A spectrofluorometric analysis to evaluate transcutaneous biodistribution of fluorescent nanoparticulate gel formulations
The investigation of the absorption of drug delivery systems, designed for the transport of therapeutic molecules inside the body, could be relatively simplified by the fluorophore association and tracking by means of bio-imaging techniques (i.e., optical in vivo imaging or confocal and multiphoton microscopy). However, when a fluorescence signal comes out from the skin, its specific detection can be problematic. Skin high autofluorescence can hinder the observation of administered exogenous fluorophores conjugated to drug delivery systems, making it more challenging to detect their biodistribution. In the present study, we have developed a method based on the spectrofluorometric analysis of skin samples to discriminate the fluorescent signal coming from administered fluorescent molecules from the background. Moreover, we gave a semi-quantitative evaluation of the signal intensity. Thus, we distinguished two gel formulations loading the fluorophore rhodamine B (called GEL RHO and GEL SLN-RHO). The two formulations of gels, one of which containing solid lipid nanoparticles (GEL RHO-SLN), were administered on skin explants incubated in a bioreactor, and the penetration was evaluated at different time points (2 and 6 hours). Cryostatic sections of skin samples were observed with confocal laser scanning microscopy, and a spectrofluorometric analysis was performed. Significantly higher signal intensity in the samples administered with SLN-RHO GEL, with a preferential accumulation in the hair bulbs, was found. Reaching also the deeper layers of the hair shaft after 6 hours, the solid lipid nanoparticles thickened with polymer represent a suitable drug delivery system for transcutaneous administration

Viral Encephalopathy and Retinopathy in groupers (<it>Epinephelus</it> spp.) in southern Italy: a threat for wild endangered species?
Abstract Background Betanodaviruses are the causative agents of Viral Encephalopathy and Retinopathy (VER). To date, more than 50 species have proved to be susceptible and among them, those found in genus Epinephelus are highly represented. Clinical disease outbreaks are generally characterized by typical nervous signs and significant mortalities mainly associated with aquaculture activities, although some concerns for the impact of this infection in wild fish have been raised. In this study, the authors present the first documented report describing an outbreak of VER in wild species in the Mediterranean basin. Case presentation In late summer - early winter 2011 (September-December), significant mortalities affecting wild Dusky grouper (Epinephelus marginatus), Golden grouper (Epinephelus costae) and European sea bass (Dicentrarchus labrax) were reported in the municipality of Santa Maria di Leuca (Northern Ionian Sea, Italy). The affected fish showed an abnormal swimming behavior and swollen abdomens. During this epizootic, five moribund fish showing clear neurological signs were captured and underwent laboratory investigations. Analytical results confirmed the diagnosis of VER in all the specimens. Genetic characterization classified all betanodavirus isolates as belonging to the RGNNV genotype, revealing a close genetic relationship with viral sequences obtained from diseased farmed fish reared in the same area in previous years. Conclusion The close relationship of the viral sequences between the isolates collected in wild affected fish and those isolated during clinical disease outbreaks in farmed fish in the same area in previous years suggests a persistent circulation of betanodaviruses and transmission between wild and farmed stocks. Further investigations are necessary to assess the risk of viral transmission between wild and farmed fish populations, particularly in marine protected areas where endangered species are present.</p

The defeat of the Spanish Armada in the Channel in 1588 turned ‘a weak and feeble woman’ into one of the most relevant characters of late 16th-century European history: Elizabeth I, the Tudor Queen, was considered a semi-divine creature whom the myth, by comparing her to Moon goddesses like Diana and Cinthia, had made known as Semper Eadem.
The identification of the queen with Cinthia became more and more evident after the events occurred in 1588, when she was hailed as the moon goddess par excellence in a series of texts written by her contemporaries. It was in that period, the last decades of the 16th century, that Sir Walter Ralegh, courtier, poet and seaman, wrote The Poems to Cynthia: a collection of short poems where Elizabeth I, the lady of the seas, is addressed as his beloved. I frammenti superstiti permettono di ricostruire la personalità di un filosofo peripatetico, allievo di Teofrasto, che pur avendo acquisito fama come 'primo grammatico' ed essersi occupato di critica letteraria, di poeti, di poetica e di 'storia', non lasciò da parte interessi di etica e probabilmente di fisica. Abstract
The decarbonisation of the built environment has increased reliance on Environmental Life Cycle Assessment (E-LCA) to evaluate the impacts of construction materials. However, social aspects—particularly those affecting workers—remain underexplored. This study presents a regionalised approach to support socially and environmentally informed decision-making in the Italian construction sector. For this purpose, we have integrated worker health and safety indicators into the E-LCA of two representative building products assessed across key life cycle stages. These indicators are incorporated into the evaluation of Global Warming Potential (GWP), thus serving as a decision-support tool during the design phase. From a design perspective, the aim is to promote a broader understanding of sustainability—encompassing both environmental and social dimensions—within building projects. Methodologically, the contribution is twofold. First, it addresses the current gap in context-specific data on the critical indicator of worker health and safety in the construction sector, an essential requirement for robust and scientifically recognised S-LCA studies. To this end, the study develops a regionalised scoring system based on publicly available occupational health and safety data from the Italian National Accident Database (INAIL), disaggregated by sector and region. Second, we propose a framework to combine these social indicators with LCA-based environmental impact metrics, which remain central to building-scale E-LCA. It is clear that no single region performs best, while a critical need for multi-criteria decision-making in sustainable design is evident
