    The role of neuraminidase-1 in muscle regeneration

    A neuraminidase-1 (Neu1) participa da regulação do catabolismo de sialoglicoconjugados nos lisossomos. A deficiência congênita da Neu1 é a base da sialidose, doença neurossomática grave associada a deformidades osteoesqueléticas, hipotonia e fraqueza muscular. Camundongos com deficiência de Neu1 (Neu1-/-) desenvolvem uma forma atípica de degeneração muscular caracterizada por expansão da matriz extracelular (MEC), invasão das fibras musculares por fibroblastos, fragmentação do citoplasma, formação vacuolar e atrofia muscular. Apesar de a degeneração muscular estar bem caracterizada nestes animais, a miogênese ainda não havia sido estudada. O objetivo desta pesquisa foi avaliar o envolvimento da Neu1 no processo de regeneração muscular, após aplicação de cardiotoxina (CTX) em camundongos Neu1-/-, em comparação com controles normais. A CTX foi administrada no músculo tibial anterior direito e os animais foram eutanasiados por deslocamento cervical 1, 3, 5, 7, 10, 14, 21 e 28 dias após a lesão. Os músculos foram analisados através de histologia; medição da área transversa das fibras musculares centronucleadas; verificação do potencial proliferativo celular por quantificação de marcação de BrdU; imunoistoquímica para inflamação, fibras regenerativas e fibrose; e expressão gênica e proteica de fatores de transcrição musculares. Os dados foram comparados estatisticamente e as variações significativas devem apresentar p <= 0,05. Nos animais com deficiência de Neu1, o processo inflamatório (especialmente a reação macrofágica) e o potencial proliferativo estavam aumentados nas fases iniciais, acompanhados da hiperexpressão de Pax7. Observamos atraso na maturação muscular caracterizado por maior expressão de miosina embrionária em estágios mais tardios da regeneração. Os genes MyoD e MyoG estavam com expressão aumentada no período de 5 a 10 dia após a lesão, embora a expressão destas proteínas estivesse reduzida. Ao final da regeneração, houve maior deposição de reticulina na MEC, indicando processo fibrótico. A Neu1 parece atuar em todos os estágios da regeneração muscular, desde a fase aguda da lesão através do controle da proliferação celular, até a maturação muscular e estágios finais em que regularia a deposição de componentes da MECNeuraminidase-1 (Neu1) participates in sialoglycoconjugates catabolism in lysosomes. Congenital Neu1 deficiency is the basis of sialidosis, a severe neurosomatic disorder associated with osteoskeletal deformities, hypotonia and muscle weakness. Mice with Neu1 deficiency (Neu1-/-) develop an atypical form of muscle degeneration characterized by abnormal fibroblast proliferation and expanded extracellular matrix (ECM), invasion of muscle fibers by fibroblast, cytosolic fragmentation, vacuolar formation and muscle atrophy. Despite muscle degeneration is well characterized in these animals, myogenesis has not been studied so far. The aim of this study was to evaluate the involvement of Neu1 in muscle regeneration process after cardiotoxin (CTX) injection in Neu1-/- mice and normal controls. CTX was applied in the right tibialis anterior muscle, and the animals were euthanized by cervical dislocation 1, 3, 5, 7, 10, 14, 21 and 28 days after injury. The muscles were analyzed through histology; cross-sectional area of regenerative muscle fibers; quantification of BrdU labeling; immunohistochemistry labelling for inflammation, regenerative fibers, and fibrosis; and gene and protein expression of muscle transcription factors. The data were compared and variances considered statistically significant in case p <= 0.05. In animals with Neu1 deficiency, both inflammatory process (mainly macrophagic response) and proliferative potential were increased in the initial stages, accompanied by overexpression of Pax7. We observed delay in muscle maturation characterized by higher expression of embryonic myosin later in muscle regeneration. MyoD and MyoG genes were overexpressed from 5 to 10 days after injury, though the expression of these proteins was reduced. At the end of muscle regeneration, reticulin deposition in ECM was increased, indicating fibrotic process. Neu1 seems to participate in all stages of muscle regeneration, since acute injury phase through the control of cell proliferation, towards muscle maturation, and at the final stages when it would regulate the deposition of ECM component

    Characterization of satellite-cells gene expression profile from dystrophic mice carrying different molecular defects

