    Die Abschlussarbeit befasst sich in ihrem theoretischen Teil mit der Gesamtkonzeption und den Zielen des deutschen Bildungswerkes BLITZ e.V. Dieses wird insbesondere durch die Beschreibung seiner Entstehung, Entwicklung und der wichtigsten Projekten vorgestellt. Weiterhin soll auf den Förderverein BLITZ e.V., und dessen unterstützende Hilfe eingegangen werden. Das darauffolgende Kapitel soll das europäische Projekt „Freiwilliger Dienst“ beschreiben. Der zweite Teil dieser Arbeit behandelt den Praxiseinsatz des Autors beim Bildungswerk BLITZ e.V. im Zuge der Veranstaltung -„Kinderdorf 2009“.Bakalářská práce se ve své teoretické části zabývá celkovou koncepcí a cíli německého "Vzdělávacího střediska BLITZ e.V.". To je představeno především popisem jeho vzniku, vývoje a jednotlivých projektů. Zmíněn je také Förderverein BLITZ e.V. a jedna z kapitol je taktéž věnována evropskému projektu "Evropská dobrovolná služba". Druhá část pojednává o autorově praxi ve vzdělávacím středisku BLITZ e.V., v dětské vesničce "Kinderdorf 2009". Ta je v závěrečné práci důkladně popsána a doplněna o vlastní zhodnocení.In its theoretical part, the Bachelor thesis deals with the overall concept and aims of the German "Educational centre BLITZ e.V.". It is introduced particularly through a chronological description of its formation, development and individual projects. Also Förderverein BLITZ e.V. is mentioned and one chapter is also dedicated to the "European voluntary service". The second part deals with author´s training in BLITZ e.V., in the children´s village, "Kinderdorf 2009".     Bungalow blitz: another history of Irish architecture

    This curating and research project is directed towards an understanding of the complex visual cultural processes that underpin and arise from changes in contemporary vernacular domestic building in Ireland, especially with regard to that specially rural and suburban form; the bungalow. Inasmuch, this represents a single, layered visual culture research outcome. The project uses the exhibition as a prospective research method. Each exhibition includes only work generated over the course of the project by researchers working on the Bungalow Blitz research team, and, as such, serves as an opportunity for critical reflection and analysis. This research is undertaken as part of a wider collaborative research project resourced by a 3 year Social Science and Humanities Council initiative on practice-led inquiry in architecture, the visual arts and curating. All exhibitions include research by Paul Antick, Andrew Kearney, Aoife Mac Namara. This single, braided research output includes A) My book ‘Bungalow Blitz' (Calgary: The Banff Centre WPG Editions, 2006), ISBN: 1-894773-20-9; which contains five piecesI have written contextualising the project.     1. General Overview MTAP is a key enzyme in the methionine salvage pathway and controls the levels of methylthioadenosine (MTA). Lost of MTAP, which is frequent in melanoma, results in higher intra and extracellular MTA levels. As MTA is an inhibitor of methylation reactions within the cell, we decided to further investigate the role of methylation in the progression of cancer and how the main signaling pathway involved in melanoma (RAS-RAF-MEK-ERK pathway) can be affected in the presence or absence of methylation inhibitors. 2. Results • Exogenous addition of MTA (and therefore blocking of methylation reactions) to melanoma cells increased total activity of the RAS-ERK pathway, highlighting a new role of methylation into controlling the amplitude of the signal transduction. • We found out that PRMT5, a protein arginine methyl transferase enzyme, binds to CRAF, a key component of the RAS-ERK pathway. In vitro and in vivo experiments showed that PRMT5 methylates CRAF and thus modifies the pathway’s output. We identified CRAF arginine 563 as the target of PRMT5 methyl transferase activity. Furthermore, in vitro kinase assays and siRNA technology experiments showed that methylation at arginine 563 can modify the kinase activity of CRAF protein, therefore modulating the total activity of the RAS-ERK pathway. • The previous results provided the first known evidence of methylation as a key modulator of the activity in MAPKs pathways, so we looked for a biological significance of this new regulatory mechanism. We used the well-established PC12 cell line system (EGF growth factor triggers proliferation whereas NGF promotes differentiation): as we had shown that PRMT5-catalyzed methylation can modify the kinase activity of the RAS-ERK pathway, we wondered whether this regulation could be part of a general mechanism controlling cell behavior. In order to test this, we tried to mimic the differentiating effect of NGF using EGF to activate the pathway in the presence of MTA or after PRMT5 siRNA. Interestingly, when triggering the cells with EGF after PRMT5 depletion or MTA treatment, they differentiated in a manner indistinguishable from that induced by NGF. Moreover, we observed that cells transfected with CRAFR563K mutant underwent differentiation, instead proliferation, when treating the cells with EGF. Together these data demonstrate a central role of methylation in the growth factors-elicited biological responses. • RAS-ERK pathway plays a key role into controlling development and maintenance of melanoma. There is a clear correlation between the activity of the pathway and melanoma growth and proliferation, so we wondered how methylation reactions could affect melanoma progression. In order to address this question we performed in vitro assays in a variety of melanoma cell lines and we found out that MTA inhibited cell proliferation and viability. Next, we injected these cells in a melanoma xenograft mouse model to induce tumoral growth: mice treated with MTA showed a significant decrease (47%) in the tumor volume with no apparent toxic effects, indicating that MTA is effective blocking melanoma in vivo tumor growth. Besides, we have been able to proof that MTA inhibitory effect was due to citostatic MTA capability rather than a pro-apoptotic effect. 3. Discussion In our reports we demonstrate that methylation is an important mechanism modulating signal transduction through the RAS-ERK pathway in response to specific growth factors. Our data implicate a specific PRMT5 methylation motif (GRG) that is conserved in all RAF proteins and we demonstrate that methylation appears to be a critical posttranslational modification controlling the RAF input to the downstream activity. We have shown that methylation is controlling ERK signal amplitude in response to specific growth factors in cells from different species. We also have identified PRMT5 as the protein performing the methylation reaction. Our results provide clues about the possible fine-tuning mechanism by which methylation regulates the total signal, indicating that PRMT5 methylates CRAF at arginine 563. Our research shows that protein arginine methylation is a universal mechanism that modulates signal transduction in response to specific growth factors and we demonstrate that PRMT5 is the methyltransferase involved in the process regulating the total CRAF kinase activity within the pathway. The mechanism we describe provides the means by which cells can modulate the time profile of ERK activation for which a particular biological response is evoked. More importantly, because cancer cells tend to heavily rely on oncogenic signalling through the RAS-RAF-MEK-ERK pathway, this additional level of signal transduction regulation identifies novel candidate targets for therapeutical intervention. Finally, we have tested the potential of the methylation inhibitor MTA regarding to tumour progression: our experiments have demonstrated that MTA inhibits, in vitro, cell proliferation and viability in a dose dependent manner in a variety of mouse and human melanoma cell lines. Importantly, MTA was also effective inhibiting in vivo tumour growth in a mouse melanoma xenograft model. In summary, here we show the therapeutic potential of the natural occurring nucleoside MTA and we demonstrate that MTA can inhibit melanoma cell proliferation and in vivo tumour growth at non-toxic rates, supporting the use of MTA in antitumoral therapies and presenting protein methylation reactions as potential targets when developing new antitumoral strategies

