Knock down of cytosolic phospholipase A2: an antisense oligonucleotide having a nuclear localization binds a C-terminal motif of glyceraldehyde-3-phosphate dehydrogenase

The subcellular localisation of oligodeoxynucleotides (ODN) is a major limitation for their use against nuclear targets. In this study we demonstrate that an antisense ODN directed against cytosolic phospholipase A(2) (cPLA2) mRNA is efficiently taken up and accumulates in the nuclei of endothelial cells (HUVEC), human monocytes and HeLa cells. Gel shift experiments and incubation of cells with oligonucleotide derivatives show that the anti-cPLA2 oligo binds a 37 kDa protein in nuclear extracts. The TAAAT sequence was identified as the major binding motif for the nuclear protein in competition experiments with mutated ODNs. Modification of the AAA tripler resulted in an ODN which failed to localise in the nucleus. Moreover, inserting a TAAAT motif into an ODN localising in the cytosol did not modify its localisation. The 37 kDa protein was purified and identified after peptide sequencing as glyceraldehyde-3-phosphate dehydrogenase (GAPDH). It was shown by confocal microscopy that GAPDH co-localises with anti-cPLA2 ODN in the nucleus and commercial GAPDH effectively binds the oligo. Competition experiments with increasing concentration of NAD(+) co-factor indicate that the GAPDH Rossmann fold is a docking site for antisense oligonucleotides containing a TAAAT motif. Extracellular DNA as matrix component in microbial biofilms.
Bacteria in nature primarily live in surface-associated communities commonly known as biofilms. Because bacteria in biofilms, in many cases, display tolerance to host immune systems, antibiotics, and biocides, they are often difficult or impossible to eradicate. Biofilm formation, therefore, leads to various persistent infections in humans and animals, and to a variety of complications in industry, where solid–water interfaces occur. Knowledge about the molecular mechanisms involved in biofilm formation is necessary for creating strategies to control biofilms. Recent studies have shown that extracellular DNA is an important component of the extracellular matrix of microbial biofilms. The present chapter is focussed on extracellular DNA as matrix component in biofilms formed by Pseudomonas aeruginosa as an example from the Gram-negative bacteria, and Streptococcus and Staphylococcus as examples from the Gram-positive bacteria. Besides the role of extracellular DNA in biofilm formation, the mechanisms involved in DNA release from P. aeruginosa, Streptococcus, and Staphylococcus are addressed
Modelamento e simulação de impacto balístico em sistema cerâmica-metal

Programa de Pós-Graduação em Ciência e Engenharia de MateriaisNovos sistemas de proteção balística vêm sendo recentemente criados com base em materiais de alta tecnologia. Um dos objetivos da indústria do ramo é desenvolver sistemas defensivos mais leves, porém mais fortes, que possibilitem ao portador, veicular ou humano, uma maior mobilidade com um maior nível de segurança. Este trabalho, baseado no trabalho original de Al-Qureshi et al [1], estuda matematicamente o comportamento de uma proteção contra um impacto de projétil. O modelo empregado inclui um sistema de proteção em camadas de cerâmica e metal, e ainda descreve o comportamento do projétil e as propriedades de absorção de impacto do sistema. A literatura ainda mostra que a taxa de erosão e desaceleração do projétil são altamente dependentes dos parâmetros geométricos e estruturais do material da proteção. O fenômeno de impacto e penetração é descrito em diferentes estágios, apresentando características particulares entre tais. As equações apresentadas demonstram diferentes propriedades do sistema ao longo dos estágios. Este trabalho ainda apresenta uma simulação matemática realizada sobre o modelo desenvolvido e aprimorado em busca de propriedades otimizadas do material para estudos futuros. Dentre as propriedades investigadas citam-se a espessura das placas utilizadas, o perfil de deformação do material metálico, a densidade da cerâmica, dentre outras características relevantes para o fenômeno. A rotina computacional possibilitou a aplicação de diferentes parâmetros nas equações propostas. Como resultado, gráficos e superfícies foram geradas, o que possibilitou uma análise mais profunda do modelo e um maior entendimento do processo de fratura em materiais por impacto de alta velocidade. Estudos futuros utilizarão estes resultados e desenvolvimentos para a produção de uma proteção balística que será utilizada para um experimento prático

Mapping Sound Worlds: A Composition Portfolio
This portfolio combines a selection of musical works I created between 2017 and 2022. In many ways, each of the eight selected pieces, no matter how sonically, conceptually, or stylistically different from each other, intertwines with the ideas of the Internal Family Systems psychological model (IFS), where the mind is naturally seen as subdivided into an indeterminate number of subpersonalities or parts that develop a complex system of interactions among themselves. This philosophy has become crucial to my daily practice over the last seven years, deeply affecting my creative process and conception of individual sounds and their sound worlds. Most importantly, it has profoundly changed the way I listen.
