    Neoepitopes on COL17 for LAD autoantibodies

    Transmembrane collagen XVII (COL17) is a hemidesmosomal component of basal keratinocytes that can be targeted by autoantibodies in autoimmune blistering disorders, including linear IgA dermatosis (LAD). COL17 can be physiologically cleaved within the juxtamembranous extracellular NC16A domain, and LAD autoantibodies preferentially react with the processed ectodomains, indicating that the processing induces neoepitopes. However, the details of how neoepitopes develop have not been elucidated. In this study, we show that C-terminal processing of COL17 also plays a role in inducing neoepitopes for LAD autoantibodies. First, the mAb hC17-ect15 targeting the 15th collagenous domain of COL17 was produced, which showed characteristics similar to LAD autoantibodies. The mAbs preferentially reacted with C-terminally deleted (up to 682 amino acids) recombinant COL17, suggesting that C-terminal processing shows neoepitopes on the 15th collagenous domain. The LAD autoantibodies also react with C-terminal deleted COL17. Therefore, neoepitopes for LAD autoantibodies also develop after C-terminal processing. Finally, the passive transfer of the mAb hC17-ect15 into human COL17-expressing transgenic mice failed to induce blistering disease, suggesting that neoepitope-targeting antibodies are not always pathogenic. In summary, this study shows that C-terminal processing induces dynamic structural changes and neoepitopes for LAD autoantibodies on COL17

    Patterns of Retirement as Reflected in Income Tax Records for Older Workers

    If retirement means a substantial and sustained reduction in the time spent working for pay or profit, measurement requires a definition of substantial and sufficient observations of the same individuals to determine whether a transition from “working” to “retired” status has occurred. Using the Statistics Canada Longitudinal Administrative Databank, a 20 percent sample of the individual income tax returns of all tax filers since 1980, we identify those with significant labour force attachment at ages 50-52, and follow them year by year. If retired means having no income from employment, the median age of retirement is about 63 for men, 62 for women. That is true for all cohorts. If earning up to half of one’s previous employment income is deemed consistent with being retired, the median age is about 60 for both men and women. Results obtained in this way are consistent with calculations based on Labour Force Survey data.retirement, older workers

Thrombosis and bleeding continue to be a major cause of morbidity and mortality in leukemias. The pathophysiology of these disorders is unique, and not only the disease but also the treatment and other factors play a role. There has been an increase in the understanding of these disorders in leukemias. However, it is still difficult to predict when and which patients will have these complications. The evidence for the management of coagulation abnormalities in leukemias is still evolving and not as established as in solid malignancies. The management of these disorders is complex, and making clinical decisions is often challenging. In the era of specialization, where there are different hematologists looking after benign- and malignant-hematology patients, opinions of thrombosis experts are often sought by leukemia specialists.     Robust Regression Shrinkage And Consistent Variable Selection Via The LAD-LASSO

    The least absolute deviation (LAD) regression is a useful method for robust regression, and the least absolute shrinkage and selection operator (lasso) is a popular choice for shrinkage estimation and variable selection. In this article we combine these two classical ideas together to produce LAD-lasso. Compared with the LAD regression, LAD-lasso can do parameter estimation and variable selection simultaneously. Compared with the traditional lasso, LAD-lasso is resistant to heavy-tailed errors or outliers in the response. Furthermore, with easily estimated tuning parameters, the LAD-lasso estimator enjoys the same asymptotic efficiency as the unpenalized LAD estimator obtained under the true model (i.e., the oracle property).     A new LAD curve fitting algorithm: slightly overdetermined equation systems in L_1

    We present a new algorithm for the discrete LAD curve-fitting problem for n points in k < eq n dimensional space. When k is about n/3 it begins to out-perform the best current methods, and the advantage increases with k. The algorithm should be of interest in approximation theory and in robust regression. Moreover our approach may be useful in linear optimization, especially in linear programming.Technical report DCS-TR-10

