    Human Bone Morphogenetic Protein-2 (hBMP2) is an osteoinductive agent physiologically involved in bone remodeling processes. A commercialized recombinant hBMP2 produced in mammalian cell lines is available in different clinical applications where bone regeneration is needed, but widespread use has been hindered due to an unfavorable cost/effective ratio. Protein bodies are very large insoluble protein polymers that originate within the endoplasmic reticulum by prolamine accumulation during the cereal seed development. The N-terminal domain of the maize prolamin 27 kD γ-zein is able to promote protein body biogenesis when fused to other proteins. To produce high yield of recombinant hBMP2 active domain (ad) in stably transformed tobacco plants we have fused it to the γ-zein domain. We show that this zein-hBMP2ad fusion is retained in the endoplasmic reticulum without forming insoluble protein bodies. The accumulation levels are above 1% of total soluble leaf proteins, indicating that it could be a rapid and suitable strategy to produce hBMP2ad at affordable costs

    O acontecer na clínica : quando o criar resiste ao cotidiano

    Este trabalho consiste numa cartografia de mapas afetivos do fazer clínico contemporâneo a partir de encontros do autor com experimentações clínicas e paisagens artísticas que componham brechas instituintes nos mecanismos disciplinares e de controle do capitalismo atual. Sociedade capitalista que repele de si o contato angustiante com o que sai fora de suas normatizações postas perante o viver, cotidianizando-o na intenção de anestesiar qualquer inusitado que se apresente. Ao partir dessa lógica que não suporta o inesperado, tentando a todo o momento controlá-lo, passamos a pensar a contribuição da clínica nessa produção social que impede qualquer um de alçar vôos distantes de uma vida tornada normativa e burocrática. Diante desse panorama, se problematiza a clínica no que ela pode se fazer enquanto prática que resista a tal sistema anestesiador da vida. Assim, discutimos ao longo do trabalho as relações de poder que ocorrem nessa sociedade de controle e no que elas possibilitam movimentos de resistência. Para pensar a clínica enquanto resistência ao cotidianizar-se, percorremos algumas ferramentas conceituais desdobrando os seguintes conceitos: acontecimento, individuação e ato criativo. Com eles trabalharemos o tema do fazer clínico e os possíveis caminhos que elevam a clínica a uma condição de recusa perante os mecanismos de controle utilizados em nossa sociedade. O impensável entra em jogo para daí poder criar existências que resistam ao cotidiano.This work expounds cartography of affective maps about the clinical job in contemporaneity. It was produced from author‟s clinical trials and artistic landscapes that would be able to built instituting gaps in disciplinary and control mechanisms of modern capitalism. The Capitalist society rejects from itself the anguishing touch with what goes out of its normalizations. Capitalism banalizes the everyday life an attempt to anesthetize any unusual that presents itself. Admitting this logic that does not support the unexpected and that try really hard to control the unannounced, we will try to think about the contributions of clinical work in this social production that prevents anyone from lift away from a life made legislative and bureaucratic control. Using as starting point this context, this works discusses what the clinical work could do as a practice that is able to resists in a system that anesthetizes the life itself. So, during the work we discuss the power relations in this control society, but we do it focused in what enable resistance moves. To think the clinical work as a piece of resistance in the way to do banalise the everyday life, we use some conceptual tools and unfold these concepts: happening, individuation and creative act. Using this we will formulate the theme of clinical work and the possible roads that put this experience in a refuse condition to the disciplinary and control mechanisms used in our society. The unthinkable then comes into play to create power existences that are able to resist to the banalization of the everyday life

    Union of tooth to implant using dental-implant supported prostheses

    Existem situações clínicas em pacientes parcialmente edêntulosque, para a confecção de uma prótese fixa, se faz necessário a união de dentes a implantes. Entretanto, esse tipo de conexão ainda é um assunto bastante controverso na literatura, tendo em vista possíveis complicações biomecânicas, pelas diferenças de mobilidade e a forma de absorver e distribuir as cargas funcionais. O objetivo desse trabalho foi analisar através de uma revisão detalhada da literatura os diferentes aspectos envolvendo esse tipo de situação. Como conclusão o autor afirma que a união de dentes naturais a implantes dentários é uma técnica passível de ser realizada, tento embasamento na literatura científica, contudo, a mesma, deve ser aplicada em casos específicos onde a prótese dento-implanto suportada é a única alternativa para uma prótese fixa. Para demais casos, próteses implanto suportadas serão sempre a primeira escolha de tratamento reabilitador.There are clinical situations in partially edentulous patients that it is necessary connecting teeth with implants to confection fixed prosthesis. However, this type of connection is still a very controversial subject in the literature, in view of possible biomechanical complications, by differences in mobility and way of absorbing and distributing functional loads. The aim of this study was to analyze through a comprehensive literature review different aspects involving this type of situation. In conclusion the author states that the union of natural teeth with dental implants is a feasible technique to be performed with basis in the scientific literature, however, the same should be applied in specific cases where the dental-implant supported prosthesis is the only alternative to a fixed prosthesis. For other cases, implant supported dentures will always be the first choice of rehabilitative treatment