My earlier piece Cositas Diminutas (2017), encapsulates my fascination with the thingness of found objects and explores how they shape a performer’s behavior in a musical context of a prepared piano. The narrative in this piece builds around the theatre of objects inside a piano, where each object is seen as an extension of the instrument and the pianist (a puppeteer) and represents a part of their multifaceted, complex entity.
My further experiments with the genre of music theatre culminate in Thousand Splinters of a Human Eye (2017), where each performer metaphorically represents a living thought inside one’s mind. Together, the “thoughts” form one intricate system in action. Carefully designed improvisatory situations in the piece invite curiosity and humor into play resulting in surprising interactions between the characters on stage. For the scores of both pieces, I developed a method of layered scores where each page consists of two or more layers (each combination is scanned separately). When read together, the pages reflect the multi-dimensionality and simultaneity of sonic situations.
The piece Silenced (2019) manifests a stylistic and conceptual turn of my compositional path, focusing on extending the sound world from within the sound itself – drilling a well of sound in pursuit of unfolding its utmost corners and amplifying its tiniest details with the means of analog electronics. This self-contained electronic eco-system enables the resonant space inside the timpani to speak for itself in the form of emerging voice – feedback, inviting for a more intimate interaction between the performers and the instrument, as well as between the performers themselves.
A Cow in the Backyard (2020) originates from the sound of a found object (a garbage truck) and explores the transformation of the mundane into art through granular synthesis, instrumental writing, and stop-motion animation. The electronic tape serves as a bridge between the ensemble and the visuals. Its emancipated rhythmical layer responds to the inherent rhythm of frames in the animation and mutates into motives over time. It plays the leading role in the musical part of the piece and brings what usually stays implicit and hidden to the foreground.
Following up on Silenced, my large-scale work for six electric guitars, Asymptotic Freedom (2020 – 2021), delves further into the possibilities of a monochrome ensemble. In this piece, I aim to expand the vibrating space within the instrument outward and create space through sound by composing sonic reflections from one guitar to another. At any given moment, the geography of Asymptotic Freedom permits the coexistence of macro- and micro-sound spaces, and local sonic events. Furthermore, I largely diversify and expand the possibilities of the feedback palette, which plays a significant role in enhancing the spatial awareness of a listener.
The composition X is Where I Am (2021) explores an intimate expression of Self – a female voice – through its many parts in the ensemble and is centered around the theme of displacement and home. A moving agent in a concert space (the voice) is transmitted via transducers inside a prepared piano on stage. The separation between the vocalist and a slightly distorted resonance of her voice naturally sets the trajectory for the unfolding. The ensemble dispersed in the hall acts as a resonant body for the voice, gradually transforming the sound space throughout the piece. The motivic language of X is Where I Am, roots in Russian folklore and alludes to the genre of Russian lament, which is explored further in my piece Whispering to the Stars.
The research on intimate expression continues further in my work Whispering to the Stars (2022). Following a gradual emergence of translucent melodic motives, the piece enters obscure corners of the originating sound field. Eventually, the three voices erupt as a choir, filtered through the vessels of the brass instruments, completing the range of emotion in the lament. The spatial quality inherent to this piece significantly advanced during its adaptation to the Wave Synthesis System in 2023.
