    Plasma drug concentrations and virologic evaluations after stopping treatment with nonnucleoside reverse-transcriptase inhibitors in HIV type 1-infected children

    BACKGROUND: The optimum strategy for stopping treatment with drugs that have different half-lives in a combination regimen to minimize the risk of selecting drug-resistant viruses remains unknown. We evaluated drug concentrations in plasma, human immunodeficiency virus (HIV) load, and development of drug resistance after a planned treatment interruption of a nonnucleoside reverse-transcriptase inhibitor (NNRTI)-containing regimen in HIV type 1-infected children. METHODS: Children with viral loads or =30% (for children aged 2-6 years) or CD4 cell percentages > or =25% and CD4 cell counts > or =500 cells/microL (for children aged 7-15 years) were randomized to either a planned treatment interruption or to continuous therapy. In the planned treatment interruption arm, either (1) treatment with nevirapine or efavirenz was stopped, and treatment with the remaining drugs was continued for 7-14 days, or (2) nevirapine or efavirenz were replaced by a protease inhibitor, and all drugs were stopped after 7-14 days. Sampling for determination of plasma drug concentrations, measurement of viral load, and drug resistance testing was scheduled at day 0, day 7 (drug concentrations only), day 14, and day 28 after interruption of treatment with an NNRTI. RESULTS: Treatment with an NNRTI was interrupted for 35 children (20 were receiving nevirapine, and 15 were receiving efavirenz). Median time from NNRTI cessation to stopping all drugs was 9 days (range, 6-15 days) for nevirapine and 14 days (range, 6-18 days) for efavirenz. At 7 days, 1 (5%) of 19 and 4 (50%) of 8 children had detectable nevirapine and efavirenz concentrations, respectively; efavirenz remained detectable in 3 (25%) of 12 children at 14 days. At 14 days, viral load was > or =50 copies/mL in 6 of 16 children interrupting treatment with nevirapine (range, 52-7000 copies/mL) and in 2 of 12 children interrupting treatment with efavirenz (range, 120-1600 copies/mL). No new NNRTI mutations were observed. CONCLUSIONS: In children with virological suppression who experienced interruption of treatment with an NNRTI, staggered or replacement stopping strategies for a median of 9 days for nevirapine and 14 days for efavirenz were not associated with the selection of NNRTI resistance mutations

    Degeneracies between self-interacting dark matter and supernova feedback as cusp-core transformation mechanisms

    Abstract We present a suite of 16 high-resolution hydrodynamic simulations of an isolated dwarf galaxy (gaseous and stellar disk plus a stellar bulge) within an initially cuspy dark matter (DM) halo, including self-interactions between the DM particles (SIDM); as well as stochastic star formation and subsequent supernova feedback (SNF), implemented using the stellar feedback model SMUGGLE. The simulations start from identical initial conditions and we regulate the strength of SIDM and SNF by systematically varying the SIDM momentum transfer cross-section and the gas density threshold for star formation. The DM halo forms a constant density core of similar size and shape for several combinations of those two parameters. Haloes with cores that are formed due to SIDM (adiabatic cusp-core transformation) have velocity dispersion profiles which are closer to isothermal than those of haloes with cores that are formed due to SNF in simulations with bursty star formation (impulsive cusp-core transformation). Impulsive SNF can generate positive stellar age gradients and increase random motion in the gas at the centre of the galaxy. Simulated galaxies in haloes with cores that were formed adiabatically are spatially more extended, with stellar metallicity gradients that are shallower (at late times) than those of galaxies in other simulations. Such observable properties of the gas and the stars, which indicate either an adiabatic or an impulsive evolution of the gravitational potential, may be used to determine whether observed cores in DM haloes are formed through self-interactions between the DM particles or in response to impulsive SNF

