    Plasmodium berghei High-Throughput (PbHiT): a CRISPR-Cas9 System to Study Genes at Scale.

    Genetic modification is essential for understanding parasite biology, yet it remains challenging in Plasmodium. This is partially due to the parasite's low genetic tractability and reliance on homologous recombination, since the parasites lack the canonical non-homologous end-joining pathway. Existing approaches, such as the PlasmoGEM project, enable genome-wide knockouts but remain limited in coverage and flexibility. Here, we present the Plasmodium berghei high-throughput (PbHiT) system, a scalable CRISPR-Cas9 protocol for efficient genome editing in rodent malaria parasites. The PbHiT method uses a single cloning step to generate vectors in which a guide RNA (gRNA) is physically linked to short (100 bp) homology arms, enabling precise integration at the target locus upon transfection. The gRNA also serves as a unique barcode, allowing pooled vector transfections and identification of mutants by downstream gRNA sequencing. The PbHiT system reliably recapitulates known mutant growth phenotypes and supports both knockout and tagging strategies. This protocol provides a reproducible and scalable tool for genome editing in P. berghei, enabling both targeted functional studies and high-throughput genetic screens. Additionally, we provide an online resource covering the entire P. berghei protein-coding genome and describe a step-by-step pooled ligation approach for large-scale vector production.     Tese (doutorado) - Universidade Federal de Santa Catarina, Centro de Comunicação e Expressão.Esta tese propõe uma leitura revisionista da poesia contemporânea através do exame do caso de um dos mais esquecidos escritores norte-americanos do século XX: Laura (Riding) Jackson (1901-1991). O objetivo é demonstrar que Riding não apenas possuía uma poética definida e singular, mas que ela permanece uma das instâncias mais extremas e paradoxais do modernismo anglo-americano, a ponto de Riding abandonar a escrita da poesia em 1938. Recorrendo a conceitos de "formação do cânone" bem como às noções de "discurso" e "função do autor", em Foucault, investigo a construção do cânone da poesia moderna anglo-americana, recuperando o contexto e as circunstâncias da ocultação de Riding. Enquanto cubro os "discursos" poéticos em circulação na primeira metade do século XX-o "imagismo" de Pound, a "dissociação da sensibilidade", "impersonalidade" e "tradição" de Eliot, a "unidade orgância" e "ambigüidade" da Nova Crítica-ofereço um panorama crítico de modernismos alternativos sendo articulados à época. Minha intenção é demonstrar que os poemas de Riding são expressões vigorosas de um escritor para quem "a mente pensando se torna a força ativa do poema", para usar a apta formulação de Charles Bernstein. Entre minhas descobertas sobre as várias e complexas razões que levaram à não-canonização de Riding estão a hegemonia da Nova Crítica, o exílio voluntário de Riding da cena literária (onde são feitas ou desfeitas as reputações), sua recusa em ser antologiada, bem como em ser explicada em termos críticos que não os dela. Todos esses fatores, mais a "dificuldade" de sua poesia, contribuíram para fazer de Riding "a maior poeta esquecida da poesia norte-americana", como escreveu Kenneth Rexroth. Ajudado pelos insights de dois importantes críticos de poesia norte-americana, Charles Bernstein e Marjorie Perloff, defendo que a "poesia da mente" de Riding-onde o que está em jogo é que o que pensamos ser a nossa realidade-representa uma mudança radical no paradigma da poética modernista: de uma poesia centrada na imagem para uma poesia centrada na linguagem. Focalizando a experiência consciente e o tempo duracional do pensamento presente em seus poemas, concluo que as "pensagens" de Riding têm o objetivo preciso de constatar um fato universal: enquanto seres humanos e pensantes, estamos numa condição permanente chamada linguagem

    CRISPR/Cas9 and genetic screens in malaria parasites: small genomes, big impact.

    The ∼30 Mb genomes of the Plasmodium parasites that cause malaria each encode ∼5000 genes, but the functions of the majority remain unknown. This is due to a paucity of functional annotation from sequence homology, which is compounded by low genetic tractability compared with many model organisms. In recent years technical breakthroughs have made forward and reverse genome-scale screens in Plasmodium possible. Furthermore, the adaptation of Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR) and CRISPR-Associated protein 9 (CRISPR/Cas9) technology has dramatically improved gene editing efficiency at the single gene level. Here, we review the arrival of genetic screens in malaria parasites to analyse parasite gene function at a genome-scale and their impact on understanding parasite biology. CRISPR/Cas9 screens, which have revolutionised human and model organism research, have not yet been implemented in malaria parasites due to the need for more complex CRISPR/Cas9 gene targeting vector libraries. We therefore introduce the reader to CRISPR-based screens in the related apicomplexan Toxoplasma gondii and discuss how these approaches could be adapted to develop CRISPR/Cas9 based genome-scale genetic screens in malaria parasites. Moreover, since more than half of Plasmodium genes are required for normal asexual blood-stage reproduction, and cannot be targeted using knockout methods, we discuss how CRISPR/Cas9 could be used to scale up conditional gene knockdown approaches to systematically assign function to essential genes