Forget Me Not (2022), the last piece in my portfolio, follows the evolution of a melodic line into a fabric of interlacing strands. The interaction between the amplified piano voice, altered via guitar pedals, and a saxophone creates a subpersonality of the latter, sonically affected by the pedals – this is a particular example of the creation’s internal complexity that reflects its ever-changing multifaceted nature. Similarly, all instruments in the ensemble permeate each other and share the gravitational pull forming a palpable layered substance curled into a vortex of sound space
БЕЗОПАСНОСТЬ АРКТИЧЕСКОГО РЕГИОНА: НАУКОМЕТРИЧЕСКИЙ АНАЛИЗ ИНФОРМАЦИОННЫХ МАССИВОВ МЕЖДУНАРОДНОЙ И РЕГИОНАЛЬНОЙ БАЗ ДАННЫХ
The article traces main trends of the scientific research development on security in the Arctic region using the international database Web of Science by Thomson Reuters Co and the regional database Scholar Sibirica generated by the State Public Scientific Technological Library of the Siberian Branch of the Russian Academy of Sciences. Using a scientometric analysis and analytical tools of DBs, the author shows countries, institutions and organizations that study this problem, as well as funds financing them; names productive periodicals and authors with high publication activity; presents monographs and dissertation studies of recent years; reveals the main research areas. In conclusion, the author notes that the study of the safety at the Arctic territories is an urgent problem for the entire world society, not only for the northern countries; the above-mentioned DBs should be regarded as an information base for further study of the Arctic security.Objective – to carry on a scientometric analysis of information arrays (IA) devoted to studying the Arctic region security.Methods: scientometric analysis.Results: countries, institutions and organizations that study this problem are represented, as well as funds financing them; productive periodicals and authors with high publication activity are designated; monographs and dissertation studies of recent years are shown; the main research fields are revealed.Practical implications: the study should be used as an information base for further research of scientists and specialists on various aspects of the Arctic region security. В статье прослежены тренды развития научных исследований по теме «Безопасность Арктического региона» с использованием баз данных – международной Web of Science (компании Thomson Reuters) и региональной Научная Сибирика (собственной генерации Государственной публичной научно-технической библиотеки Сибирского отделения Российской академии наук). В базах данных с использованием булевых операторов и ключевых слов выявлены документальные массивы по теме, проведен их сравнительный наукометрический анализ. Аналитические сервисы баз данных позволили назвать страны, учреждения и организации, занимающиеся изучением данной проблемы, а также финансирующие их фонды. Определены наиболее продуктивные периодические издания и авторы с высокой публикационной активностью. Представлены научные мероприятия, проводимые по теме, монографии и диссертационные исследования последних лет. Показана тематическая структура информационных массивов. Отмечено, что изучение безопасности арктических территорий является актуальной проблемой всего мирового сообщества, а не только северных стран. Вышеназванные базы данных могут служить информационной основой дальнейших исследований различных аспектов безопасности Арктического региона.Цель – проведение наукометрического анализа информационных массивов, посвященных изучению безопасности Арктического региона.Методы: наукометрический анализ.Результаты: представлены страны, учреждения и организации, которые изучают эту проблему, а также фонды, финансирующие их; обозначены продуктивные периодические издания и авторы с высокой публикационной активностью; показаны монографии и диссертационные исследования последних лет; выявлены основные направления исследований.Область применения результатов: исследование могут служить информационной основой дальнейших исследований ученых и специалистов по различным аспектам безопасности Арктического региона
ОСОБЛИВОСТІ РЕГУЛЮЮЧИХ МЕХАНІЗМІВ ІНТЕГРАЦІЇ КАПІТАЛУ В КОРПОРАТИВНОМУ СЕКТОРІ (НА ПРИКЛАДІ ПОЛЬЩІ)
The development of the corporate sector in Poland has become an important element in the formation of a new economic environment and the transformation of social relations. The article analyses the regulatory mechanism in the corporate sector of Poland, which concerns, in particular, the mechanisms of integration and capital development, with the help of the content analysis method. Considerable attention is paid to updating Polish legislation in accordance with the requirements of the European Union (EU). The author carried out a retrospective analysis of the process of the development of market economy in Poland and the place of privatization in it. The study describes significance of privatization processes in Poland for new progressive tendencies of development of public relations. The article analyses the problems that still remain relevant for the development of the corporate sector in Poland. The regulatory features of