    Chemical characterization, computational analysis and biological views on Daphne gnidioides Jaub. & Spach extracts: Can a new raw material be provided for biopharmaceutical applications?

    The scientific world tends to turn to natural products such as medicinal and aromatic plants because of the inadequacy of commercially available synthetic drugs as antibiotics or anticancer, and their adverse effects on healthy tissues. One of these plants is Daphne gnidioides Jaub. & Spach, which belongs to the Thymelaeaceae family, and there is no data in the literature on its biological activity. This study is aimed to elucidate the chemical profiles and in vitro anticancer, antibacterial and DNA protection and enzyme inhibitory properties of methanol extracts of root, stem, and leaf of D. gnidioides Jaub. & Spach. Polyphenolic components of the extracts were characterized by HPLC-MS/MS. The highest phenolic content was detected in the leaf extract (TIPC = 43.5 ± 0.5 mg/g DE), followed by stem (TIPC = 27.3 ± 0.7 mg/g DE) and root (TIPC = 18.3 ± 0.2 mg/g DE) extracts. Vicenin-2 and 3-O-p-coumaroyl-5-O-caffeoylquinic acid were the main identified compounds in leaf and both root and stem extracts, respectively. The extracts did not show any protective effect on DNA against experimental Fenton's reagent. The minimum inhibitory concentration and the minimum bactericidal concentration values for the root and leaf extracts against tested bacterial strains ranged from 31.25 to 500 μg/mL. After 48 h interaction of the cancer cell lines with the extracts, only the stem extract had significant cytotoxicity on HeLa cells (IC50 = 86.16 μg/mL). No remarkable activity of the extracts, which was tested against MDA-MB-231, was detected (IC50 > 1000 μg/mL). These data showed that D. gnidioides Jaub. & Spach stem extract inhibited the survival of HeLa cells in a time-dependent manner. After the treatment of IC50 concentration of stem extract with HeLa cells, an increase in LC3-II autophagic gene expression was detected. Also, the extracts exhibited significant tyrosinase inhibitory effects which were confirmed by molecular docking. To sum up, the tested extracts could be used as a starting point for the development of new multifunctional drugs

    Genome-wide Identification of PMEI Genes in Wild Olives (Olea europaea sylvestris L.) by Bioinformatic Analysis

    In the present study, 47 PMEI type 1 genes and 57 PMEI type 2 genes were identified with bioinformatic analysis. The PMEI genes were localized separately on chromosomes 1, 2, 3, 7, 8, 9, 10, 11, 12, 13, 14, 15, 16, 17, 18, 20, 21 and 22, but mainly at the level of the scaffold. The biological functions of the PMEI type 1 genes were found to be in the areas of biological regulation, metabolism and cellular functions. Their cellular localization appears to be associated with cell parts. For the PMEI type 2 genes, the biological functions were determined as biological regulation, metabolic and cellular functions. A total of 393 Arabidopsis miRNAs targeting 47 olive PMEI type 1 genes were identified. Two specific miRNAs targeting the OePMEI1-07 gene were found (ath-miR8168 and ath-miR774b-5p). For the PMEI type 2 genes, 269 Arabidopsis miRNAs were found, including 14 specific miRNAs targeting OPMEI2-02, OPMEI2-03, OPMEI2-27, OPMEI2-28, OPMEI2-29, OPMEI2-30, OPMEI2-40 and OPMEI2-54. These results suggest that PMEI genes in olives may not only play a role in cell development, germ cell formation and plant growth, but also play an important role in abiotic and biotic stress conditions in the olive

    SIYIRCIK (Daphne gnidioides) ve SIRIMBAĞI (Daphne pontica) BİTKİ EKSTRAKTLARININ BİYOLOJİK ETKİNLİĞİNİN İNCELENMESİ

    Thymelaeaceae (Serçedilgiller) familyasına ait olan Daphne pontica ve Daphne gnidioides halk arasında sırasıyla sırımbağı ve sıyırcık olarak bilinmektedirler. Ülkemizin kuzeyi ile kıyı kesimlerinde yetişmektedirler. Geleneksel tıpta bitkisel tedaviden süs bitkisine kadar birçok kullanım alanına sahiptirler. Bu çalışma ile sırımbağı ve sıyırcık bitkilerinin çeşitli kısımlarından elde edilen ekstraktların antimikrobiyal, DNA koruma, antikanser özellikleri ile kanser hücrelerini hangi programlı hücre ölümü modeline götürdüğünün belirlenmesi ve bu bitkilere ait içerik analizlerinin gerçekleştirilmesi amaçlanmıştır. Yapılan analizlerde, sıyırcık bitkisinin yaprak özütü MIC deneyi sonucunda Gram pozitif olan Alfa streptococcus haemolyticus bakterisinde 31,25µg’a kadar etki ettiği, sırımbağı bitkisinin yaprak özütünün ise çalıştığımız Gram (+) ve Gram (-) bakterilerin tümüne etki ettiği sonucuna ulaşılmıştır. Sırımbağı ve sıyırcık bitkilerinin DNA koruma aktivitelerinin ise olmadığı görülmüştür. IC50 değerinin 86,16 µg olarak bulunduğı sıyırcık bitkisinin gövde özütünün HeLa (serviks) kanser hattında en düşük değerde en yüksek etkiye sahip olduğu sonucuna varılmıştır. Çalışma ile sırımbağı ve sıyırcık bitkilerinin biyolojik etkinliği hakkında ön bilgiler elde edilmiş olup, bu bitkilerin antimikrobiyal veya antioksidant olarak kullanımı için de yeni bir alan sağlanmıştır. Böylece sentetik ve yan etkileri bulunan kanser ilaçlarının aksine, doğal ve sadece kanser hücrelerine etki eden bir ilaç olma potansiyelinin de değerlendirilmesine imkân verilmiş olacaktır

