    Determination of low level methyl tert-butyl ether, ethyl tert-butyl ether and methyl tert-amyl ether in human urine by HS-Spme gaschromatography/mass spectrometry

    Methyl tert-butyl ether (MTBE), ethyl tert-butyl ether (ETBE) and tert-amyl methyl ether (TAME) are oxygenated compounds added to gasoline to enhance octane rating and to improve combustion. They may be found as pollutants of living and working environments. In this work a robotized method for the quantification of low level MTBE, ETBE and TAME in human urine was developed and validated. The analytes were sampled in the headspace of urine by SPME in the presence of MTBE-d12 as internal standard. Different fibers were compared for their linearity and extraction efficiency: carboxen/polydimethylsiloxane, polydimethylsiloxane/divinylbenzene, and polydimethylsiloxane. The first, although highly efficient, was discarded due to deviation of linearity for competitive displacement, and the polydimethylsiloxane/divinylbenzene fiber was chosen instead. The analysis was performed by GC/MS operating in the electron impact mode. The method is very specific, with range of linearity 30-4600 ng L(-1), within- and between-run precision, as coefficient of variation, <22 and <16%, accuracy within 20% the theoretical level, and limit of detection of 6 ng L(-1) for all the analytes. The influence of the matrix on the quantification of these ethers was evaluated analysing the specimens of seven traffic policemen exposed to autovehicular emissions: using the calibration curve and the method of standard additions comparable levels of MTBE (68-528 ng L(-1)), ETBE (<6 ng L(-1)), and TAME (<6 ng L(-1)) were obtained

    Papillomavirus E5: the smallest oncoprotein with many functions

    Papillomaviruses (PVs) are established agents of human and animal cancers. They infect cutaneous and mucous epithelia. High Risk (HR) Human PVs (HPVs) are consistently associated with cancer of the uterine cervix, but are also involved in the etiopathogenesis of other cancer types. The early oncoproteins of PVs: E5, E6 and E7 are known to contribute to tumour progression. While the oncogenic activities of E6 and E7 are well characterised, the role of E5 is still rather nebulous. The widespread causal association of PVs with cancer makes their study worthwhile not only in humans but also in animal model systems. The Bovine PV (BPV) system has been the most useful animal model in understanding the oncogenic potential of PVs due to the pivotal role of its E5 oncoprotein in cell transformation. This review will highlight the differences between HPV-16 E5 (16E5) and E5 from other PVs, primarily from BPV. It will discuss the targeting of E5 as a possible therapeutic agent

Programa de Pós-Graduação em Educação.O estudo analisa a educação oferecida aos povos do campo, no curso de formação de educadores organizado pela Unioeste, campus de Francisco Beltrão-PR. O planejamento e a implementação do referido curso demandou muitos debates entre os professores em busca da clareza necessária para encaminhar questões polêmicas que acompanharam o processo de elaboração da proposta do curso: o currículo a ser adotado, as especificidades do campo e as peculiaridades da formação de educadores atuantes no Movimento dos Trabalhadores Rurais Sem Terra. A pesquisa desenvolvida junto a seis professores da universidade e inclui o dirigente do Movimento dos Trabalhadores Rurais Sem Terra. Dentre os professores entrevistados, os ministrantes do curso, deixaram clara a importância de formar educadores comprometidos com as lutas das mulheres e homens que vivem e trabalham no campo, garantindo essas especificidades sem perder de vista o caráter universalizante da formação docente, mas, respeitando as matrizes culturais, a formação da identidade, a manutenção dos valores, dos ideários e da terra. O estudo aponta ainda a necessidade de continuar pesquisando a temática da educação do campo, pois são muitas as dúvidas que ainda preocupam os educadores, tanto os da Unioeste que atuam nos movimentos sociais quanto os acadêmicos do Curso de Formação para Educadores do Campo, que são educadores atuantes nas escolas do campo, em busca do ensino superior

    Self-collected urine sampling to study the kinetics of urinary toluene (and o-cresol) and define the best sampling time for biomonitoring

    Purpose To study the excretion kinetics of urinary toluene, TOL-U, and o-cresol, o-C, following occupational exposure to toluene in order to define the best time for sample collection, to apply a non-invasive approach based on self-collected urine sampling. Methods Five rotogravure printing workers exposed to uncontrolled levels of toluene collected spot urine samples over three consecutive working days and the following day of rest. In each sample TOL-U and o-C were measured and kinetics of excretion evaluated. Results Toluene exposure ranged from 48.3 to 75.3 mg/ m3; TOL-U and o-C ranged from 1.4 to 34.6 lg/L and from 0.013 to 1.012 mg/L. A time course trend was obtained: TOL-U and o-C increased during the shift and peaked at the end of exposure and up to 2 h later, respectively; afterwards they rapidly decreased following apparent first order kinetics. Considering TOL-U, the elimination halflife for the first fast phase was 79 (±35 standard error) min, and for the second slow phase was 1,320 (±1,162) min. For o-C the elimination half-life for the first fast phase was 231 (±48) min. Considering a toluene uptake of 86%, TOL-U and o-C excreted in urine were about 0.0067 and 0.18% of the up taken. Conclusion Our results support the use of end shift TOLU as a short term biomarker of occupational exposure to toluene and show the feasibility of self-collected urine sampling to investigate the elimination kinetics of industrial toxics in humans

    Development of a gas chromatography/mass spectrometry method to quantify several urinary monohydroxy metabolites of polycyclic aromatic hydrocarbons in occupationally exposed subjects