    Expression of CLAVATA3 fusions indicates rapid intracellular processing and a role of ERAD

    The 12 amino acid peptide derived from the Arabidopsis soluble secretory protein CLAVATA3 (CLV3) acts at the cell surface in a signalling system that regulates the size of apical meristems. The subcellular pathway involved in releasing the peptide from its precursor is unknown. We show that a CLV3-GFP fusion expressed in transfected tobacco protoplasts or transgenic tobacco plants has very short intracellular half-life that cannot be extended by the secretory traffic inhibitors brefeldin A and wortmannin. The fusion is biologically active, since the incubation medium of protoplasts from CLV3-GFP-expressing tobacco contains the CLV3 peptide and inhibits root growth. The rapid disappearance of intact CLV3-GFP requires the signal peptide and is inhibited by the proteasome inhibitor MG132 or coexpression with a mutated CDC48 that inhibits endoplasmic reticulum-associated protein degradation (ERAD). The synthesis of CLV3-GFP is specifically supported by the endoplasmic reticulum cha- perone endoplasmin in an in vivo assay. Our results indicate that processing of CLV3 starts intracellularly in an early compartment of the secretory pathway and that ERAD could play a regulatory or direct role in the active peptide synthesis

    The WD40-repeat proteins nfc101 and nfc102 regulate different aspects of maize development through chromatin modification mechanisms

    The maize (Zea mays) nucleosome remodeling factor complex component 101 and 102 (nfc101/nfc102) are putative paralogs encoding WD-repeat proteins that exhibit homology with plant and mammalian components of various chromatin modifying complexes. In this study, we generated transgenic lines with simultaneous nfc101 and nfc102 down-regulation and analyzed phenotypic alterations, along with effects on RNA levels, the binding of nfc101/nfc102 and Rpd3- type histone deacetylases (HDACs), and histone modifications at selected targets. Direct nfc101/nfc102 binding and negative correlation with mRNA levels were reported for key activators of the floral transition, such as indeterminate 1 (id1) and the florigen ZCN8. In addition, the abolition of nfc101/nfc102 association with repetitive sequences of different transposable elements (TEs) resulted in tissue-specific up-regulation of transcripts produced by these regions. All direct nfc101/nfc102 targets showed histone modification patterns linked to active chromatin in nfc101/nfc102 down-regulation lines. However, different mechanisms may be involved because nfc101/nfc102 proteins mediate HDACs recruitment at id1 and TE repeats, but not at ZCN8. These results, along with the pleiotropic effects observed in nfc101/nfc102 down-regulation lines, suggest that nfc101 and nfc102 are components of distinct chromatin modifying complexes, which operate in different pathways and influence diverse aspects of maize developmen

