Early Chronic Memantine Treatment-Induced Transcriptomic Changes in Wild-Type and Shank2-Mutant Mice

Shank2-mutant mice with a homozygous deletion of exons 6 and 7 (Shank2-KO mice) show decreased NMDA receptor (NMDAR) function and autistic-like behaviors at juvenile [∼postnatal day (P21)] and adult (>P56) stages that are rescued by NMDAR activation. However, at ∼P14, these mice show the opposite change – increased NMDAR function; moreover, suppression of NMDAR activity with early, chronic memantine treatment during P7–21 prevents NMDAR hypofunction and autistic-like behaviors at later (∼P21 and >P56) stages. To better understand the mechanisms underlying this rescue, we performed RNA-Seq gene-set enrichment analysis of forebrain transcriptomes from wild-type (WT) and Shank2-KO juvenile (P25) mice treated early and chronically (P7–21) with vehicle or memantine. Vehicle-treated Shank2-KO mice showed upregulation of synapse-related genes and downregulation of ribosome- and mitochondria-related genes compared with vehicle-treated WT mice. They also showed a transcriptomic pattern largely opposite that observed in ASD (reverse-ASD pattern), based on ASD-related/risk genes and cell-type–specific genes. In memantine-treated Shank2-KO mice, chromatin-related genes were upregulated; mitochondria, extracellular matrix (ECM), and actin-related genes were downregulated; and the reverse-ASD pattern was weakened compared with that in vehicle-treated Shank2-KO mice. In WT mice, memantine treatment, which does not alter NMDAR function, upregulated synaptic genes and downregulated ECM genes; memantine-treated WT mice also exhibited a reverse-ASD pattern. Stretchable, soft, and wireless biomedical electronics
Electrophysiology that records various bio-signals from living organisms has broad impacts on clinical diagnosis and study. Having a recording system, effectively small and compatible with wireless communication, not only leads to the new environment in which health monitoring can commonly take place outside the hospital with portable recording devices, but also contributes to development in neuroscience by enabling studies on neural activity with the help of an implantable neural interface. This thesis presents a soft, stretchable, and wireless biomedical electronics system for both skin-mountable and implantable applications. The wireless capability associated with Bluetooth Low Energy (BLE) allows continuous monitoring in portable devices in user-friendly environments. This machine has a 3 KJ, 600 KV, 12 stages Marx generator that charges 6.8 ohm water filled pulseforming line, which is then connected to a load through a self breakdown spark switch. The triggering characteristics of Marx generator is shown to be very stable and output reproducibility is also fairly good. This peaked pulse generator is designed to be versatile in output parameters of polarity, pulse length and impedance. It is shown that the energy conversion efficiency of "SNU" versatile electron beam accelerator from the Marx generator to the electron beam is reached up to about 63% in a condition where the gas pressures of Marx and pulse line switches are 0.3 Kg/㎠ and 3 Kg/㎠ , respectively, and the vacuum of diode chamber is Torr.
The steady-state one dimensional theoretical model of the intense relativistic electron beam propagation within the vacuum diode and its numerical results are presented. The characteristic curves of injection current density and its beam radius are exactly obtained as a function of the diode voltage. The experimental investigation of intense relativistic electron beam propagation in vacuum diode has also been reported. The beam current rises to its peak value 30 KA in about 60 nsec, which is nearly same as pulse width, and then decayed without any positive part of tail oscillation. It is shown that the virtual cathode is not formed in vacuum diode regime from the both theoretical and experimental results.
The steady-state equilibrium properties of bipolar electron and ion flow in a relativistic planar diode were calculated within the framework of the cold-fluid-Poisson equations. Generation of Background Model Image Using Foreground Model
Proper consideration of the temporal domain and the spatial domain is essential to perform robust foreground object detection in visual surveillance. However, there are difficulties in considering long-term temporal information with CNN-based methods. To solve this limitation, classical algorithms and some deep learning-based algorithms have used a background model image. However, acquiring a sophisticated background model image is also one of the complex problems. Most of the algorithms take a lot of time to initialize the background model image and generate many errors in the presence of a static foreground. This paper proposes an algorithm for generating a background model image using a deep-learning-based segmenter to solve this problem. The proposed method shows a 66.25% lower mean square error (MSE) than the background subtraction (BGS) algorithm and 79.25% lower than the latest deep learning algorithm in the SBI dataset. In addition, in the deep learning-based segmenter that uses a background image as input, replacing the background image of BGS algorithm with the background image of the proposed method shows a 38.63% reduction in the false detection rate (PWC)
