olmayan imaj inşası ve etik olmayan tutundurma faaliyetleri de pazarlama tarafından artık bir kenara bırakılmalıdır. Tüketim karşıtlarının nitelikli insan kaynağı özellikleri taşıdıkları görülmekte, farkındalığı yüksek ve etik değerler ile hareket ettiği düşünülen bu bireylerin toplumsal gelişme yolunda kilit oyuncu rolleri üstlenebileceği anlaşılmaktadır
Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more