    About the Ash Lad is inspired by a Dano-Norwegian folktale collection by Peter Christen Asbjørnsen and Jørgen Moe that was published in 1859. It is said that Henrik Ibsen, author of the original Peer Gynt drama, was inspired by this collection of folk tales of which the story “About Ash Lad , Who Stole the Troll’s Silver Ducks, Coverlet, and Golden Harp” (some other titles it is known by include “Boots and the Troll”), is a part of. In this piece, the unfolding of the story in the form of a suite is also inspired by the famous Grieg setting of Peer Gynt, as we follow the hero along his journey, through perils and triumphs. There are a total of nine small movements, all connected by a dance-like ‘Passage’ theme, that parallel the Ash Lad’s story as he ‘borrows’ three items from a Troll in order to win the local princesses hand in marriage and a decent half of the kingdom on top of that. It was an absolute joy to compose this work for the thoroughly amazing Inventi Ensemble, and I thank them for the opportunity to transport myself creatively into a world of fantasy and imagine my own sounds within this old Dano-Norwegian realm.     Sequential graft was used frequently in clinical studies. In this study, the hemodynamic effect of one kind of sequential graft in two different conditions of the lesion was discussed and some recommendations on the surgical procedures were made. A patient-specific three-dimensional (3D) model of left anterior descending artery (LAD) was reconstructed. A moderate stenosis exist in the trunk of LAD between the first and the second diagonal branch (D1 and D2). Another 3D model without stenosis was also reconstructed based on the patient-specific model. Sequential graft and single graft were applied on these two 3D model. Thus four 3D models were built so that the hemodynamic effect of sequential graft can be discussed. The zero-dimensional (0D)/3D coupling method was used to perform the numerical simulation by coupling the 3D artery model with a 0D lumped parameter model of the cardiovascular system. The flow rates in the branches of LAD and the graft flow were calculated and illustrated in this paper. The wall shear stress (WSS) and oscillatory shear index (OSI) were also calculated and depicted. If the native LAD is stenosis, sequential graft should be applied for the short-term outcomes. Moreover, the long-term patency of the sequential graft applied on the stenosis LAD is good. The long-term patency of the single graft was bad. But the short-term outcomes are almost the same when LAD is not stenosis.     Right ventricular function assessment in single LAD lesion patients using strain and strain rate imaging

    AbstractBackgroundStrain and strain rate imaging is currently the most popular echocardiographic technique that reveals subclinical myocardial damage, and data are not available on this imaging method with regard to assessing right ventricular involvement in single LAD lesion.AimTo evaluate right ventricular regional functions using strain and strain rate imaging tissue Doppler method in patients with single LAD lesion.MethodsThe patient group was composed of 60 patients who had experienced first anterior myocardial infarction and had undergone successful percutaneous coronary intervention for LAD lesion. Twenty patients were selected for the control group. The right ventricular myocardial samplings were performed in three regions: the basal, mid, and apical segments of the lateral wall. The individual myocardial velocity, strain, and strain rate values of each basal, mid, and apical segment were obtained.ResultsThe right ventricular tissue Doppler parameters (Sm, E, A, E/A ratio, IVA, E/E″) of the patients group were significantly decreased than controls. In addition, changes in the right ventricular mean strain and strain rate values were significantly lower in patient group than controls.ConclusionRight ventricular involvement in LAD lesion patient is significant even after PCI and recanalization of LAD. TDI, strain and strain rate are new, useful imaging techniques for detection of subclinical RV dysfunction in patients with LAD lesion

    Quotidian Classics: Nick Hornby's Lad Lit

    This essay explores the genre of lad lit and Nick Hornby's contribution to the genre. Hornby's focus is on the quotidian, specifically the male experience in the 1990s and their relationship with football and Popular culture, thus bringing ordinary lives closer to canon. Lad lit has only been addressed academically to a certain extent, but Hornby's work holds up to critical review and deserves to be treated as such. Firstly, Hornby's works are brought into historical context and viewed through feminist lens. Secondly, we apply psychoanalytic approaches, citing Freud as well as Lacan, and viewing Hornby's work through discourses on melancholia and addiction. Finally, we address identity and readership. Through these methods we find that Hornby’s lad literature is made distinct by the obsessions that seem to rule the protagonists’ lives and define their identities – so much so that they border on addiction. To re-join society they must adapt, which requires deep introspection for the lads to lead meaningful lives. What we can take away from this discussion is that Hornby's work pertains to the human experience, and because he treats it with humor and skill it is made that much more accessible to the average reader. Hornby asserts that the joy of reading is of the upmost importance, and the author of this paper agrees wholeheartedly. Hornby's wilting taps directly into the reader's stream of consciousness in an automatic process such that time ceases to exist until the pagers run out. The ease of Hornby writing has become increasingly important in our day and age where multiple agents compete for every second of our attention and close reading of canonical classics can be strenuous

    Empirical likelihood for LAD estimators in infinite variance ARMA models

    In this paper, we use an empirical likelihood method to construct confidence regions for the stationary ARMA(p, q) models with infinite variance. An empirical log-likelihood ratio is derived by the estimating equation of the self-weighted LAD estimator. It is proved that the proposed statistic has an asymptotic standard chi-squared distribution. Simulation studies show that in a small sample case, the performance of empirical likelihood method is better than that of normal approximation of the LAD estimator in terms of the coverage accuracy.
