Making neurons from stem cells : molecular mechanisms and spider silk substrates
The understanding of the function of the nervous system and the brain is one of the major intellectual challenges in life sciences. Neurological and psychiatric disorders are in addition major issues for the society, and new approaches are needed to learn more about the brain and to develop new treatments. The development of the mammalian brain is a highly regulated process that involves extra- and intracellular signaling to efficiently regulate gene expression in a precise spatial and temporal manner. The understanding of the differentiation mechanisms into neurons, glia and other cell types in the developing forebrain however is still incomplete. Studies of embryonic telencephalic neural stem cells (NSCs) in vitro may increase the understanding of the molecular mechanisms of brain development, and aid in developing new protocols for defined differentiation of stem cells for clinical use.This thesis is aimed at investigating the mechanisms underlying bone morphogenetic protein(BMP4)-mediated differentiation of NSCs, and to explore the use of recombinant spider silk protein-based matrices in combination with signaling factors, especially BMP4, to generate functional neural cell circuits in vitro. In the first study we discovered that BMP4 treatment of NSCs resulted in a dramatic increase in the expression of the BMP4-inhibitor Noggin. BMP4 mediated non-neural differentiation into mesenchymal cells at low seeding densities, neuronal differentiation at high seeding densities, and astrocyte differentiation in any condition. As the Noggin levels increased linearly at higher densities, we hypothesized that the endogenous Noggin production predominantly mediated an inhibition of mesenchymal differentiation. We further observed that BMP4 stimulation induced an AMPA responsive neuron population at high seeding densities, and that this population was increased by co-stimulation of the signaling factor Wnt3a. By applying whole transcriptome sequencing, we aimed at elucidating the molecular mechanisms responsible for the increased neuronal differentiation by BMP4+Wnt3a. This approach, however revealed an unexpected increase in the expression of genes associated with inhibitory GABAergic neurons, and also functional the expression of the neurogenic bHLH factor Hes6.To apply these novel protocols for differentiation of NSCs into functional neurons, we introduced a novel way of culturing NSCs in substrates generated from recombinant spider silk protein (4RepCT). Spider silk protein is a promising biomaterial due to its biocompatibility, biodegradability, and possibility to use in various forms both in 2D and 3D. NSCs cultured in 2D cultures on 4RepCT “film” structures showed no significant differences in cell proliferation, viability, or differentiation potential compared to control cultures in optimized conditions. 4RepCT substrates generated as “foam” structures could be used for 3D culturing of NSCs, and these NSC cultures differentiated nicely into astrocytes and neurons. Calcium imaging assays revealed that BMP4+Wnt3a-treatment of NSCs grown in 3D4RepCT-matrices resulted in efficient generation of functional excitatory neurons.These studies have thus revealed new molecular mechanisms underlying neural differentiation of cortical stem cells, and point to the versatility of using spider silk protein-based substrates for stem cell cultures. Future studies aim at testing these new concepts in vivo for improved treatment of neurological disease.List of scientific papersI. Abstract
Introduction. Children's and adolescents' injuries are a serious medical problem, as they lead to substantial medical and socio-economic effects.
Aim of the paper. The aim of the conducted research was to analyze incidence and causes of hospitalizing children between birth and 18 years of age on the grounds of injuries.
Material and methods. Medical histories of 2 832 children between birth and 18 years of age treated for injuries in 2006-2010 in Wladyslaw Bieganski Regional Specialist Hospital in Grudziadz were analyzed.
Results. Correlations between treatment method and subsequent years of research (p<0,05), between children's age group and type of injury, and between type of injury and season of the year and hospitalization time, were shown to be statistically significant. No correlation was found in the studied population between subsequent years of research and injury types or residence of the injured.
Conclusions. 1. The most frequent causes for placing children in the hospital were cranioencephalic injuries, injuries to the osteoarticular system, and burns. 2. The children's age determined the type of injury inflicted. Children in the youngest age group were most frequently hospitalized for burns, children in the age of 3-6 years for foreign bodies, and in the group of 7-18 years of age for injuries to the chest and to the osteoarticular system. 3. Boys were more frequently injured in a manner requiring hospital treatment than girls were. 4. A decisive majority of injured children required only conservative treatment and a stay in the hospital for no more than 3 days. 5. A constant tendency of higher occurence of injuries in children during spring and summer, was observed.