    W. H. Auden's inter-war poetry: a political use of ambiguity

Programa de Pós-Graduação em Letras/Inglês e Literatura CorrespondenteThis dissertation focuses the historical context of the inter-war period in England and Wystan Hugh Auden#s poetry. Auden#s poetic work has been read, interpreted, analysed and criticised considering his biography, a form of reading that subtly differs from autobiography. I think that his texts are autobiographical pieces that denote the 1920s and 1930s social, cultural, economic and political changes through which England went. For this I analyse autobiographical traces in his poetry and his specific use of ambiguity that constitutes in my point of view a political strategy that shelters the author before the social censorship against homosexuals in that period of the English History. The poet disguises his subjectivity in masks or third person as a protagonist forced to behave performatively in order to survive in his society. The poems show individuals living in a conservative society and their impossibility to live a love relationship in its completion. Protected by ambiguity, the poet is able to keep a place in society free from the cruelties engendered against homosexuals who were considered subversive individuals in that epoch. Auden#s political use of ambiguity is thus a strategy to hide his homosexuality, what elicits his concern with gender matters. Moreover, the poems show the poet#s awareness towards social class. By bringing up gender and class, Auden#s inter-war poetics can contribute to gay, lesbian and queer studies as a form to show socio-cultural views of homosexuality and homosexuals# quotidian lives. Esta tese focaliza o contexto histórico do período entre-guerras na Inglaterra e a poesia de Wystan Hugh Auden. A obra poética de Auden tem sido lida, interpretada, analisada e criticada considerando-se sua biografia, uma forma de leitura que sutilmente difere de autobiografia. Eu penso que seus textos são peças autobiográficas que denotam as mudanças sociais, culturais, econômicas e políticas pelas quais a Inglaterra passou. Para isto, eu analiso os traços autobiográficos em sua poesia e seu uso específico da ambiguidade que se constitui em meu ponto de vista em uma estratégia política que protege o autor diante da censura social contra os homossexuais naquele período da História da Inglaterra. O poeta esconde sua subjetividade em máscaras ou em terceira pessoa como um protagonista forçado a comportar-se performaticamente a fim de sobreviver em sua sociedade. Os poemas mostram indivíduos vivendo em uma sociedade conservadora e sua [dos indivíduos] impossibilidade de viver relacionamentos de amor em sua completude. Protegido pela ambigüidade, o poeta é capaz de manter um lugar na sociedade, livre das crueldades engendradas contra os homossexuais que eram considerados indivíduos subversivos naquela época. O uso político da ambiguidade por Auden é assim uma estratégia para esconder sua homossexualidade, o que demonstra sua preocupação com questões de gênero. Além disso, os poemas mostram a consciência do poeta para com classes sociais. Ao abordar gênero e classe, a poética entre-guerras de Auden pode contribuir para os estudos gays, lésbicos e queer como forma de mostrar as visões sócio-culturais da homossexualidade e das vidas quotidianas dos homossexuais

    Traditional medicine in sub-Saharan Africa : its importance and potential policy options

    This report indicates that about 20 percent of Africans who seek medical care first consult traditional healers. Patients tend to consult modern health care services for infectious or acute diseases, or those for which modern health care has been shown to be highly effective. But patients tend to consult traditional practitioners for chronic diseases, for diseases related to psychological or social disruption or to reproductive systems, for diseases that are slow to respond to treatment or deemed to be"magical"in origin. The prestige and credibility of traditional healers have been waning in the face of modernization and an increasingly educated public. Even so many highly educated people consult traditional practitioners. A survey in Ibadan of two groups - one educated elite, the other a traditional, less privileged group - found that roughly 70percent of both groups used traditional health care, particularly traditional drugs. The author shows that traditional medicine is an important source of health care for significant number of Africans and that traditional healers, particularly those who wield authority within their communities, are an important human resource for health care.Gender and Health,Health Monitoring&Evaluation,Health Systems Development&Reform,Regional Rural Development,Hygiene Promotion and Social Marketing