    Stout Kafur Ağacında LEA Genlerinin Biyoinformatik Analizi (Cinnamomum micranthum f. Kanehirae)

    LEA proteinleri bitkilerin abiyotik streslere karşı tepkilerinde önemli bir role sahiptir. Lauraceae ailesine ait tıbbi ve aromatik bir bitki olan Cinnamomum micranthum f. Kanehirae veya Stout Kafur ağacının genom dizisi yakın zamanda tamamlanmıştır. Stout Kafur genomunda çalışmalar olmasına rağmen LEA genleri ile alakalı herhangi bir çalışma bulunmamaktadır. Bu nedenle bu çalışmada biyoinformatik araçlar kullanılarak Stout Kafur genomunda yer alan LEA genlerinin genom çapında analizinin yapılması amaçlanmıştır. Stout Kafur genomunda 57 LEA geni (CmiLEA) tanımlandı. CmiLEA filogenetik analize göre 8 ayrı kümeye ayrılmıştır. CmiLEA'nın subselüler lokalizasyonları incelendiğinde daha çok sitoplazmada lokalize oldukları ve sadece bir miRNA hedefleyen toplam 13 gen tanımlanmıştır. CmiLEA’ da toplam 23 genin yalnızca ekzon bölgelerine sahip olduğu ve intronsuz olduğu tespit edilmiştir. Toplamda 35 korunmuş motif belirlenirken, CmiLEA-42'de yalnızca bir korunmuş motif bulunmuştur. 3B yapı sonuçlarına uygun olarak LEA_2 alt ailesinden CmiLEA-21, CmiLEA-31, CmiLEA-44, CmiLEA-45 ve CmiLEA-57 %90'ın üzerinde doğruluk göstermiştir. Bu çalışma, Cinnamomum micranthum f. kanehirae bitkisinde LEA genleri ile ilgili yapılmış ilk biyoinformatik çalışma olup, Cinnamomum cinsinde gelecekte ileri fonksiyonel analizler için bir temel oluşturabileceği düşünülmektedir

    Sosyal Bilimler EnstitüsüAile işletmeleri aile üyelerinden bir ya da daha fazla kişinin çalıştığı ve kararlarında etkili olduğu, hisselerin yarıdan fazlasına ailenin sahip olduğu ve yönetim kurulunda ailenin çoğunlukta olduğu işletmeler olarak tanımlanabilir. Aile işletmelerinde birbiriyle etkileşim halinde olan ve iç içe geçmiş; aile, işletme ve mülkiyet olmak üzere üç boyut vardır. Bunlar arasındaki dengeyi kurmak ve gözlemlemek gerek. Aile işletmeleri her ülkede ekonomilerin lokomotifi durumundadırlar. Türkiye’de de işletmelerin %90-95’i aile işletmesi özelliğini taşımaktadır. Aile işletmelerinin; 2. kuşakta çözüldüğü ve 3. kuşakta neredeyse yok olma noktasına geldiği bir vakıadır. Bu durum işletmelerin sürdürülebilirliğinde büyük bir problem olarak karşımıza çıkmaktadır. Hâlbuki aile işletmeleri istihdamın desteklenmesinde daha fazla rol üstlenirler ve bunu bir görev olarak kabul ederler. Böylece kriz ortamlarında adeta bir dalga kıran vazifesi görürler. Krizlerden daha çabuk çıkılmasında öncülük ederler. Aile işletmeleri diğer işletmelerden daha avantajlı olmalarına rağmen aile içi problemlerden dolayı kendi kendileri için bir risk alanı oluştururlar. Aile; bir yandan başarının, umudun kaynağı iken, diğer yandan da bunların yok edilmesine neden olabilecek bir potansiyel taşımaktadır. Bu nedenlerle, aile işletmelerinde, “işlerin kişilere göre değil, kurallara göre yönetilmesi” ilkesinin, yani kurumsallaşmanın hayata geçmesi gerekmektedir. Bu bağlamda işletme ve ailenin kurumsallaşması birlikte düşünülmelidir. Aile işletmeleri ile ilgili pek çok çalışma yapılmıştır. Bununla birlikte, aile işletmelerinin kurumsallaşma sürecindeki, aileden kaynaklanan ”tuzak davranışların önlenmesi” konusunda ise yeterli araştırma yapılmadığı görülmektedir. Bu çalışmamızda aile işletmelerinin uzun ömürlü olmaları için gerekli olan kurumsallaşma sürecinde aile bireylerinden kaynaklanan kurumsallaşma engelleri ve tuzak davranışlara karşı neler yapılabileceğine dair tutum ve davranışlar test edilmiştir. Önerilerin saha bulgularına göre analizleri yapılmıştır. Netice olarak; Kahramanmaraş’da aile işletmelerinde yapılan bu çalışmamızda aile üyelerinden kaynaklanan kurumsallaşma engelleri ve tuzak davranışların ortadan kaldırılması, hatta ortaya çıkmadan önlenebilmesi için xxxii önerilerde bulunulmuştur. Burada; güven, şeffaflık, adil paylaşım, hesap verebilirlik, açıklık, saygı, etkinlik, doğru iletişim, ortak akıl, uzman desteği, profesyonellerle çalışma, planlama alışkanlığı, yazılı kurallara uyma, kurum kültürünü önemseme, geleceği görebilecek bir devir planına sahip olma, liyakat, toplumsal sorumluluk gibi tercihler öne çıkmaktadır.     Assessment of Water Resources Status Using the Water Footprint Concept: The Case of Tekirdağ Province

