The interaction of RecA with both CheA and CheW is required for chemotaxis

As in other microorganisms, chemotaxis affords key physiological benefits, including enhanced access to growth substrates, but also plays an important role in infection and disease. Chemoreceptor signaling core complexes, consisting of CheA, CheW and methyl-accepting chemotaxis proteins (MCPs), modulate the switching of bacterial flagella rotation that drives cell motility. These complexes, through the formation of heterohexameric rings composed of CheA and CheW, form large clusters at the cell poles. RecA plays a key role in polar cluster formation, impairing the assembly when the SOS response is activated. In this study, we determined that RecA protein interacts with both CheW and CheA. The binding of these proteins to RecA is needed for wild-type polar cluster formation. In silico models showed that one RecA molecule, attached to one signaling unit, fits within a CheA-CheW ring without interfering with the complex formation or array assembly. Activation of the SOS response is followed by an increase in RecA, which rises up the number of signaling complexes associated with this protein. This suggests the presence of allosteric inhibition in the CheA-CheW interaction and thus of heterohexameric ring formation, impairing the array assembly. STED imaging demonstrated that all core unit components (CheA, CheW, and MPCs) have the same subcellular location as RecA. Activation of the SOS response promotes the RecA distribution along the cell instead of being at the cell poles. CheA- and CheW- RecA interactions are also crucial for chemotaxis, which is maintained when the SOS response is induced and the signaling units are dispersed. Our results provide new molecular-level insights into the function of RecA in chemoreceptor clustering and chemotaxis determining that the impaired chemoreceptor clustering not only inhibits swarming but also modulates chemotaxis in SOS-induced cells, thereby modifying bacterial motility in the presence of DNA-damaging compounds, such as antibiotics

Perspectivas interdisciplinarias: contribuciones significativas desde el campo editorial, la ilustración y el emprendimiento al medio cultural
A lo largo de su trayectoria como diseñadora gráfica profesional, la
autora del presente informe incursiona en diversos sectores del mercado
laboral, destacándose especialmente en el diseño editorial y el branding. El
objetivo principal de este informe es recapitular y respaldar su desempeño
profesional alineado con sus motivaciones personales. En el presente
documento se detallan, de manera concisa, tres proyectos seleccionados que
abarcan los últimos siete años de su carrera. En primer lugar, se analiza el rol
desempeñado en el diseño editorial para más de veinte publicaciones bajo el
sello editorial del Fondo de Cultura Económica (Perú), utilizando cuatro como
ejemplos, y se destaca cómo esta experiencia ha constituido una plataforma
de aprendizaje para proyectos futuros. Seguidamente, se explor en detalle su
contribución al diseño de marca, planificación, conceptualización y gestión de
contenido para el perfil de Instagram de Vuelo Ámbar Print Studio, espacio
especializado en grabado y letterpress y cofundado por la autora, Giselle
Adrianzén, junto con Diego Cabanillas, (grabador de la PUCP), resaltando
también su labor como directora artística para diversos proyectos. Por último,
se examina su aporte como ilustradora editorial para la reedición del libro
"Autobiografía de un Yogui" de Paramahansa Yogananda, destacando cómo
sus ilustraciones enriquecen la interpretación reflexiva del lector, según la
editora del proyecto, Cindy Torrejón. Cada proyecto está respaldado con
material gráfico que ilustra los resultados del diseño, detallando la
metodología empleada para fundamentar sus aspectos estéticos,
comunicacionales, técnicos y funcionales.Throughout her career as a professional graphic designer, the author of
this report has ventured into various sectors of the job market, particularly
excelling in editorial design and branding. The main objective of this report is
to recapitulate and support her professional performance aligned with her
personal motivations. This document will detail, concisely, three selected
projects spanning the last seven years of her career. Firstly, the role played in
editorial design for over twenty publications under the editorial imprint of Fondo
de Cultura Económica (Peru) will be analyzed, utilizing four as examples, and
emphasizing how this experience has served as a learning platform for future
projects. Subsequently, her contribution to brand design, planning,
conceptualization, and content management for the Instagram profile of Vuelo

Análisis de la representación romántica en el género de cómics Boys Love (BL) de horror como subversión del Boys Love (BL) idílico para el diseño de una propuesta innovadora y creativa
Como tópico, el Boys Love (BL) de horror no tiene una exploración a nivel
académico, en contraste con su contraparte general, siendo esta el BL regular; sin
embargo, aunque este tema sí es tocado por textos académicos, el análisis no suele adentrarse en el diseño visual de estos textos narrativos. El objetivo principal de esta investigación es describir la idealización de la representación romántica en el género BL de cómics que crea estereotipos sobre cómo debe representarse a los hombres homosexuales en los medios de entretenimiento en comunidades latinas de
Facebook y su efecto en la recepción del cómic BL de horror en estos círculos. Los objetivos secundarios son analizar las consecuencias de la solidificación del anti en espacios latinos e investigar la censura en la distribución del BL de horror en
espacios occidentales. Esta investigación se enfoca en definir qué es el género BL y BL de horror, y analizar las características de este subgénero. Se utilizan métodos
cualitativos y cuantitativos. Se genera un producto visual digital e impreso que
pertenece al género BL de horror y responde como subversión a los tropos habituales de los géneros BL y BL de horror. Se concluye que el análisis de la
representación romántica del género de cómics BL de horror como subversión del
BL idílico permite el diseño de una propuesta innovadora y creativa a nivel narrativo, gráfico y editorial

Truncated, non-networking versions of the coupling protein chew retain chemoreceptor control of knase cheA
Bacterial chemoreceptors control the activity of the associated CheA kinase in response to chemical gradients and, consequently, regulate the swimming behavior of the cell. However, such control is not direct but requires the participation of the essential coupling protein CheW, which is structurally homologous to the carboxy-terminal domain of the kinase. The actual role of this small coupling protein is somehow intriguing. It has been demonstrated that it is absolutely essential for chemoreceptor control of the kinase, in spite of the occurrence of direct contacts between chemoreceptors and CheA. In addition, CheW plays an essential role in the assembly of the large macromolecular arrays that combine chemoreceptors of different specificities, and it is therefore responsible for molecular interactions that provide such arrays with remarkable signaling properties. In this work, we analyze truncated CheW derivatives that are still able to control the kinase but have lost the ability to connect signaling units. We demonstrate that these two activities can work separately and speculate about the significance of the roles of these two different activities in the context of the chemoreceptor cluster.Fil: Pedetta, Andrea. Spatially-localized time dependent solutions including turbulence and their interactions in 2D Kolmogorov flow
In 2D Kolmogorov flow in small aspect ratio domains, spatially-localized solutions such as kink, traveling or time-dependent kink-antikink pars coexist. However, the conservation of the flow rate in the y direction strongly restrict combination of localized solutions and their positioning. We find that by adding a homogeneous flow U y their positioning is controlled and each of localized solutions including a spatially-localized chaos is isolated. Numerical results suggest that these isolated solutions can be elements constructing a whole flow