    La Ética de la Escucha Clínica en Tiempos de Biopoder

    Esta tese trata de pensar a ética da escuta clínica em tempos de biopoder. Baseado sobretudo na filosofia esquizoanalítica como intercessora da clínica psicanalítica e na poética de Fernando Pessoa, o estudo definiu como campo problemático a Rede de Atenção Psicossocial brasileira, a partir de variados serviços e intervenções de cuidado que percorreram a atenção à saúde mental, como: Consultório na Rua, Centros de Atenção Psicossocial, Unidades Básica de Saúde, Estratégias de Saúde da Família e Pesquisa na Atenção Básica. Em um contexto de biopoder, produtor de uma subjetividade mínima, entende-se que a escuta volta-se ao corpo como organismo biológico, sem mediação com o desejo. Nesta lógica, a medicalização da vida e o anestesiamento do sofrer, tornam-se as principais práticas de cuidado. No contraponto a este controle capitalístico característico do biopoder, a tese propõe pensar o ato da escuta clínica que se passa na interação junto à potência da desrazão, da loucura e do inconsciente. O ato da escuta clínica emerge aí como dispositivo que se pretende sensibilizador para os acontecimentos inventivos da vida, apoiando cada sujeito, coletivo ou serviço que possui uma angústia a enfrentá-la de maneira inventiva. Trata-se, pois, da criação de dispositivos que lancem cada sujeito a um obrar de si e do mundo. A pesquisa indica que a clínica e o clínico que conduzem o escutar a partir do inconsciente, em sua maquinaria desejante, se posicionam, na acolhida do outro que chega para ser cuidado, em um plano intensivo de dessubjetivação de si e do mundo no que tange à formatação moral exercida pelos mecanismos de controle do biopoder: a anatomopolítica, a biopolítica e a tanatopolítica O ato da escuta clínica implica a ética de questionar um viver conservador, moral e não reflexivo, para disso desviar, inventando outros mundos. Evoca, pois, um devir artista, despersonalização que o plano da arte coloca em jogo quando assombra o mundo com suas novidades que ainda não tomaram forma petrificada. Pautada pelo método cartográfico, a pesquisa se desenrola entrecruzando escritas que se confundem, se misturam, confluem e desfluem à medida que o texto vai ganhando delineamentos. Da escrita dissertativa e acadêmica às narrativas de um conto e de crônicas, que insistem em restar, há um íntimo agenciamento que maquina o escrever sobre a clínica. Tal maquinação inicia-se em um percurso afectivo-experimentativo vivenciado pelo autor que instala um campo problemático a partir do cotidiano das práticas clínicas pelas quais exercitou o escutar. Após este anúncio, é apresentada uma análise sobre os mecanismos do biopoder e a decorrente produção de uma subjetividade mínima, subscrita por um modo narcisista de ser e pela morte do desejo, que invade o cotidiano clínico. Deste pano de fundo passa-se a uma exploração textual que envolve diários de campo sobre intervenções do autor entre seu escutar clínico e uma escuta pesquisante viabilizada pela investigação sobre o cuidado em saúde mental na atenção básica. Por fim, faz-se uma aproximação da clínica com o plano da arte, articulando uma escuta do inconsciente ao ato inventivo que se movimenta na direção de um obrar-se.This thesis deals with the ethics of clinical listening in times of biopower. The study defined the Brazilian Psychosocial Attention Network as a problematic field based,especially, on the schizoanalytic philosophy, as an intercessor of the psychoanalytic clinics, and in the poetics of Fernando Pessoa. For this purpose, it is considering several services and care interventions that covered mental health care, as:Consultation Offices in the Street, Psychosocial Care Centers, Basic Health Units, Family Health Strategies, and Primary Care Research. In a context of biopower, a producer of a minimal subjectivity, it is understood that the listening turns to the body as a biological organism without mediation with desire. In this logic, the medicalization of life and the anaesthetization of suffering become the main practices of care. In contrast to this capitalistic control characteristic of biopower, the thesis proposes to think of the act of clinical listening that takes place in the interaction with the power of unreason, madness, and the unconscious. The act of clinical listening emerges as a device that is intended to sensitizer to the inventive events of life, supporting each subject, collective or service that has an anguish to face it in an inventive way.It is, therefore, the creation of devices that launch each subject to work on himself and on the world.The research indicates that the clinical and the clinician who lead the listening from the unconscious, in their desiring machinery, are positioned in the reception of the other that arrives to be taken care of in an intensive plan of desubjectivation of oneself and of the world with regard to the formatting morality exerted by the mechanisms of control of biopower: anatomo-politics, biopolitics, and thanatopolitics The act of clinical listening causes the ethics to question a conservative, moral, and nonreflective life, to divert it, inventing other worlds. It evokes, then, a becoming artist, a depersonalization that the plane of art puts at stake when it haunts the world with its novelties that have not yet taken on petrified form. Guided by the cartographic method, the research is carried out by crisscrossing writings that merge, mingle, conjoin, and no longer converge as the text gains its outlines. From dissertation and academic writing to narratives of a short story and chronicles, that insist on remaining, there is an intimate agency that machinates writing about the clinical practice. Such machination begins in an affective-experiential path experienced by the author thatsettles a problematic field based on the daily routine of the clinical practices through which he exercised the listening. After this announcement, it is presented an analysis on the mechanisms of biopower and the resulting production of a minimal subjectivity, underwrote by a narcissistic way of being and by the death of desire, which are common on clinical routine. From this background, there is a textual exploration that involves field journals about the author's interventions from his clinical listening and hisresearcher's listening, enabled by the research on mental health care in basic care. Finally, the clinical practice is approached with the plan of art, articulating a listening from the unconscious to the inventive act that moves in the direction of a work on itself.Esta tesis trata de pensar la ética de la escucha clínica en tiempo de biopoder. Embasado sobretodo en la filosofía esquizoanalítica como intercesora de la clínica psicoanalítica y en la poética de Fernando Pessoa, el estudio determinó como campo problemático la Red de Atención Psicosocial brasileña, a partir de variados servicios e intervenciones de cuidado que recorrieron la atención a la salud mental, como: Consultorio de Calle, Centros de atención Psicosocial, Unidades básicas de Salud, Estrategias de Salud de la Familia y Pesquisa en la Atención Básica. En un contexto de biopoder, productor de una subjetividad mínima, se entiende que la escucha se vuelve hacia cuerpo como organismo biológico, sin mediación con el deseo.     Progressive Aggregation of 16 kDa Gamma-Zein during Seed Maturation in Transgenic Arabidopsis thaliana

