    Selección de aislados de Bacillus spp. con fines agropecuarios e industriales a partir del bioproducto IHPLUS®

    El presente trabajo tuvo como objetivo seleccionar aislados de Bacillus sp. con características promotoras del crecimiento vegetal y con potencialidades industriales, a partir del producto IHPLUS®. Para el aislamiento se inocularon 10 mL del producto en medio Caldo Nutriente y posteriormente se realizó un golpe térmico, para iniciar el proceso a partir de formas productoras de esporas. Se realizaron diluciones seriadas y se realizaron siembras en medio Agar Nutriente. Los aislados fueron purificados y se verificó que se correspondieran con formas bacilares Gram positivas productoras de endosporas. Se evaluó la producción de ácido indolacético y la capacidad de los aislados de solubilizar fosfafos. Se determinaron las actividades quitinolítica y/o glucanolítica, lipídica, proteolítica, amilolítica, celulolítica y mananolítica. Se obtuvieron un total de cuatro aislados de Bacillus sp. Gram positivos y con capacidad de esporular. Los aislados IH7 e IH5 mostraron propiedades bioestimuladoras y biofertilizantes al producir ácido indolacético (23,55 μg.mL-1) y solubilizar fosfato tricálcico, respectivamente. El aislado IH7 evidenció potencialidades como agente antagónico de fitopatógenos ya que produjo cianuro de hidrógeno y actividades β-1,3-glucanolítica y/o quitinolítica (95,55 %) y proteolítica (43,98 %). IH5 mostró actividad amilolítica y mananolítica, mientras que IH7 es productor de amilasas y celulasas, lo cual indica que el producto IHPLUS® constituye una fuente para la obtención de aislados de Bacillus con aplicaciones en diferentes industrias

    Estudio de la expresión de la formaldehído deshidrogenasa dependiente de glutatión de Arabidopsis thaliana y su función en la patogénesis

    Consultable des del TDXTítol obtingut de la portada digitalitzadaLa formaldehído deshidrogenasa dependiente de glutatión (FALDH) (EC 1.2.1.1), conocida también como ADH clase III, es un enzima ubícuo, presente en el reino vegetal y animal. La eliminación del formaldehído dentro de la célula se realiza principalmente mediante la FALDH, que utiliza NAD+ como cofactor. El formaldehído es un intermediario del metabolismo celular normal, aunque puede tener también un origen exógeno, siendo un polucionante atmosférico y de aguas residuales. Por otra parte, el formaldehído se forma intracelularmente durante la peroxidación lipídica en situaciones de estrés oxidativo. La eliminación de este formaldehído, que es muy tóxico para la célula, es necesaria para su supervivencia. Recientemente se ha descubierto que la FALDH tiene también actividad S-nitrosoglutatión reductasa. El S-nitrosoglutatión (GSNO) es uno de los más abundantes metabolitos endógenos del oxido nítrico (NO), que actúa como uno de los señalizadores moleculares en los mecanismos de defensa de las plantas. En esta Tesis Doctoral hemos demostrado que la expresión de la FALDH está regulada por herida y por diferentes hormonas vegetales, tales como el ácido jasmónico y el ácido salicílico. También hemos generado plantas transgénicas de A. thaliana portadoras de una construcción antisentido, que presenta una importante disminución de la actividad enzimática FALDH, directamente relacionados con los niveles de proteína. Dichas plantas transgénicas tienen una menor capacidad de metabolizar formaldehído exógeno y presentan un aumento de la resistencia basal a patógenos fúngicos y bacterianos. Por otra parte, las plantas transgénicas con niveles de FALDH modificados (tanto las antisentido como las de sobreexpresión) muestran un fenotipo diferente, principalmente en la raíz. La inmunolocalización en tejidos reveló que la FALDH presenta un patrón de expresión en hojas y en raíz que es específico del tipo celular. A nivel subcelular, la FALDH está presente en citoplasma, núcleo y cloroplasto. Esta es la primera vez que se describe la localización cloroplástica de la FALDH.The glutathione-dependent formaldehyde dehydrogenase (FALDH) is the main enzyme of the formaldehyde detoxification system in eukaryotes. In Arabidopsis, it is coded by a single gene, which is constitutively expressed. To gain more insight into the functional role of this enzyme in plants, we have studied the spatial distribution of the enzyme at cellular and subcellular level. By immunolocalization experiments on root and leaf sections we have demonstrate that the pattern of expression of the enzyme is cell specific. At subcellular level, FALDH localizes in cytoplasm, nucleus and chloroplasts. We have demonstrated that Arabidopsis plants with reduced levels of FALDH, bearing antisense constructs, show a slower rate in formaldehyde elimination. These results confirm the central role of FALDH in formaldehyde metabolism in plants and have important implications in the phytoremediation of environmental formaldehyde. We observed also that the antisense transgenics lines were more resistance to the infection with pathogens (bacteria and fungus). The importance of FALDH has been greatly increased by the discovery of its potent activity toward S-nitrosoglutathione, the condensation product of glutathione and nitric oxide (NO). NO and NO-related metabolites, such as S-nitrosothiols (SNOs) play a central role in signal transduction and host defense. We have investigated the gene response to mechanical wounding and plant hormones involved in signal transduction, showing that the gene is down-regulated by wounding in a JA-dependent pathway, and that it is transcriptionally activated by salicylic acid. This is the first time that regulation of FALDH in response to signals associated with plant defense has been demonstrated

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
