Universidad Libre -- Ingenieria -- Ingenieria de sistemasThe presentation graphically shows a series of initial tests to develop an artificial intelligence-assisted legal consultancy. Introduction to transcription factor structure and function

    No full text
Transcription of eukaryotic genes is regulated by DNA binding proteins known as transcription factors. These factors interact specifically with sequences located in the promoter regions of the genes they regulate. Transcription factors are classified in families according to the structure of their DNA binding domain. Some transcription factor families are present in most eukaryotes, while others are specific of certain lineages, suggesting that they are more recent acquisitions. In addition to the DNA binding domain, transcription factors possess other domains involved in activating or repressing gene expression, dimerization and establishing protein-protein interactions. Transcription factor action is finely modulated by mechanisms involving their synthesis, subcellular location and activity, usually through the interaction with other proteins and/or post-translational modifications. In this chapter, we summarize general properties of eukaryotic transcription factors as an introduction to the specific chapters dealing with the structure and function of plant transcription factors of this book.Fil: Gonzalez, Daniel Hector. This Research Topic is aimed at establishing a collection of articles that focus on the different processes involved in the biogenesis of respiratory complexes in plants as a means to highlight recent advances. In this way, it intends to help to construct a picture of the whole process and, not less important, to expose the existing gaps that need to be addressed to fully understand how plant cells build and modulate the complex structures involved in respiration.Fil: Gonzalez, Daniel Hector. The Effect of Ambient Ageing on the Corrosion Protective Properties of a Lithium-Based Conversion Layer

    No full text
The influence of ageing under ambient conditions on the corrosion protective behaviour of a lithium-based conversion layer on AA2024-T3 is studied in this work. Conversion layers aged at ambient conditions for relatively short times (0 h and 4 h), show an initial high degree of corrosion inhibition but a much lower protectiveness after the inhibition stage terminates. Conversion layers with relatively long ageing times (24 h and 72 h) show a rather stable corrosion resistance which is higher than that of short-time aged samples. It is hypothesized that the freshly-formed conversion layer has trapped a certain amount of lithium ions and water molecules, leading to ongoing and heterogeneous growth of the conversion layer with time under ambient indoor conditions. Moreover, conversion layers with short ageing times show early-stage active corrosion protection by lithium-ion release.Green Open Access added to TU Delft Institutional Repository 'You share, we take care!' - Taverne project https://www.openaccess.nl/en/you-share-we-take-care Otherwise as indicated in the copyright section: the publisher is the copyright holder of this work and the author uses the Dutch legislation to make this work public.Team Arjan MolTeam Yaiza Gonzalez GarciaTeam Peyman Taher

Cytochrome c, a hub linking energy, redox, stress and signalling pathways in mitochondria and other cell compartments

    No full text
Cytochrome c (CYTc) is a soluble redox-active heme protein that transfers electrons from Complex III to Complex IV in the cyanide-sensitive mitochondrial respiratory pathway. CYTc biogenesis is a complex process that requires multiple steps until the mature active protein is obtained. CYTc levels and activity are finely regulated, revealing the importance of this protein not only as electron carrier but also in many other processes. In this review, we describe the role of CYTc in mitochondrial respiration, from its canonical role as electron carrier for ATP production to its involvement in protein import and the stabilization of respiratory complexes and supercomplexes. In plants, CYTc is connected to the synthesis of the antioxidant ascorbate and the detoxification of toxic compounds. Finally, CYTc is also a multi-functional signalling molecule that influences the balance between life and death, acting in energy provision for cellular functions or triggering programmed cell death. The confluence of several metabolic routes into a single protein that links redox reactions with energy producing pathways seems logical from the point of view of cellular economy, control and organization.Fil: Welchen, Elina. Neurocysticercal antigens stimulate chemokine secretion from human monocytes via an NF-kappaB-dependent pathway.