    As células-satélite musculares vêm sendo muito estudadas em diferentes pesquisas, especialmente com o objetivo de aumentar a compreensão do mecanismo de sua ação na regeneração muscular e as respectivas implicações nas diferentes miopatias, visando identificar possíveis alvos terapêuticos. Dois modelos para distrofias, os camundongos Largemy d e Lama2dy2j/J possuem um padrão de degeneração intenso e bastante semelhante, mas com diferenças na expressão de genes envolvidos na cascata de regeneração. Por isso, estes constituem interessantes modelos para o estudo de possíveis diferenças no mecanismo de ativação e atuação das células-satélite no músculo distrófico. Assim, os objetivos específicos deste projeto consistiram em: 1) isolar e caracterizar por citometria de fluxo as populações de células-satélite dos modelos Largemy d e Lama2dy2j/J em comparação com o normal C57Black6, quanto a expressão de marcadores de miogênese e de células-tronco pluripotentes; e 2) estudar e comparar o perfil de expressão gênica destas populações de células satélites através de microarray de expressão. Na caracterização fenotípica das células isoladas do músculo normal, aquelas que aderem mais precocemente (PP1) e mais tardiamente (PP2) mostram um padrão fenotípico semelhante entre si e com características mais próximas às miogênicas. Já a população de células de adesão bem mais tardia (PP6) apresentou um padrão de marcação misto, mantendo as características miogênicas, mas apresentando tambem padrão de células-tronco mesenquimais, sugerindo o fenótipo de células mais imaturas. Nos músculos distróficos, identificamos diferenças na constituição do pool de células presentes inicialmente no músculo, onde na linhagem Lama2dy2j/J há indícios de uma população em estágio proliferativo, enquanto que na linhagem Largemy d há presença de células com maior imaturidade. Os resultados da analise de expressão gênica nas populações caracterizadas se mostraram concordantes com os fenótipos celulares avaliados por citometria. Identificamos a hipo-expressão de genes ligados à regeneração e remodelamento muscular em ambos modelos distróficos. Considerando os genes diferentemente expressos somente em cada um dos modelos, os resultados sugerem ativação de proliferação celular e inibição da diferenciação na linhagem Lama2dy2j/J e distúrbios na miogênese na linhagem Largemy d. Assim, pudemos identificar vias importantes alteradas em cada um dos modelos que explicam parte das diferenças encontradas nos trabalhos anterioresMuscle satellite cells have been widely studied, especially to understand their mechanism of action in muscle regeneration and correspondent implications in the different dystrophic processes, aiming the identification of potential therapeutic targets. Two mice models for muscular dystrophies, Largemyd and Lama2dy2j/J, have a pattern of an intense and very similar degeneration, but with differences in the expression of genes involved in the regeneration cascade. Therefore, they are interesting models to study possible differences in the mechanism of activation and action of satellite cells in the dystrophic muscle. The main objectives of this project are: 1) to isolate and characterize by flow cytometry, populations of satellite cells from Largemyd and Lama2dy2j/J models, as compared to normal C57Black6, evaluating the presence of myogenic and pluripotent stem cells markers; and 2) to study and compare gene expression profiles of these populations of satellite cells using microarray technique. In the phenotypic characterization of cells harvested from normal muscle, both faster (PP1) and slower (PP2) populations to adhere in culture flasks show similar phenotypic characteristics, which were closer to myogenic phenotype. On the other hand, the population of cells with very delayed adhesion ability (PP6) presented a mixed pattern, maintaining the myogenic characteristics, but associated to positive mesenchymal stem cell´s markers, suggesting a phenotype of more immature cells. In dystrophic muscles, we could identify differences in the constitution of the first pool of cells present in the Lama2dy2j/J muscle where there is evidence of a population in proliferative stage, while in the Largemyd strain, we found more immature cells. Gene expression profile in the characterized populations showed consistent concordance with the cellular phenotypes assessed by flow citometry. In both dystrophic models, we identified down-regulated genes related to regeneration and remodeling of the muscle. Considering only the genes differently expressed in each dystrophic model, data suggest the activation of cell proliferation and inhibition of differentiation pathways in Lama2dy2j/J strain and altered myogenesis in the Largemyd model.     As distrofias musculares constituem um grupo heterogêneo de doenças genéticas, caracterizadas por uma degeneração progressiva e irreversível dos músculos. Modelos murinos distróficos, como o mdx, SJL/J, Largemyd e Lama2dy-2J/J, são ferramentas importantes para o estudo destas doenças. O objetivo deste trabalho consistiu em estabelecer parâmetros de avaliação funcional que visem a sua utilização para elucidar os benefícios clínicos de futuras terapias. Para tanto, foram avaliadas as quatro linhagens distróficas, em diferentes idades, e comparadas a controle normal. Os testes padronizados consistiram em nado forçado, avaliação de resistência/equilíbrio pelos membros anteriores e pelos quatro membros, caminhar em plataforma suspensa, suspensão pela cauda, grip strength e rota rod. Comprovou-se a existência de diferentes padrões de força, resistência, coordenação motora e aprendizagem/memória ao longo do tempo de vida de cada linhagem, o que permitiu traçar parâmetros a serem utilizados em futuras pesquisas de terapia celular e farmacológica.Muscular dystrophies are a heterogeneous group of genetic diseases characterized by a progressive and irreversible degeneration of the muscles. Dystrophic mouse models, like the mdx, SJL/J, Largemyd and Lama2dy-2J/J, are important tools for studying these diseases. The aim of this study was to establish parameters for functional evaluation aiming its use to elucidate the clinical benefits of future therapies. Thus, we evaluated the four strains of dystrophic mice, at different ages, and compared to normal control. Standardized tests consisted of forced swimming, evaluation of resistance/balance by forelimb and four members, walking on suspended platform, suspension by the tail, grip strength and rota rod. We observed the existence of different patterns of strength, endurance, coordination and learning / memory over the lifetime of each strain, which allowed tracing parameters to be used in future studies of cell and pharmacology therapies

