VIABILIDADE ECONÔMICA DA UTILIZAÇÃO DO PÓ DE ROCHA NA CULTURA DO BRÓCOLIS (Brassica oleracea)
As atividades olerícolas sempre foram de fundamental importância para a sustentabilidade de pequenas propriedades agrícolas das quais a principal característica baseia-se no emprego de mão de obra familiar em suas atividades. São pequenas produções constituídas principalmente de produtos agroecológicos, os quais são de ótima qualidade, mais saudáveis e sem a presença de resíduos tóxicos e a venda é feita diretamente ao consumidor, aumentando a renda familiar, eliminando o atravessador e havendo consequente aumento da renda na propriedade. A presente pesquisa teve por objetivo avaliar a produtividade e os teores de nitrogênio, fósforo e potássio na produção de brócolis de cabeça submetido a diferentes doses de pó de rochas, visando à possibilidade da utilização desse pó em sistemas produtivos. Além da produtividade do brócolis por hectare, foram avaliados os componentes de rendimento do brócolis, sendo: diâmetro das inflorescências, diâmetro do caule, altura de planta (cm), medindo desde a superfície do solo até a inflorescência, e valor nutritivo. No Brasil, a utilização de pó de rocha é pouco difundida e está restrita a zonas próximas às minas de extração, sendo essa prática direcionada a cultivos orgânicos. O pó de rocha possui baixa relação PK e nenhum N, inviabilizando, muitas vezes, o transporte do produto a longas distâncias o que aumentaria o custo de produção. No entanto, em razão da liberação lenta do pó de rocha, espera-se um efeito residual ao solo, o qual viabilizará a produção de culturas subsequentes, o que será estudado com a análise de solo posterior à colheita do brócolis. Nesse sentido, salienta-se que até o presente momento não foram realizados experimentos demonstrando a capacidade produtiva de olerícolas, especialmente brócolis, com diferentes doses de pó de rocha nas condições climáticas e de solo em que o experimento foi conduzido. Estudos que demonstram o real aumento da produtividade do brócolis com adição de pó de rocha são de fundamental importância para definir estratégias e introduzir tecnologias a produtores que se enquadram nesse perfil tecnológico. O pó de rocha é uma alternativa ecologicamente correta e vem se tornando uma alternativa a produtores orgânicos em todo o país. Além disso, a adoção de tecnologias que visem ao aumento da produtividade de pequenos produtores, com baixo custo econômico e ambiental é necessária para a manutenção da viabilidade agrícola.Palavras-chave: Brassica olerace. Expression In Stressed Mouse And Human Cardiomyocytes
Transcriptional changes in cardiomyocytes drive heart failure progression, however, precise control over endogenous gene expression remains challenging. The expression of Krueppel-like factor 15 ( KLF15 ), an evolutionary conserved nuclear and cardiomyocyte specific inhibitor of WNT/CTNNB1 signalling in the heart, is lost upon cardiac remodelling, and accompanied by aberrantly active WNT/CTNNB1 resulting in heart failure progression. We investigated KLF15 expression dynamics employing CRISPR/Cas9-based tools in mouse cardiomyocytes in vivo and in human induced pluripotent stem cell derived cardiomyocytes (hiPSC-CM) under the hypothesis that re-establishment of KLF15 levels in myocardial stress conditions prevents heart failure progression. Using a mouse model expressing enzymatically inactive Cas9 (dCas9) fused to transcriptional activators (VPR) under Myh6 -promoter control, we activated Klf15 in a murine pressure overload model by transverse aortic constriction. Delivery of Klf15 gRNAs targeted to the Klf15 promoter region via AAV9 induced Klf15 expression sufficiently to re-normalize Klf15 expression to transcript levels comparable to sham surgery hearts. This was accompanied by reduced decrease of fractional shortening as well as reduced cardiomyocyte hypertrophy in stressed Klf15 re-activated hearts compared to non-trageted (NT) gRNA hearts (n=3-8 per group, echo data from 4 and 8 weeks post-surgery). We achieved titratable KLF15 activation in dCas9VPR transgenic hiPSC-CM by selection of single and multiple gRNAs (n=3-4 replicates) and used these cells to generate human engineered myocardium by combining hiPSC-CM and fibroblasts which we subjected to isometric contractions in order to induce mechanical stress, which resulted in KLF15 expressional decrease in line with our in vivo data. This transcriptional loss was rescued in CRISPR/dCas9VPR hiPSC-CM targeted to the KLF15 locus compared to controls (n=6-9/2/4 tissues per group/casting sessions/differentiations). Additionally, TGFB1 induced cardiomyocyte stress resulted in decreased KLF15 expression levels in 2D hiPSC-CM cultures which were rescued by dCas9VPR- KLF15 targeting (n=3 experiments). In conclusion, we report controllable gene activity by CRISPR/dCas9VPR to restore the loss of KLF15 in stressed mouse and human cardiomyocytes. VIABILIDADE ECONÔMICA DA UTILIZAÇÃO DO PÓ DE ROCHA NA CULTURA DO BRÓCOLIS (BRASSICA OLERACEA)
