High-throughput in vitro processing of human primary microRNA by the recombinant microprocessor
© 2021 The Author(s)We describe a protocol to conduct a high-throughput in vitro processing assay, using 1,881 human primary microRNAs (pri-miRNAs) and recombinant Microprocessor complex, followed by deep sequencing library generation. This comprehensive approach allows the mapping of cleavage sites and the measurement of processing efficiency of a large number of substrates simultaneously. Our protocol is readily modifiable to investigate the effects of chemicals and regulatory proteins. Mass production of ultra-pure NaI powder for COSINE-200
COSINE-200 is the next phase experiment of the ongoing COSINE-100 that aims to unambiguously verify the annual modulation signals observed by the DAMA experiment and to reach the world competitive sensitivity on the low-mass dark matter search. To achieve the physics goal of the COSINE-200, the successful production of the low-background NaI(Tl) detectors is crucial and it must begin from the mass production of the ultra-low background NaI powder. A clean facility for mass-producing the pure-NaI powder has been constructed at the Center for Underground Physics (CUP) in Korea. Two years of operation determined efficient parameters of the mass purification and provided a total of 480 kg of the ultra-pure NaI powder in hand. The potassium concentration in the produced powders varied from 5.4 to 11 ppb, and the maximum production capacity of 35 kg per 2 weeks was achieved. Here, we report our operational practice with the mass purification of the NaI powder, which includes raw powder purification, recycling of the mother solution, and recovery of NaI from the residual melt after crystal growth.11Nsciescopu
Controlled Assembly of Icosahedral Colloidal Clusters for Structural Coloration
Icosahedral colloidal clusters are a new class of spherical colloidal crystals. This cluster allows for potentially superior optical properties in comparison to conventional onion-like colloidal supraballs because of the quasi-crystal structure. However, the characterization of the cluster as an optical material has until now not been achieved. Here we successfully produce icosahedral clusters by assembling silica particles using bulk water-in-oil emulsion droplets and systematically characterize their optical properties. We exploit a water-saturated oil phase to control droplet drying, thereby preparing clusters at room temperature. In comparison to conventional onion-like supraballs with a similar size, the icosahedral clusters exhibit relatively strong structural colors with weak nonresonant scattering. Simulations prove that the crystalline array inside the icosahedral cluster strengthens the collective specular diffraction. To further improve color saturation, the silica particles constituting the cluster are coated with a thin-film carbon shell. The carbon shell acts as a broad-band absorber and reduces incoherent scattering with long optical paths, resulting in vibrant blue, green, and red colors comparable to inorganic chemical pigments. © 2020 American Chemical Society
