Using rapid city surveys to inform municipal social policy : an application in Cali, Colombia
No full textMany developing countries assign local governments increasing responsibilities in fighting poverty. This requires local social policy to go beyond the execution of centrally designed and funded education and health programs. Hence, local governments and their partners have both an opportunity and a need to analyze key local bottlenecks for poverty reduction and social development. Drawing on an example from Cali, Colombia, The author describes a tool for such policy formulation at the local level-a rapid city household survey. Although the survey uses pre-coded and closed- ended questions, it is contextual in the sense that it is tailor-made to social and economic conditions in Cali. The survey places particular emphasis on collecting key quantitative information, such as household welfare and service access, as well as qualitative information, such as service evaluations and population priorities. Combining the quantitative and qualitative data allows, for example, the mapping of population budget priorities or service satisfaction levels by welfare group. No full textEn los marcos del proyecto global de investigación que involucra cinco ciudades, nos proponemos en este capítulo presentar el caso de la violencia urbana en Cali, entre 1980 y 2012. Para desarrollarlo, hemos vinculado antecedentes históricos de la violencia en la ciudad, destacando sus contextos socioeconómicos, con tres temas cruciales que son objetivos de la investigación: el de los escenarios más típicos de la violencia urbana, el de las memorias que tienen los ciudadanos de estos y el de posibles políticas públicas de seguridad ciudadana. No full textEsta investigación tiene como propósito analizar la siniestralidad en accidentes de tránsito en bicicleta en Santiago de Cali, la cual se ha planteado como objetivos específicos el estudiar la legislación aplicable a los accidentes causados por bicicletas e identificar las principales causas de siniestralidad relacionada con biciusuarios en la ciudad. Para ello la investigación parte de considerar el aumento de la accidentalidad por situaciones en las que se ven involucradas las bicicletas, además de tener en cuenta el aumento de los viajes en este vehículo de parte de los habitantes de Cali, para así, aplicar una metodología que combine, desde un enfoque cualitativo, que permita comprender los hechos importantes existentes tras establecer la correlación entre lo que indica la legislación, en especial la Ley 769 de 2002, basada esta, a su vez, en el artículo 24 de la Constitución Política.The purpose of this research is to analyze the accident rate in traffic accidents involving bicycles in Santiago de Cali, which has set as specific objectives to study the legislation applicable to accidents caused by bicycles and to identify the main causes of accidents related to bicycle users in the city. For this purpose, the research starts by considering the increase in accidents caused by situations involving bicycles, in addition to taking into account the increase in trips in this vehicle by the inhabitants of Cali, in order to apply a methodology that combines, from a qualitative approach, to understand the important existing facts after establishing the correlation between what the legislation indicates, especially Law 769 of 2002, based, in turn, on Article 24 of the Political Constitution.Introducción. --

Quantification of organelle contact sites by split-GFP-based contact site sensors (SPLICS) in living cells
No full textMembrane contact sites between organelles are essential for maintaining cellular homeostasis, which requires the continuous exchange of signaling molecules, ions, nutrients and lipids. Alterations of different contact sites are associated with a wide spectrum of human diseases. However, visualizing and quantifying these contact sites remains a challenge. This protocol describes the use of split-GFP-based contact site sensors (SPLICS) in microscopy applications for mapping organelle contact sites both in fixed and living cells. SPLICS sensors are engineered to express equimolar amounts of the organelle-targeted nonfluorescent β11 and GFP1-10 portions of the split-GFP protein in a single vector, and are capable of reconstituting fluorescence when two opposing membranes come into proximity. Reconstitution will occur only over the cell volume at defined contact sites resulting in a bright signal that can be detected easily and quantified automatically with specific custom-made plugins. The use of minimal targeting sequences facilitates targeting specificity and membrane coverage, avoiding artifacts due to full-length fusion protein overexpression and, thus, possible perturbations of the cell’s physiology. SPLICS sensors engineered to simultaneously detect multiple contact sites within the same cell have been generated by exploiting the ability of the β11 GFP fragment to reconstitute different color-shifted variants of the GFP1-10 fragment. Here, we describe a detailed protocol to set up SPLICS expression in living cells (2–3 d), detection and acquisition (1 d), and automated quantification with custom plugins (1–2 d).
