    Cerium ammonium nitrate: A new catalyst for regioselective protection of glycols

    The regioselective introduction of a methoxymethyl (MOM) group on different type of glycols via an orthoester intermediate was investigated. The novelty presented in this study is the use of ceric ammonium nitrate instead of the previously employed camphorsulfonic acid as catalyst.     Design and synthesis of conformationally restricted carbocyclic nucleosides

    Los nucleósidos carbocíclicos son análogos de nucleósidos en los que el átomo de oxígeno del anillo furanósico está reemplazado por un grupo metileno. La ausencia de una verdadera unión glicosídica le otorga mayor estabilidad metabólica pues no son afectados por fosforilasas e hidrolasas que sí atacan a los nucleósidos convencionales. Las enzimas responsables de distintas acciones farmacológicas de los nucleósidos convencionales también reconocen los análogos carbocíclicos correspondientes, obteniéndose un amplio rango de propiedades biológicas, especialmente como agentes antivirales y antitumorales. La conformación en este tipo de drogas cumple un rol fundamental en la actividad biológica; sin embargo, los carbanucleósidos presentan una conformación distinta a la de los nucleósidos convencionales en solución, por lo que son menos efectivos. N-metano-carba-timidina (50), un carbanucleósido sintético rígido en la conformación Norte y estructuralmente relacionado al nucleósido carbocíclico natural neplanocina C (48), construido en un sistema bicíclico[3.1.0]hexano, exhibió una potentísima acción antiherpética, incluso mayor que aciclovir. Por este motivo, se llevaron a cabo estudios sintéticos para optimizar su estructura química. En los dos primeros capítulos de esta tesis se describen los antecedentes de nucleósidos, sus análogos carbocíclicos y aquellos que presentan restricciones conformacionales. También se incluye un resumen de los métodos de síntesis más comunes para la preparación de carbanucleósidos y la síntesis y actividad biológica de los compuestos más relevantes de esta familia de análogos. En el tercer capítulo se extendió el estudio de la monoprotección regioselectiva de glicoles por tratamiento con DIBAL-H y ortoformiato de metilo catalizado por nitrato cérico amónico. Este estudio surgió de la necesidad de proteger regioselectivamente un hidroxilo alílico secundario frente a uno homoalílico, durante la síntesis enantioselectiva de (+)-neplanocina F (183), realizada en nuestro laboratorio. En los capítulos cuatro y cinco se presenta la primera síntesis de nucleósidos carbocíclicos conformacionalmente restringidos en una conformación de tipo Norte, construidos en un sistema tiabiciclo[3.1.0]hexano. Para dicho estudio se desarrollaron nuevos métodos para la preparación de episulfuros fusionados a anillos de cinco miembros sintetizándose primero modelos simples de carbanucleósidos construidos sobre este sistema azufrado. N-tia-carba-timidina (273) se comportó como un agente antiherpético muy potente, comparable a aciclovir.Carbocyclic nucleosides are nucleosides analogues in which the oxygen atom of the furanose ring is substituted by a methylene group. These analogues have great metabolic stability because they are unaffected by phosphorylases and hydrolases that cleave the glycosidic bond of conventional nucleosides. The responsible enzymes for pharmacological actions of conventional nucleosides also recognize the carba-analogues displaying a wide range of biological properties, especially as antiviral and antitumoral agents. The conformation of this type of drugs plays an important role in the biological activity. In solution, carbanucleosides present a different conformation than for conventional nucleosides, for that reason they are less effective in molecular recognition processes. N-methano-carba-thymidine (50), a carbocyclic nucleoside locked in the North conformation and structurally related to the natural carbanucleoside neplanocin C (48), built on a bicyclo[3.1.0]hexane system, exhibited potent antiherpetic activity, even greater than acyclovir, a well-known antiherpetic agent. Synthetic studies motivated to optimize the chemical structure of 50 were performed. Chapters one and two provide an introduction to nucleosides, carbocyclic nucleosides and those with conformational restrictions. A summary of the most common synthetic methods to prepare carbanucleosides and the synthesis and biological activity of representative members of this family of compounds are included. Chapter three describes the study of the scope about the regioselective monoprotection of glycols by treatment with DIBAL and methyl orthoformate catalyzed by ceric ammonium nitrate. This study emerged as a consequence of the need of protecting regioselectively a secondary allylic hydroxyl group over a homoallylic in the course of the enantioselective synthesis of (+)-neplanocin F (183), developed in our laboratory. Chapter four and five present the first synthesis of conformationally locked carbocyclic nucleosides built on a thiabicyclo[3.1.0]hexane system, in a Northern-type geometry. For this purpose, new methods for the preparation of an episulfide group fused to a five-member ring were developed. At the beginning, simple models of carbanucleosides built on this sulfur-containing system were prepared.     Synthesis of conformationally locked carbocyclic nucleosides built on a thiabicyclo[3.1.0]hexane system as a pseudosugar surrogate