    Prolamins constitute a unique class of seed storage proteins, present only in grasses. In the lumen of the endoplasmic reticulum (ER), prolamins form large, insoluble heteropolymers termed protein bodies (PB). In transgenic Arabidopsis (Arabidopsis thaliana) leaves, the major maize (Zea mays) prolamin, 27 kDa γ-zein (27γz), assembles into insoluble disulfide-linked polymers, as in maize endosperm, forming homotypic PB. The 16 kDa γ-zein (16γz), evolved from 27γz, instead forms disulfide-bonded dispersed electron-dense threads that enlarge the ER lumen without assembling into PB. We have investigated whether the peculiar features of 16γz are also maintained during transgenic seed development. We show that 16γz progressively changes its electron microscopy appearance during transgenic Arabidopsis embryo maturation, from dispersed threads to PB-like, compact structures. In mature seeds, 16γz and 27γz PBs appear very similar. However, when mature embryos are treated with a reducing agent, 27γz is fully solubilized, as expected, whereas 16γz remains largely insoluble also in reducing conditions and drives insolubilization of the ER chaperone BiP. These results indicate that 16γz expressed in the absence of the other zein partners forms aggregates in a storage tissue, strongly supporting the view that 16γz behaves as the unassembled subunit of a large heteropolymer, the PB, and could have evolved successfully only following the emergence of the much more structurally self-sufficient 27γz

    Environment and self-regulation in galaxy formation

    The environment is known to affect the formation and evolution of galaxies considerably best visible through the well-known morphology–density relationship. It is less clear, though, whether the environment is equally important at a given galaxy morphology. In this paper, we study the effect of environment on the evolution of early-type galaxies as imprinted in the fossil record by analysing the stellar population properties of 3360 galaxies morphologically selected by visual inspection from the Sloan Digital Sky Survey in a narrow redshift range ( 0.05 ≤z≤ 0.06 ). The morphological selection algorithm is critical, as it does not bias against recent star formation. We find that the distribution of ages is bimodal with a strong peak at old ages and a secondary peak at young ages around ∼2.5 Gyr containing about 10 per cent of the objects. This is an analogue to 'red sequence' and 'blue cloud' identified in galaxy populations usually containing both early- and late-type galaxies. The fraction of the young, rejuvenated galaxies increases with both decreasing galaxy mass and decreasing environmental density up to about 45 per cent, which implies that the impact of environment increases with decreasing galaxy mass. The rejuvenated galaxies have lower α/Fe ratios than the average and most of them show signs of ongoing star formation through their emission line spectra. All objects that host active galactic nuclei in their centres without star formation are part of the red sequence population. We confirm and statistically strengthen earlier results that luminosity weighted ages, metallicities and α/Fe element ratios of the red sequence population correlate well with velocity dispersion and galaxy mass. Most interestingly, however, these scaling relations are not sensitive to environmental densities and are only driven by galaxy mass. We infer that early-type galaxy formation has undergone a phase transition a few billion years ago around z∼ 0.2 . A self-regulated formation phase without environmental dependence has recently been superseded by a rejuvenation phase, in which the environment plays a decisive role possibly through galaxy mergers and interactions
