Evolution of executive and permissive mechanisms of paradoxical sleep during aging in the rat : implication of the inducible NO-synthase : effects on the stress response : recovery of the stress response with an antidepressant
Cette étude a analysé l’influence qu’exerce le vieillissement sur les mécanismes du sommeil chez le rat. Dans un premier temps, nous avons abordé le rôle joué par la NO-synthase inductible (iNOS) dans la régulation du sommeil paradoxal (SP). Nous rapporterons que l’injection d’un inhibiteur spécifique de la iNOS dans le tegmentum latérodorsal (LDT), inhibe l’apparition du SP chez le rat âgé (24 mois) mais pas chez l’adulte (4 mois). Cet effet est antagonisé par une injection secondaire d’un donneur de NO. Les corps cellulaires exprimant la iNOS sont localisés dans des noyaux voisins du LDT, mais, seuls des prolongements neuronaux expriment la iNOS dans ce noyau. Aucune expression de la iNOS n’est apparente dans le cerveau du rat adulte. Ces résultats sont la première preuve du rôle joué par la iNOS dans la régulation du SP chez l’animal âgé. Nous avons ensuite comparé la capacité des rats adultes et âgés à répondre à un stress d’immobilisation sur le plan polysomnographique. Les résultats obtenus confirment que, chez le rat adulte, le stress d’immobilisation d’une heure, imposé en début de période d’obscurité, provoque un rebond de SP. Chez l’animal âgé, au contraire, le stress d’immobilisation reste sans effets sur les états de veille et de sommeil. Enfin, nous avons tenté de mettre en évidence si l’agomélatine, un antidépresseur de nouvelle génération, était capable de compenser l’absence de rebond de SP observée chez l’animal âgé après un stress d’immobilisation. Les résultats obtenus indiquent que l’agomélatine, administrée au début de la période d’obscurité pendant 3 jours consécutifs, pouvait vraisemblablement favoriser la réapparition d’un rebond de SPThis study analysed the influence exerted by aging on the rat sleep-wake cycle. We first considered the part played by the inducible NO-synthase (iNOS) in the regulation of paradoxical sleep (PS or REM sleep). We report that the injection of an iNOS inhibitor, within the laterodorsal pontin tegmentum (LDT), suppresses the occurrence of PS in the aged animals (24 months), but not in the adult ones (4 months). This effect can be reversed by a secondary injection of a NO donor. While the cell bodies expressing the iNOS are localized in the structures neighbouring the LDT, only nerves ending impinge this nucleus. The iNOS is not expressed in the brain of the adult rat. These data constitute the first demonstration of the part played by the iNOS in the regulation of PS in the aged animal. Afterwards, we compared the ability of adult and aged rats to reply to an immobilization stress by a sleep rebound. Data obtained confirm that a sleep rebound occurs after a 1h immobilization stress in the adult rat. In the same conditions, such a sleep rebound is not observed in the aged animal. Finally, we checked whether agomelatine, a new type of antidepressant, was able to compensate the loss of the sleep rebound observed in the aged rat after an immobilization stress. Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. Mechanisms of stress vulnerability : pharmacologic and naturalistic approaches
Le syndrome de stress post-traumatique est une pathologie caractérisée par des symptômes de réexpérimentation, d’évitement et d’hypervigilance. L’apparition d’un syndrome de stress posttraumatique après un événement traumatisant peut être prédit par un déficit de sécrétion en glucocorticoïdes. Le rôle de ce déficit dans la vulnérabilité individuelle face au stress a été analysé par deux approches différentes chez le rat. Dans une première approche, une déplétion pharmacologique a été induite par la métyrapone puis les réactions physiologiques et comportementales pendant le stress ont été caractérisées. Dans une seconde approche, les différences interindividuelles d’une population soumise à un stresseur intense ont été caractérisées en termes de réponse physiologique, comportementale et d’expression génique dans l’hippocampe. Ces études n’ont pas mis en évidence de vulnérabilité accrue induite par un déficit en glucocorticoïde. Au contraire, la métyrapone a montré des propriétés anxiolytiques indépendamment de son effet sur les glucocorticoïdes. La métyrapone a également modifié le métabolisme central et périphérique d’une façon qui pourrait expliquer ses propriétés neuroprotectrices. Enfin, l’approche naturaliste a mis en évidence l’importance de la plasticité synaptique dans la résilience face au stressPost-traumatic stress disorder is a pathology characterized by reexperience, avoidance and hyper-arousal symptoms. Emergence of post-traumatic stress disorder after a traumatic event can be predicted, to a certain extent, by a glucocorticoid secretion deficiency. The role of this deficiency in stress vulnerability has been analysed by two different approaches in the rat. In a first approach, a pharmacological depletion has been induced by metyrapone, then, physiological and behavioural reactions during stress have been characterized. In a second approach, inter-individual differences of a population submitted to an intense stressor has been characterized in terms of physiological, behavioural and hippocampal gene expression responses. These studies failed to evidence an increased vulnerability induced by a glucocorticoid deficiency. On the contrary, metyrapone exhibited anxiolytic-like properties independently of its effects on glucocorticoids. Metyrapone has also modified central and peripheral metabolism in a way that could explain its neuroprotective properties. Nitric oxide (NO) and experimental african trypanosomiasis in the rat
Grâce à un modèle expérimental de la trypanosomose humaine Africaine (THA ou maladie du sommeil), le rat infecté par Trypanosoma brucei brucei, nous avons examiné l’implication du monoxyde d’azote (NO) dans le développement de cette pathologie. Des variations opposées de la concentration de ce composé ont été observées chez les animaux infectés, au niveau des compartiments périphérique et central : le NO diminue au niveau du sang et augmente au niveau cérébral. Ces changements sont dépendants de la NO-synthase inductible (iNOS). Au niveau périphérique, la diminution du NO qui survient favorise l’installation du parasite car la pression trypanocide de ce composé est diminuée. Dans cette situation, la L-arginine, le substrat à la base de la synthèse du NO, est utilisée pour la synthèse de polyamines, des composés nécessaires à la croissance du parasite. Ces mécanismes sont très probablement déclenchés par le trypanosome via ses facteurs solubles. Au niveau cérébral, la synthèse du NO est aussi soumise à des régulations qui impliquent l’arginase et la NG, NG-diméthylarginine diméthylaminohydrolase (DDAH). Tandis que l’activité de l’arginase demeure constante, celle de la DDAH augmente au cours de l’infection en accord avec les données des western-blot et des amino-acides. Cette augmentation, qui dépend essentiellement de l’isoforme DDAH-2, conduit à une augmentation du NO cérébral dont les propriétés sont trypanocides. Ces changements, contraires à ceux observés en périphérie, sont défavorables à la survie du trypanosome au niveau du cerveau. In the infected animals, opposite variations in NO concentration were observedeither at peripheral or brain compartments: NO decreases in blood but increases in brain. These changes are dependent on the activity of the inducible NO-synthase (iNOS). In periphery, the decrease observed in NO concentration favors the parasite entrance because the trypanocidal pressure exerted by NO is decreased. In such a situation, L-arginine, the substrate conducing to the synthesis of NO, is employed for the synthesis of polyamines, a category of compounds necessary for the parasite growth. It is likely that above mechanisms might be triggered by parasites. In brain, NO synthesis is submitted to additive complex regulatory processes implying arginase and NG, NG-dimethylarginine dimethylaminohydrolase (DDAH). While the arginase activity remains constant, that of DDAH increases throughout the infection process in keeping with western-blot and amino acids data. This increase, depending mainly on DDAH-2 isoform, lasts in a brain NO increase which enhance the trypanocidal pressure.