    The synthesis of prototype models of purine and pyrimidine carbanucleosides built on a 6-thiabicyclo[3.1.0]hexane system as pseudosugar moiety has been investigated. These pyrimidine carbanucleosides proved to be very stable compounds, in contrast to the parent epoxy analogs, which experienced epoxide ring-opening due to intramolecular enol base attack. In addition, as the synthesis of a thiirane moiety fused to a five-membered ring is not a trivial synthetic task, validation and optimization of the existing methods for episulfide preparation were required to access the committed synthetic precursor of the title compounds: (±)-(1RS,2RS,5SR)-6-thiabicyclo[3.1.0]hexan-2- ol, compound 28. © Wiley-VCH Verlag GmbH & Co.     High‐Throughput Experimentation (HTE) Applied to the Synthesis of API Impurities: A Case Study in Oxidative Decarboxylation of Aryl Acetic Acids

    High-throughput experimentation (HTE) has dramatically impacted experimental reaction development by enabling the rapid exploration of a diverse set of reaction conditions. During the past few decades, HTE has evolved as a tool to expedite reaction discovery and optimization. This work details the application of HTE to synthesize impurities of active pharmaceutical ingredients (APIs) with ketone/aldehyde functionality, specially focusing on ibuprofen impurity E. Initial experiments using K2S2O8 as oxidant yielded moderate results. Subsequent HTE screens identified cerium ammonium nitrate (CAN) and RuCl3-NaIO4 as new effective decarboxylative oxidants, with RuCl3-NaIO4 in the presence of tetrabutylammonium bromide (TBAB) achieving the highest yield of 65%. This optimized method was successfully applied to synthesize ibuprofen impurity E on a gram scale. Additionally, the applicability of these methods to obtain other API related substances, such as naproxen impurity L and ketoprofen impurity A, was demonstrated. This research highlights the potential of HTE to streamline the synthesis of API impurities, making them widely accessible for pharmaceutical development purposes.Fil: Paolo, Luciano.     Design and synthesis of aryloxyethyl thiocyanate derivatives as potent inhibitors of Trypanosoma cruzi proliferation

    As a part of our project directed at the search of new chemotherapeutic agents against American trypanosomiasis (Chagas' disease), several drugs possessing the 4-phenoxyphenoxy skeleton and other closely related structures employing the thiocyanate moiety as polar end group were designed, synthesized, and evaluated as antiproliferative agents against Trypanosoma cruzi, the parasite responsible for this disease. These thiocyanate analogues were envisioned bearing in mind the potent activity shown by 4- phenoxyphenoxyethyl thiocyanate (compound 8) taken as lead drug. This compound had previously proved to be an extremely active growth inhibitor against T. cruzi with IC50 values ranging from the very low micromolar level in epimastigotes to the low nanomolar level in the intracellular form of the parasite. Of the designed compounds, the ethyl thiocyanate drugs connected to nonpolar skeletons, namely, arylthio, 2,4-dichlorophenoxy, ortho-substituted aryloxy, and 2-methyl-4-phenoxyphenoxy (compounds 15, 34, 47, 52, 72, respectively), were shown to be very potent antireplicative agents against T. cruzi. On the other hand, conformationally restricted analogues as well as branched derivatives at the aliphatic side chain were shown to be moderately active against T. cruzi growth. The biological activity of drugs bearing the thiocyanate group correlated quite well with the activity exhibited by their normal precursors, the tetrahydropyranyl ether derivatives, when bonded to the same nonpolar skeleton. Compounds having the tetrahydropyranyl moiety as polar end were proportionally much less active than sulfur-containing derivatives in all cases. Drugs 47 and 72 also resulted to be very active against the amastigote form of the parasite growing in myoblasts; however, they were slightly less active than the lead drug 8. On the other hand, compounds 34 and 52 were almost devoid of activity against myoblasts. Surprisingly, the dithio derivative 15 was toxic for myoblasts.Fil: Szajnman, Sergio Hernan.     Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    Variations on the Author

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
