    Análise epidemiológica e de associação dos genes kir com artrite reumatoide

Programa de Pós-Graduação em Biologia Celular e do DesenvolvimentoA artrite reumatoide (AR) é uma doença autoimune caracterizada por inflamação crônica que causa erosões e deformidades nas articulações e manifestações inflamatórias sistêmicas como acometimentos pulmonar e cardíaco. As células natural killer (NK) são células efetoras que participam tanto da resposta imune inata quanto adaptativa através de produção de citocinas e ação citolítica. As NK são encontradas com frequência nas articulações afetadas pela AR. Dentre os receptores das células NK, destaca-se o receptor das células killer semelhante à imunoglobulina (KIR) que possui ação ativadora e/ou inibidora das NK. O objetivo deste trabalho foi investigar a presença/ausência dos genes KIR na população de Santa Catarina e a associação da presença dos genes KIR ativadores com pacientes de AR e com dados clínicos. Neste estudo foram avaliados 147 pacientes com AR (casos) e 164 indivíduos sem histórico de doença autoimune (controle) para a presença/ausência de 16 genes KIR, através da técnica de PCR por primer de sequência específica (PCR-SSP). No entanto, apenas os genes KIR ativadores (KIR2DS1, KIR2SD2, KIR2DS3, KIR2DS4, KIR2DS5, KIR3DS1) foram investigados para o estudo de associação caso-controle. As frequências gênicas foram estimadas e o odds ratio (OR) foi calculado para avaliar associação dos genes KIR ativadores com AR (p<0,05 foi considerado significativo). As amostras de pacientes e controles tiveram predominância de mulheres (87%) com média de 53(±12) anos e com ascendência europeia (86%). As frequências de presença/ausência dos 16 genes KIR em pacientes com AR assemelharam-se aos estudos com AR e outras doenças autoimunes no Brasil, bem como as frequências destes genes no grupo controle aproximaram-se aos demais estudos brasileiros para populações eurodescendentes. Não foi encontrada associação do polimorfismo de ausência e presença dos genes ativadores com AR. No entanto, a presença do gene KIR2DS2 foi mais frequente em pacientes com AR que apresentaram manifestação articular (MA) em relação ao grupo controle (OR=2,312; p=0,010). Além do mais, não houve associação do tagabismo com a AR, apenas com fator reumatoide (FR) em pacientes com AR (OR=2,947; p=0,02). Os genes KIR não foram associados ao desenvolvimento de AR neste estudo. Entretanto, o gene KIR2DS2 foi mais frequente em pacientes com a presença de MA, enquanto que o hábito tabagista foi relacionado com FR

    Clinical equivalence of amlodipine medications in arterial hypertension patients

    The study was focused on clinical equivalence of two amlodipine medications (cardilopin and norvasc) in patients with Stage I-III arterial hypertension (AH). Forty-two patients (mean age 52.1±3.9years) were divided into two groups, according to cardilopin or norvasc administration. At baseline and 6 months later, echocardiography (EchoCG), Doppler cardiac and vessel sonography were performed. Monthly blood pressure (BP) control demonstrated that BP target level was achieved by the end of Month One and sustained till the study's end, at similar doses of norvasc and cardilopin (5-10 mg/d). By the end of the study, left ventricular (LV) myocardial mass reduced, and prevalence of normal LV geometry increased, with significant improvement in LV diastolic function in both groups. Initially decreased linear blood flow parameters and pulse index, as well as increased extracranial vascular resistance, normalized by 6 months. Hemodynamic parameters were similar in both groups. According to its beneficial influence on initially disturbed hemodynamics, LV geometry and cerebral blood flow in AH patients, cardilopin is as effective as norvasc