Użycie języka ojczystego na lekcjach języka angielskiego – opinie studentów filologii angielskiej
For years there have been debates among applied linguists and in-service teachers as to whether to allow any inclusion of learners’ mother tongue in the process of foreign language teaching. Since second language acquisition theories do not offer a precise answer, the actual use of the first language in the classroom depends on the beliefs of teachers and learners. Recent research studies concerning teachers and students’ views on whether learners’ L1 should be present or avoided in the teaching context seem to suggest that at present the controlled use of the L1 is perceived as beneficial. This article presents the results of a survey study on the perception of the usefulness of the L1 conducted among pre-service teachers of English at the University of Białystok. The findings support the view that the purposeful employment of the L1 may result in an increase in learning outcomes.Od wielu lat toczą są dyskusje wśród językoznawców i praktykujących nauczycieli, czy język ojczysty uczniów powinien być obecny w procesie nauczania języka obcego. Teorie nabywania drugiego języka nie dają jednoznacznych odpowiedzi. Zakres użycia języka ojczystego zatem zależy od przekonań nauczycieli i uczących się. Ostatnie badania na ten temat przeprowadzone wśród nauczycieli i uczniów zdają się wskazywać, że ograniczone stosowanie języka pierwszego przynosi korzyści dydaktyczne. Artykuł przedstawia wyniki badań sondażowych dotyczących postrzegania użyteczności języka ojczystego przeprowadzone wśród studentów filologii angielskiej na Uniwersytecie w Białymstoku. Phase transitions of ferruginous minerals in the course of thermal processing of feldspar-quartz raw materials from the Sobótka region (Lower Silesia)
AbstractThis paper presents the results of analyses of feldspar-quartz raw materials from deposits of leucogranites located in the Sobótka region. This is a successive stage of research carried out by the author on reasons for colour variation of ceramic materials obtained from them. This step encompassed the firing of sample pairs of analogous chemical composition in different conditions: in a standard cycle lasting more than 2 hours (at a maximum temperature of 1200°C), and a fast one - lasting around 50 minutes (at a max. temperature of 1260°C). The obtained ceramic bodies were analysed using the XRD method, scanning microscopy SEM-EDS and57Fe Mössbauer spectroscopy at room temperature. The XRD investigations revealed the presence of quartz in both samples, while remnants of sodium feldspar were observed in the one fired in the fast cycle. The scanning microscopy confirmed that the principal component of the examined bodies is the aluminosilicate melt, resulting from the thermal decomposition of mainly feldspars and quartz. Single quartz grains and other minerals of high melting temperatures, i.e. zirconium, were also observed in the course of microscopic examinations. Products of other mineral phases’ transformations at high temperature, such as: titanomagnetite (spinel Fe-Ti), magnetite, biotite, xenotime, sphalerite as well as probably chlorites and garnets, were also relatively frequent. Mössbauer studies demonstrated the presence of three basic components constituting 30% (each) of the spectra, i.e. Fe2O3(presumably hematite), Fe3+ions (dispersed in aluminosilicate glassy phase), and nanocrystalline or amorphous oxide phase of iron (Fe-O). Some relatively small amounts (5 and 10%) of mixed valence iron cations (Fe2+/3+) that are not expected to influence the colour difference between samples after firing were also found. A paramagnetic doublet referring to them can be attributed to a titanomagnetite spinel or products of its thermal decomposition. The above-mentioned observations and examinations lead to the finding that the main reason for colour variation in the examined bodies as well as their different microstructure and advancement of phase transitions in the course of firing are different conditions of thermal treatment. These probably also influenced the forms in which iron and other colouring elements occur in the samples studied.</jats:p

A concept presented in the form of an architectural design becomes most valuable when it is put down on paper through soul and hand, like a violinist transferring a melody to the strings with a sensitivity of an author. In spite of the development of computer technology, at the stage of impressive concept designing there is an even greater need for the return to hand-drawn designs. In view of electronic media, it shows a high quality of the architect’s technique, original and individual approach and specific value of handmade creation

Abstract
The rights patient as a set of rights due to the use of health services in Poland result from the Constitution of the Republic of Poland. They are regulated by the whole regulations. The aim of this work is to present the rights patient and duties in primary care.

Prawa pacjenta definiowane są jako zespół uprawnień przysługujących człowiekowi z tytułu korzystania ze świadczeń zdrowotnych. W Polsce wynikają z Konstytucji Rzeczypospolitej Polskiej i Karty Praw Pacjenta. Odwołują się do głównych dokumentów międzynarodowych. Uregulowane są przez przepisy szczegółowe.
Celem niniejszej pracy jest przedstawienie praw i obowiązków pacjenta - świadczeniobiorcy w podstawowej opiece zdrowotnej.