    The aim of this studywas the development of a method for the determination of 12 urinary monohydroxy metabolites of PAHs, namely 1-hydroxynaphthalene, 2-hydroxynaphthalene, 2-hydroxyfluorene, 9-hydroxyfluorene, 1-hydroxyphenanthrene,2-hydroxyphenanthrene, 3-hydroxyphenanthrene, 4-hydroxyphenanthrene, 9-hydroxyphenanthrene, 1-hydroxypyrene,6-hydroxychrysene, and 3-hydroxybenzo[a]pyrene. Analytes were determined in the presence of deuterated analogues as internal standards, by GC/MS operating in the electron impact mode. Sample preparationwas performed by enzymatic hydrolysis of glucoronate and sulphate conjugates of hydroxy metabolites of PAHs, liquid–liquid extraction with n-hexane, and derivatization with a silylating reagent. The method is very specific, limits of quantification are in the range 0.1–1.4 μg/l, and range of linearity is from limit of detection to 208 μg/l.Within- and between-run precision, expressed as coefficient of variation, are <20%; accuracy for most analytes is within 20% of the theoretical value. An application of the proposed method to the analysis of 10 urine samples from coke-oven workers shows that 1-hydroxynaphthalene and 2-hydroxyfluorene were the most abundant compounds (median 61.4 and 69.7 μg/l, respectively), while 6-hydroxychrysene,and 3-hydroxybenzo[a]pyrene were always below the quantification limit

    Dissertação (mestrado) - Universidade Federal de Santa Catarina, Centro Sócio-Econômico, Programa de Pós-Graduação em Serviço Social, Florianópolis, 2014.Este trabalho é um primeiro ensaio que tem como objetivo aproximar o conceito de campo em Bourdieu para o entendimento do Serviço Social no Campo Jurídico. A proposta partiu da necessidade de se ampliar o horizonte analítico sobre o que constituiria esse espaço sócio-ocupacional. Sabe-se que na literatura profissional um importante esforço vem sendo realizado desde o início do presente século, com o claro objetivo de delimitar o chamado "campo sociojurídico". No entanto, ao questionarmos o que seria esse "campo", não encontramos na literatura profissional - salvo alguns poucos artigos - a problematização do que consistiria essa noção para a apreensão desse âmbito profissional e, tampouco, uma formulação teórica que explicaria a junção dos termos "sócio" e "jurídico". Nesse sentido, ao buscarmos novos parâmetros analíticos para o entendimento da noção de campo, encontramos em Bourdieu um forte respaldo teórico para pensarmos não só os elementos que constituem um campo qualquer, mas também para apreender as especificidades do campo jurídico, através de outras chaves heurísticas utilizadas pelo autor francês para a compreensão de um campo. Dentre as principais categorias utilizadas, além da noção de campo, estão a noção de habitus, tipos de capital, inclusive o capital simbólico, a illusio e a violência simbólica. Dessa forma, na primeira seção do trabalho, partiu-se de uma revisão bibliográfica acerca dos principais termos que auxiliam na compreensão de campo em Bourdieu e os elementos que constituiriam o campo jurídico. Na segunda seção, situamos um breve histórico da inserção do Serviço Social no Campo Jurídico e como a profissão se constituiu no interior desse campo. Foi realizada uma pesquisa bibliográfica em dois dos principais eventos da categoria profissional (ENPESS e CBAS), relativo a um mesmo período histórico, a fim de identificar não somente a definição do chamado "campo sociojurídico" e suas variantes, mas também, levantar indicadores a partir da representação da prática profissional nos trabalhos pesquisados, das estratégias e técnicas de intervenção, daquilo que apareceu enquanto atribuição e atuação profissional, para pensarmos a dinâmica do trabalho do assistente social no interior do campo jurídico. Por fim, as análises empreendidas em torno do conceito de campo e, consequentemente do campo jurídico, aliadas aos resultados da pesquisa, mostraram que o Serviço Social no Campo Jurídico está em permanente disputa pelo direito de dizer o direito.Abstract : This work aims to bring the concept of field in Bourdieu to a better understanding of Social Work in the Legal Field. The proposal came from the need to expand the analytical horizon of what would constitute this socio-occupational space. It is known that an important effort has been made in the professional literature since the beginning of this century, with the clear objective of defining the so-called "socio-legal field." However, when we question on what this "field" would be and the problematic of what this notion would be consisted of on the professional context, and even a theoretical formulation that would explain the join of "social" and "legal", we find no professional literature, except for a few articles. In this sense, as we seek new analytical parameters to understand the concept of field, we find in Bourdieu, a strong theoretical support to think about not only the elements that constitute any field , but also to grasp the specifics of the legal field, through other key heuristics used by the French author to understand a field. Among the main categories used, besides the notion of field, there are: notion of habitus, types of capital, including the symbolic capital, the illusio and symbolic violence. Thus, the first section of this work begins with a literature review on main terms that assist in understanding of field in Bourdieu and the elements that would constitute the legal field. In the second section, we place a brief history of the insertion of Social Work in the Legal Field and how the profession is constituted within that field. A literature survey in two major events in the professional category (National Meeting of Researchers in Social Work (ENPESS) and the Brazilian Congress of Social Workers (CBAS) concerning the same historical period was conducted. This was done in order to identify the definition of the "social legal field" and its variants. Also, it has raised the indicators from professional practice on the work surveyed, strategies and intervention techniques, in what appeared as allocation and professional experience, to understand the dynamics of the work of social workers within the legal field. Finally, the analyses undertaken around the 18 concept of field and, consequently the legal field, allied to research results showed that the Social Work in the Legal Field is in constant dispute over the right to say what is right
