Physical and functional interactions between STAT3 and Kaposi's Sarcoma-associated Herpesvirus-encoded LANA
Get PDFThe Kaposi’s sarcoma-associated herpesvirus (KSHV)-encoded latency-associated nuclear antigen (LANA) is known to modulate viral and cellular gene expression. We show that LANA directly associates with an IL-6 signal transducer, STAT3 and that LANA enhances the transcriptional activity of STAT3. Coimmunoprecipitation studies documented a physical interaction between LANA and STAT3 in transiently transfected 293T cells as well as the KSHV-infected primary effusion lymphoma (PEL) cell line. Furthermore, small-interfering RNA-mediated reduction of LANA expression decreased the STAT3-dependent transcription in KSHV-positive PEL cells, whereas overexpression of LANA enhanced STAT3 activity in KSHV-negative B lymphoma cells. These data demonstrate that LANA is a transcriptional co-activator of STAT3, and may have implications for the pathogenesis of KSHV-associated diseases

Kaposi's sarcoma-associated herpesvirus-encoded LANA associates with glucocorticoid receptor and enhances its transcriptional activities
Get PDFKaposi's sarcoma-associated herpesvirus (KSHV)-encoded latency-associated nuclear antigen (LANA), which interacts with cellular proteins, plays a central role in modification of viral and/or cellular gene expression. Here, we show that LANA associates with glucocorticoid receptor (GR), and that LANA enhances the transcriptional activity of GR. Co-immunoprecipitation revealed a physical interaction between LANA and GR in transiently transfected 293T and HeLa cells. In human B-lymphoma cells, LANA overexpression enhanced GR activity and cell growth suppression following glucocorticoid stimulation. Furthermore, confocal microscopy showed that activated GR was bound to LANA and accumulated in the nucleus, leading to an increase in binding of activated GR to the glucocorticoid response element of target genes. Taken together, KSHV-derived LANA acts as a transcriptional co-activator of GR. Our results might suggest a careful use of glucocorticoids in the treatment of patients with KSHV-related malignancies such as Kaposi's sarcoma, primary effusion lymphoma, and multicentric Castleman disease. She took part in the Fortissat Science Alliance podcast recordings in March 2021.What is the Fortissat Science Alliance?The Fortissat Science Alliance was a Wellcome Trust & Children In Need "Curiosity" project. This scheme provided informal STEM learning opportunities for young people who attended the community centre Getting Better Together Shotts (GBT Shotts) between 2019 and 2023. Due to the COVID-19 pandemic, deliveries had to pivot online so the podcast was founded. These recordings were made via Zoom with warm-up STEM activities sent to every young person in advance, along with a profile page for each researcher, so that they were relaxed and able to ask excellent questions.Link to episode on Spotify.Depending on the broadcast date, podcast deliveries were co-sponsored by Glasgow Science Festival, EXPLORATHON 2021, or EXPLORATHON 2022/23.For the duration of the project, it was supported jointly by Children in Need and the Wellcome Trust. These three components are closely interconnected and are evolving strongly and rapidly due to both widespread practices and technological developments. On the one hand, authorship is becoming increasingly blurred and less tied to writing; on the other, Artificial Intelligence systems appear increasingly capable of intervening heavily in writing, if not even replacing the human author; and the role of the reviewer, already difficult, is becoming even more complicated. We need to reflect in a manner appropriate to the times on what authorship, writing and peer review are and what we want them to be.La producción científica pone en juego la autoría, la escritura y la revisión por pares. Estos tres componentes están estrechamente interconectados y evolucionan con fuerza y rapidez debido tanto a las prácticas generalizadas como a los avances tecnológicos. Por un lado, la autoría es cada vez más difusa y está menos vinculada a la escritura; por otro, los sistemas de Inteligencia Artificial parecen cada vez más capaces de intervenir fuertemente en la escritura, cuando no incluso de sustituir al autor humano; y el papel del revisor, ya de por sí difícil, se complica aún más. Get PDFEditada en la Universidad Carlos IIISe trata de llamar la atención en e este artículo sobre el papel de primera fila que jugaron los grandes señores de ganados en la actividad ganadera trashumante en el ámbito que abarcaba la cuadrilla mesteña de Soria durante los siglos XV y XVI. Para ello se establecen comparaciones entre el tamaño de sus rebaños y los de otros medianos y pequeños propietarios de la región, teniendo en cuenta entre otros indicios las noticias sobre sus ventas de lanas. Al mismo tiempo se profundiza en su caracterización desde el punto de vista sociopolítico, matizando algunas hipótesis que se han venido manejando para definir la base social mesteña y su evolución a lo largo del siglo XVI. Y por fin se repasan algunos aspectos referentes a la gestión de las explotaciones ganaderas trashumantes, que revelan la presencia de numerosos factores que hacían más rentables a las grandes frente a las medianas y pequeñas.Big sheep owners played a key role in nomadic sheeprearing in the Spanish region around Soria during the XVth. and XVIth. centuries. Century. Lastly, some aspects of the management of sheeprrearing enterprises are examined, and its factors identified. the Gramscian writings hidden in the huge corpus of unsigned articles published by the newspapers where Gramsci usually wrote. The study adopts quantitative authorship attribution methods, which were extensively tested for this specific study on these texts, yielding very interesting results.
A very brief history of quantitative methods for authorship attribution is drawn, with a focus on those used for this study: both based on a mathematical model of texts and the author/text relationship, and both using similarity distances (the first compares the statistics for sequences of n characters – n-grams – in the texts, while the second is based on the concept of entropy of a symbolic sequence). These methods have until now usually been in the toolbox of textual scholars and historians who could fear being dispossessed of their role and interpretative authority, but things are far more complex