A olericultura, mais especificamente as atividades relacionadas a cultura de brócolis (Brassica oleracea) vem adquirindo grande importância nos últimos anos na região Oeste de Santa Catarina Além de contribuir significativamente na economia de pequenas propriedades, seja pela venda direta aos consumidores ou pela comercialização nos mercados e feiras da região. A presente pesquisa avaliou os teores de nitrogênio no tecido vegetal seco e o rendimento do brócolis na produção de brócolis de cabeça única, (cv. Lord Summer) submetida a diferentes doses de pó de rochas. Além dos componentes de rendimento do brócolis, sendo: Diâmetro do caule (cm), Altura de planta (cm), medindo desde a superfície do solo até a inflorescência e Massa Verde. Para tanto os 05 tratamentos foram distribuídos em blocos casualizados possuindo 04 repetições totalizando 20 parcelas, sendo D02 0,8kg de pó de rocha por parcela, o equivalente a 2t de pó de rocha ha¹, D04 1,6kg/parcela ou 4t ha¹, D08 3,2kg/parcela ou 8t ha¹, D12 4,8kg/parcela ou 12t ha¹. A área da parcela 4m² (2m de largura X 2m de comprimento) totalizando uma área total de 16m² por tratamento, com espaçamento de 50cm entre linhas e entre plantas totalizando uma área útil de 2m² por parcela e 8 m² por tratamento, utilizando 16 plantas uteis por tratamento. Os resultados da pesquisa apontam que a tecnologia da rochagem, que é considerada uma tecnologia social ou de baixo impacto, tem um papel importante na conservação dos solos e diminuição dos custos de produção nas culturas olerícolas, porém a rochagem deve ser implantada como uma prática corretiva para o manejo do solo, pode ser considerada uma modalidade de investimento a longo prazo, como a calagem ou fosfatagem corretiva, inicialmente deve ser associada a adubação orgânica ou química, existe a necessidade ainda de adequar a dosagem e utilização destes para cada cultura de modo a permitir um incremento de renda e de segurança alimentar e nutricional.Palavras-chave: Brócolis, Pó de rocha, Nitrogênio, Rendiment
Abstract 498: Krueppel-like Factor 15 Modulation: A Crispr-based Re-establishment of Homeostatic Transcription in Diseased Mouse and Human Cardiomyocytes
An altered transcriptomic profile of cardiomyocytes is a hallmark of cardiac remodelling and heart failure. Re-activation of gene programs necessary for myocardial homeostasis and inhibition of deleterious signalling pathways, such as Wnt/beta-catenin signalling, is therefore therapeutically desirable, however challenging to achieve tissue-specifically. We identified Krueppel-like factor 15 (KLF15) as a cardiac specific inhibitor of Wnt/beta-catenin signalling, lost during heart failure progression in the mouse and human heart. To re-establish endogenous Klf15 expression, we designed a cardiomyocyte specific CRISPR activation mouse model expressing enzymatically inactive Cas9 (dCas9) fused to a transcriptional activator (VPR) under Myh6 promoter control. The dCas9VPR protein is directed by guide RNAs (gRNA), delivered by AAV9, to the promoter region of Klf15 and induces gene expression. We tested activation of Klf15 in the adult myocardium by injecting 15 weeks old Myh6-dCas9VPR mice systemically with AAV9 carrying Klf15 gRNAs and increasing Klf15 transcript levels 8 weeks later up to 1.6 fold compared to controls. Since we found a 0.5 fold change of Klf15 expression upon transverse aortic constriction compared to sham operated mice, we expect the CRISPR activation induced Klf15 activation to be sufficient to re-establish Klf15 expression. As a translational approach and since we demonstrated KLF15-WNT regulation in human cardiomyocytes, we integrated dCas9VPR into human induced pluripotent stem cells (hIPSC) by targeted genome editing at the AAVS1 safe harbour locus and confirmed dCas9VPR expression in hIPSC and in hIPSC-derived cardiomyocytes (hIPSC-CM). We tested KLF15 gRNAs in HEK293 cells and selected the most efficient candidate gRNAs to test Klf15 activation in dCas9VPR-IPSC resulting in an up to 1.7 fold activation compared to controls. For gene activation in dCas9VPR hIPSC-CM, we are currently testing lentivirus constructs to deliver up to 3 gRNAs simultaneously to tailor KLF15 expression. With these prerequisites, we aim to re-activate master regulators of normal cardiomyocyte function in failing cardiomyocytes at physiologically relevant levels, such as KLF15, to test potential therapeutic targets for the prevention of heart failure progression