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     Influence of force and functionality of the upper limbs in patients with muscular dystrophy duchenne in corticosteroids

    O nosso objetivo foi de avaliar evolutivamente a perda da força muscular e das habilidades motoras, bem como a progressão da distribuição da fraqueza muscular nos diferentes segmentos dos membros superiores em pacientes com distrofia muscular de Duchenne em corticoterapia. Selecionamos seguintes testes de fácil aplicação: Teste de força Manual Muscular Escala Medical Research Council; Teste ABC provas de coordenação visual-motora e fatigabilidade provas n° 1, 3, 7 e 8; Grau funcional de Brooke; Índice de Barthel. Os testes foram aplicados em 40 pacientes com idades entre 5 e 15 anos, deambulantes e não deambulantes, os quais foram avaliados três vezes, com intervalos de seis meses entre cada avaliação. Os resultados mostraram que a progressão da distribuição da força muscular nos membros superiores ocorreu dos segmentos proximais para os distais em todos os pacientes e foi maior nos pacientes não deambulantes e com maior idade. O ato de escrever (pegada no lápis e/ou caneta) não foi influenciado pela progressão; no entanto, o aumento da fatigabilidade foi um fator limitante contribuindo para a redução do ritmo e da qualidade da escrita. O grau funcional de Brooke confirmou a variação das medidas de força muscular e nas atividades de vida diária o nível de dependência foi maior nos pacientes com maior idade, acontecendo nestes compensações funcionais importantes que lhes permitiram realizar a atividade de forma adaptada.Our aim was to analyze the progression of the involvement of muscle strength and functional motor hability as well as the progression of the weakness pattern in the upper limbs of children with Duchenne muscular dystrophy who were receiving steroid therapy. We evaluated 40 patients with DMD, aged 5-15 years, by using the following simple tests: MRC score based on the upper limbs muscles; ABC tests number 1, 3, 7 and 8 for assessing visual motor coordination and fatigability; Brooke functional ability scale and modified Barthel index. All boys were evaluated every 6 months along a period of 18 months. The loss of muscular strength showed a proximal to distal progression and was greater in non- ambulant and in older patients. The act of writing (to hold pencil or pen) was not influenced by the progression; however, the fatigability increased and was a limiting factor for the speed and the quality of the hand writing. The Brooke functional ability scale confirmed the changes in the muscular strength. The performance in daily activities showed a greater dependence in the older patients who adopted functional compensations for performing the activities in an adapted way