    Modulation of Rage and CD147 in Cerebral Amyloid Angiopathy <i>in vitro</i>

    Introduction. In the study of Alzheimer’s disease (AD), the cause-and-effect relationship between neurodegenerative changes and the accompanying amyloid angiopathy is becoming increasingly important. The accumulated clinical data indicates that an important contribution to the pathogenesis of AD is made by neurovascular unit dysfunction, including disruption in permeability of the blood-brain barrier (BBB), microcirculation, and metabolic coupling of cells.Aim. To study the molecular mechanisms of disturbed brain microcirculation and the structural and functional integrity of the BBB in experimental models of AD in vitro under the modulation of CD147 and RAGE.Materials and methods. The study was carried out on C57BL/6 mice. First, we formed an AD model in animals of the experimental group. Then, we isolated and cultured primary cells of the brain, modulated the activity of CD147 and RAGE in endothelial cells using siRNA CD147, siRNA RAGE, cyclophilin A and Aβ1-42, and formed a BBB model in vitro. Further, we assessed transendothelial electrical resistance in the BBB model in vitro, registered the marker molecules of angiogenesis and analyzed the expression of APP in endothelial cells. Statistical processing of the obtained data was carried out using the methods of nonparametric statistics: the Mann – Whitney U test for comparing independent samples and the Wilcoxon test for comparing dependent samples. The level of statistical significance of differences was p ≤ 0.05.Results and discussion. Knockdown of RAGE led to a statistically significant increase in TEER, an intensification of neoangiogenesis, and a decrease in the level of APP expression. At the same time, although CD147 knockdown led to an increase in TEER, it also led to controversial effects on angiogenesis and an increase in APP expression.Conclusion. Analyzing the data obtained, it can be concluded that RAGE and CD147 silencing in the cells of cerebral microvessels can become a promising method for reducing their pathological permeability

    Abstract-Soil organic matter (SOM) was studied in relation to vertic processes (i.e., shrinking/swelling, cracking, vertical turbation, lateral shearing, gilgai formation) in Vertisols and vertic soils of the North Cau casus in Russia, and Texas and Louisiana in the USA. Their impact on SOM properties and distribution was analyzed according to various levels of soil organization, such as soil cover, profile, horizon, and aggregate structure using chemical methods, micromorphology, isotopic analyses, and physical fractionation. The greatest variations both in the distribution and properties of SOM were found in mature Vertisols at the level of soil cover including C tot , organic carbon stocks, stable carbon isotopic composition, and SOM 14 C age, chemical composition. The distribution of SOM at the profile and horizon levels was related to the function ing of Vertisols during wet dry cycles. The isotopic and chemical study of densi granulometric fractions at the aggregate level reflected the minor role of vertic processes

    Central image of Vertisols: evolution of concepts of their morphology and genesis

    This paper discuss the changes in understanding of the central image of Vertisols and leading processes of their formation. The early concept described Vertisols as black or dark clayey soils with homogenous undifferentiated profile resulted basically of pedoturbation. The further studies discovered vertical differentiation of Vertisol attributes. The application of trench method discovered spatial heterogeneity of Vertisols with alternation of bowl and diapiric structures. Such spatial complex subsurface pattern seems to be rather common and can be found even in the absence of gilgai microrelief. A new central image of a mature Vertisol is a combination of two structural types, one being homogenous and monotonous, generally corresponding to the initial central image, and the other - heterogeneous profile with fragmented horizons. A leading process forming the new central image of Vertisols was defined as lateral shearing or plastic deformations, i.e., plastic movements and gradual upward pushing of moist material (analogy of defluction process). Pedoturbation or more exactly the vertical falling of surface material into the cracks results in the vertical mixing rather than in deformations. Micromorphological features typical of Vertisols and associated with shrink-swell phenomena, cracking, mixing and lateral shearing that are reflected in the central image of Vertisols are summarized in the paper and illustrated by microphotographs

    THE DYNAMICS OF THE MICROBIOTA OF URINE IN PATIENTS WITH ACUTE OBSTRUCTIVE PYELONEPHRITIS IN PERIODS OF OBSERVATION.

    Abstract. Analysis of urine culture in 72 patients with acute obstructive pyelonephritis within 6 months. When hospitalization was performed urine culture before and after the elimination of the upper urinary tract obstruction (VMP). Over the next 5 days the patients received empirical antibiotic therapy until the results of the microbiological examination of urine, after which all patients were divided into two groups. 1 group of patients consisted of 38 people who received antibiotic therapy based on antibiotic susceptibility obschedokazannyh pathogens (E.coli, Proteus sp., Klebsiella sp., Enterococcus sp., Etc.), The 2 group of patients included 34 men treated with antibacterial treatment based antibiotic susceptibility of microorganisms isolated from urine. After the completion of antibiotic therapy, all patients underwent clinical and laboratory and bacteriological monitoring of urine through the 1,3 and 6 months follow-up