Neurocysticercosis, infection with larval Taenia solium, is a common, serious neuroparasitic infection. Larval degeneration results in inflammatory cell influx and granuloma formation which leads to clinical symptomatology. The role of chemokines in such cell influx is unknown. We demonstrate that monocyte stimulation by T. solium larval antigen (TsAg) results in a differential profile of CXCL8/IL-8 (146.5+/-8.5ng/ml after 24h), CCL2/MCP-1 (267+/-4 ng/ml after 48 h) and CCL3/MIP-1alpha (1.72+/-0.43 ng/ml after 8 h) secretion. There was coordinate mRNA accumulation reaching maximum at 1h for CCL3 and 2 h for CXCL8 and CCL2. TsAg induced maximal nuclear binding of p65, p50 and c-rel subunits of the transcriptional regulator NF-kappaB by 2 h. IkappaBalpha but not IkappaBbeta was degraded within 10 min before resynthesis by 2 h. Pre-treatment with the broad-spectrum NF-kappaB inhibitor pyrrolidine dithiocarbamate caused complete abrogation of TsAg-induced CCL2 secretion (p=0.005) and 91% reduction of CXCL8 secretion (p=0.0003). TsAg was unable to induce CXCL8 promoter activity in Toll-like receptor (TLR)-2 or TLR-4/MD-2 transfected HeLa cells in the absence of lectins or other adaptor molecules. In summary, our data demonstrate that TsAg induces chemokine secretion via specific pathways dependent on NF-kappaB but not TLR-4/TLR-2, and indicate a potential mechanism whereby larval degeneration results in brain inflammation

Amidinoquinoxaline N-oxides: Synthesis and activity against anaerobic bacteria

    No full text
We present herein an in-depth study on the activity of amidinoquinoxaline N-oxides 1 against Gram-positive and Gram-negative anaerobic bacteria. Based on 5-phenyl-2,3-dihydropyrimidoquinoxaline N-oxide 1a, the selected structural variations included in our study comprise the substituents α− to the N-oxide function, the benzofused ring, substitution and quaternization of the amidine moiety, and the amidine ring size. Compounds 1 showed good to excellent antianaerobic activity, evaluated as the corresponding CIM50 and CIM90 values, and an antimicrobial spectrum similar to metronidazole. Six out of 13 compounds 1 had CIM90 values significantly lower than the reference drug. Among them, imidazoline derivatives 1i-l were the most active structures. Such compounds were synthesized by base-promoted ring closure of the corresponding amidines. The N-oxides under study showed no significant cytotoxicity against RAW 264.7 cells, with high selectivity indexes. Their calculated ADME properties indicate that the compounds are potentially good oral drug candidates. The antianaerobic activity correlated satisfactorily with the electron affinity of the compounds, suggesting that they may undergo bioreductive activation before exerting their antibacterial activity.Fil: Gruber, Nadia. Spatiotemporally resolved corrosion protection of AA2024-T3 by a lithium-based conversion layer

    No full text
In this work, the corrosion mechanism of AA2024-T3 covered by a lithium-based conversion layer is studied with high spatial and temporal resolution. Although the aluminium alloy surface is protected by a multi-layered conversion layer, areas around intermetallic phases (IMPs) represent weak spots due to an insufficient generation of a protective inner dense layer. For the freshly formed conversion layer, both the top and the inner layer undergo a gradual dissolution upon exposure to relatively dilute NaCl solution within 2 h due to their chemical instability. For the ambiently-aged conversion layer, most corrosion activity around IMPs is related to the S-phase and large constituent phases, due to their active nature and the lower local conversion layer quality, respectively. Moreover, S-phase-related corrosion activity lasts approximately 8 h due to fast dissolution whereas reactions induced by large constituent particles remain active over the entire re-immersion period of 12 h.Team Arjan MolTeam Yaiza Gonzalez Garci

A method for detecting the profile of an author

    No full text
This paper presents a method for detecting an author’s profile using the following two elements: gender and age. This is based on a set of dialogues, written in two languages: English and Spanish, provided for Author Profiling competence within the evaluation forum "Uncovering Plagiarism, Authorship, and Social Software Misuse" (PAN2018). Counts of lexical, semantic, and syntactic characteristics are used to generate a two-phase classification system, which first classifies gender and then age. The results obtained show that, with the amount of data available, it is possible to characterize both the age and gender of an author with an accuracy greater than 50%. However, these values could be improved by having more evidence of information in the training data.Silva, JesusGarcía, SilviaBinda, María AlejandraMarin Gonzalez, FredyBarrios, RosioLeon Castro, Bellani