    Wudjari Country Bush Blitz: Inland Aquatic Molluscs (Final Report)

    Sixty-three lots of inland aquatic molluscs were collected from 2 classes (Gastropoda and Bivalvia) comprised of seven families and 11 identified species, one of which may be new to science. Of the 19 sites surveyed in ponds, wetlands, lakes, rivers, and streams on Wudjari County, including regions around Kepa Kurl (Esperance), Madoowernup (Cape Le Grand National Park) and Gauroojeninya (Cape Arid National Park)), nine supported aquatic molluscs. An important aspect of this survey was to confirm whether the threatened freshwater mussel, Westralunio carteri Iredale, 1934 (listed as vulnerable on the Western Australian Biodiversity Conservation Act 2016 (BC Act) and the Commonwealth of Australia Environment Protection and Biodiversity Conservation Act 1999 (EPBC Act)). One Western Australian Museum record indicated that the species was found from catchments draining to Esperance Town Beach in the 1970s. However, surveys of the Bandy Creek catchment failed to locate the species. Two exotic/pest species of freshwater snail were confirmed to be inhabiting modified wetland sites within and near the town of Esperance. The study also resulted in seven new records/range extensions for six species of inland aquatic mollusc. Additionally, the collection of a potentially undescribed species of what appears to be Coxiella from standing water on a sheet of granite near the base of Boyatup Hill is an unexpected outcome given this genus of aquatic snails typically occurs in inland salt lakes and seldom found in freshwater habitats. The revelation of at least one putatively undescribed species of mollusc during the survey achieved the “species discovery” objective of the Bush Blitz program and follow-up work through the WA Museum will aim to formally describe the species in consultation with project partners.Full Tex

    BLITZ-HF: a nationwide initiative to evaluate and improve adherence to acute and chronic heart failure guidelines

    Aims: To assess adherence to guideline recommendations among a large network of Italian cardiology sites in the management of acute and chronic heart failure (HF) and to evaluate if an ad-hoc educational intervention can improve their performance on several pharmacological and non-pharmacological indicators. Methods and results: BLITZ-HF was a cross-sectional study based on a web-based recording system with pop-up reminders on guideline recommendations used during two 3-month enrolment periods carried out 3 months apart (Phase 1 and 3), interspersed by face-to-face macro-regional benchmark analyses and educational meetings (Phase 2). Overall, 7218 patients with acute and chronic HF were enrolled at 106 cardiology sites. During the enrolment phases, 3920 and 3298 patients were included, respectively, 84% with chronic HF and 16% with acute HF in Phase 1, and 74% with chronic HF and 26% with acute HF in Phase 3. At baseline, adherence to guideline recommendations was already overall high for most indicators. Among acute HF patients, an improvement was obtained in three out of eight indicators, with a significant rise in echocardiographic evaluation. Among chronic HF patients with HF and preserved or mid-range ejection fraction, performance increased in two out of three indicators: creatinine and echocardiographic evaluations. An overall performance improvement was observed in six out of nine indicators in ambulatory HF with reduced ejection fraction patients with a significant increase in angiotensin receptor-neprilysin inhibitor prescription rates. Conclusions: Within a context of an already elevated level of adherence to HF guideline recommendations, a structured multifaceted educational intervention could be useful to improve performance on specific indicators. Extending this approach to other non-cardiology healthcare professionals, who usually manage patients with HF, should be considered