the activity of transnational companies in modern Poland and their significance for the economy are studied. The author notes the role of Poland’s membership in the EU to ensure further development of the market economy, the development of the corporate sector in it on the basis of capital integration. It is concluded about a significant capitalization of Polish companies. Access of Polish companies to capital is a necessary condition for dynamic development, as small Polish businesses are unable to accumulate investment funds on their own and incur high loan costs.The development of the corporate sector in Poland has become an important element in the formation of a new economic environment and the transformation of social relations. Tato bakalářské práce je zaměřena na analýzy rizik v podniku. Obsahem teoretické části je vymezení pojmů, souvisejících se strategickým řízením podniku a analýzou rizik. V praktické části je stručně představena restaurace Burger King Area Bory DRIVE THRU a rizika navrhovaných strategií rozvoje podniku zpracovávané autorkou. Navrhované strategie jsou vytvořené na základě provedené PEST analýzy a Porterova modelů pěti sil. Následně je provedena identifikace, analýza, ovládnutí rizik pomocí modelu 4T a následné opatření rizik navrhovaných strategií restaurace. Závěrečnou kapitolu tvoří celkové zhodnocení praktické časti.ObhájenoThis bachelor's thesis is focused on risk analyses in the company. The content of the theoretical part is the definition of terms related to strategic management of the company and risk analysis. In the practical part, the restaurant Burger King Area Bory DRIVE THRU is briefly presented, and also the risks of the proposed business development strategies processed by the author. The proposed strategy is based on a PEST analysis and Porter's five forces model. Subsequently, the identification, analysis, control of risks using the 4T model and subsequent measures of the risks of the restaurant's proposed strategies are carried out. The final chapter is an overall evaluation of the practical part
Анализ особенностей речевой стратегии кандидата в президенты Франции Э. Макрона и их перевод на русский язык
Предмет исследования данной работы— речевая стратегия кандидата в президенты Франции Э. Макрона. В работе представлены результаты анализа речей и публичных выступлений Э. Макрона во время президентских выборов во Франции в 2016-2017 гг. на предмет лексических и грамматических особенностей. Кроме того, автором предложены варианты и способы эквивалентного перевода рассматриваемых характерных черт на русский язык. В настоящем исследовании была выдвинута и подтверждена гипотеза о том, что речевой стратегии Э. Макрона свойственна большая неформальность стиля и языка, чем это традиционно допустимо для дискурса французских политиков, и что именно подобная речевая стратегия стала если не достаточным, то необходимым условием быстрого обретения популярности среди избирателей Э. Макроном и его дальнейшей победы на президентских выборах. Актуальность работы обусловлена современностью рассматриваемого языкового материала и значительным интересом политологов и специалистов по политическому дискурсу.The subject of the present research is the speech strategy of Emmanuel Macron, a candidate for the french presidential elections in 2016-2017. The work presents the results of grammar and lexicology analysis of the public speeches presented by E. Macron during his electoral campaign. Also presented in the work are the ways of translating characteristic grammar and lexical features found in the speeches into Russian. The hypothesis of the research suggests E. Macron's speech strategy being more informal than political discourse is generally thought to be. The author also suggests it is his speech strategy that has provided for such a burst in popularity among the french electorate of E. Macron. The importance of the subject addressed is explained by an already existing attention to the subject in the scientific fields as well by an overall modern-day language material treated
Evaluation of German Automatic Speech Recognition solutions in the context of speech and language therapy support of people with aphasia
Those who suffer from aphasia benefit from digital speech and language therapy solutions, and automatic speech recognition (ASR) has been already used for giving feedback on the correctness of the answers in naming exercises. AphaDIGITAL application is to provide German-speaking users with detailed feedback on phonemic/phonetic and semantic errors, based on automatic speech and language processing. For this purpose, open-source ASR solutions for German were evaluated on different corpora of atypical speech, including two small datasets with aphasic speech samples. Character error rate, the number of precisely recognized items and empty outputs served as evaluation metrics. The four selected models are generally robust to the deteriorated condition of speech and audio quality and consistently outperform commercial models in atypical speech recognition. Applying error acceptance threshold, additional use of phonemic error rate, and other valuable insights for ASR implementation in aphaDIGITAL are discussed