    One of the fundamental steps in the protection and sustainability of water resources is monitoring and evaluation. By assessing the resources in terms of both quality and quantity, a clear depiction of the current situation can be established, which will form a solid inventory for the necessary actions. From the perspective of our country, the main issues concerning our water resources include the reduction in water quantity during periods of need due to excessive and uncontrolled use, the uncontrolled increase in pollution due to negligence linked to sectoral developments, and globally, the expected intense impact of climate change on the Mediterranean Basin, where we are located. The concept of the water footprint is one of the accepted methods for diagnosing the current state of water resources in terms of management planning and sustainability. The water footprint concept can effectively reveal how agricultural, industrial, and domestic uses impact water resources. In the present study, the agricultural water footprint of Tekirdağ, one of the most economically powerful provinces in the Thrace Region in terms of agriculture and industry, has been calculated and evaluated. Agriculture water footprint was found to be 1.33 billion cubic meters (BCM) in total, 0.61 BCM in crop production and 0.72 BCM in animal production. The green, blue and grey water footprint values for crop production were calculated as 0.11, 0.48 and 0.02 BCM, respectively.     The purpose of the study is to investigate and characterize the hydrogeological conditions at the Alpu Sector-A Coal Field which is located in the Eskisehir Province in Turkey. This is required to provide baseline hydrogeological information before environmental impact assessment of the planned mining operations can be done. The characterization studies included hydrogeological and hydrochemical analysis of groundwater and surface water. The spatial and temporal variations in groundwater levels are determined by measuring the groundwater levels in drilled observation and pumping wells and analyzing the pore pressure from the vibrating wire piezometers installed in different depths. The hydraulic parameters of the groundwater system are estimated by conducting aquifer tests in drilled pumping wells. The hydrochemical characteristics of the waters are based upon measured field water quality parameters and chemical analyses of the samples taken at periodic intervals from both surface and groundwater. The most prolific aquifer in the Sector-A is the alluvium aquifer which interacts with the Porsuk Stream. There is a vertical upward hydraulic gradient with excessive pore pressure beneath the coal seams.     Hay&acirc;l&acirc;t-ı Dil, published in 1868 by Hasan Tevfik, is one of the most important milestones of the transition to the novel in Turkish literature. The adventure of the Murg-ı Dil, who is in love with C&acirc;n&acirc;n, is told in the book. In the first part of it, there is a conversation in which Murg-ı Dil tells C&acirc;n&acirc;n about his suffering and complaints about the sorrow he feels. Since she gets angry at these complaints and disrespects, C&acirc;n&acirc;n leaves her lover and goes to an unknown place. Then, Murg-ı Dil begins to search his beloved. Throughout his journey, Murg-ı Dil faces with some challenges such as being captured by the savages and being a commander in a war. Thanks to the supports of P&icirc;r and N&acirc;y&acirc;b, he gets rid of these situations and finally reaches C&acirc;n&acirc;n. In the frame of the adventure of a lover who struggles in order to come together with his beloved, Hay&acirc;l&acirc;t-ı Dil aims to give the reader some advice, to criticize the political and military policies and social disorders of the epoch. Although Hay&acirc;l&acirc;t-ı Dil maintains the tradition of Folk literature and Divan literature in general, it comes closer to the novel in terms of content and characters. Despite the importance in the history of Turkish literature, no independent study has been conducted about the book yet. In the first section of this book, the place of Hay&acirc;l&acirc;t-ı Dil in Turkish literature will be emphasized and the text will be evaluated in point of its form and content. In the second section, there will be the modern Turkish translation of Hay&acirc;l&acirc;t-ı Dil and in the third section, there will be the Latinised form of it. In the last section of the book, there will be the facsimile text.</p