    Sneutrino DM in the NMSSM with inverse seesaw mechanism

    In supersymmetric theories like the Next-to-Minimal Supersymmetric Standard Model (NMSSM), the lightest neutralino with bino or singlino as its dominant component is customarily taken as dark matter (DM) candidate. Since light Higgsinos favored by naturalness can strength the couplings of the DM and thus enhance the DM-nucleon scattering rate, the tension between naturalness and DM direct detection results becomes more and more acute with the improved experimental sensitivity. In this work, we extend the NMSSM by inverse seesaw mechanism to generate neutrino mass, and show that in certain parameter space the lightest sneutrino may act as a viable DM candidate, i.e. it can annihilate by multi-channels to get correct relic density and meanwhile satisfy all experimental constraints. The most striking feature of the extension is that the DM-nucleon scattering rate can be naturally below its current experimental bounds regardless of the higgsino mass, and hence it alleviates the tension between naturalness and DM experiments. Other interesting features include that the Higgs phenomenology becomes much richer than that of the original NMSSM due to the relaxed constraints from DM physics and also due to the presence of extra neutrinos, and that the signatures of sparticles at colliders are quite different from those with neutralino as DM candidate.National Natural Science Foundation of China (NNSFC) [11575053]SCI(E)ARTICLE1

    Suitability of the TBA method for assessing lipid oxidation in a meat system with added phenolic-rich materials

    Numerous protocols and modifications of the thiobarbituric acid (TBA) test are available in the literature. The present paper compares the effectiveness of different TBA tests in minimizing the interferences caused by the addition of phenolic-rich materials (wild fruits) as antioxidants in cooked burger patties. The aqueous acid extraction procedure (EM) and a modified distilation TBA method (DM) were tested with different conditions of incubation - boiling (B) vs. room temperature (RT) - for monitoring lipid oxidation in cooked burger patties during refrigerated storage. DM-B and DM-RT were more suitable than EM procedures for assessing TBA-reactive substances (TBA-RS) in meat samples containing compounds such as anthocyanins, with similar spectral properties than that of the TBA-malondialdehyde (MDA) adduct. Additionally, interferences caused by browning development during incubation were avoided by DM procedures or by performing RT incubations. Correlations between TBA-RS numbers and hexanal contents in cooked pork burger patties were calculated in order to corroborate the suitability of the tested TBA procedures. The DM-RT procedure showed the highest correlation with hexanal content (R2 = 0.90; p < 0.001). © 2010 Elsevier Ltd.     Policy-driven Data Sharing over Attribute-Based Encryption supporting Dual Membership

    Attribute-Based Encryption (ABE) plays an important role in current secure data sharing through fine-grained customizable policies. However, the existing ABE schemes only support simple predicates, = and ≠, but cannot express a more general membership predicates, ∈ and ∉, in policies. The low expressivity of ABE will enlarge the ciphertext storage and reduce the communication efficiency. To overcome this problem, we propose an ABE supporting Dual Membership (DM-ABE). The core problem for implementing this scheme is how to use cryptographic methods to decide the membership between the verified element and the given set. In order to solve this problem, we design a cryptographic algorithm, called Secure Decision of Membership (SDM), based on aggregation functions. In this algorithm, any set can be aggregated into one cryptographic element, and the verified element and the given set can be converted into another cryptographic element in decision process. The membership between them can be decided by the above two cryptographic elements. Furthermore, we construct the DM-ABE by using SDM. Because of the good expressivity of our DM-ABE, we further propose a novel cryptographic data sharing framework by integrating DM-ABE and attribute-based access control to provide fine-grained access control and security protection for private data. In the security proof of DM-ABE, we prove that the DM-ABE satisfies the semantic security against chosen-plaintext attacks under the DBDHE assumption in the standard model through a unified way, considering both two encryption methods for ∈ and ∉ at the same time. Finally, we analyze our scheme in terms of time and space complexity, and compare it with some existing schemes.     Constraining the Inner Galactic DM Density Profile with H.E.S.S.

This article is an open access article distributed under the terms and conditions of the Creative Commons Attribution (CC BY) license (https://creativecommons.org/licenses/by/4.0/).In this short review, corresponding to a talk given at the conference “Cosmology 2023 in Miramare”, we combine an analysis of five regions observed by H.E.S.S. in the Galactic Center, intending to constrain the Dark Matter (DM) density profile in a WIMP annihilation scenario. For the analysis, we include the state-of-the-art Galactic diffuse emission Gamma-optimized model computed with DRAGON and a wide range of DM density profiles from cored to cuspy profiles, including different kinds of DM spikes. Our results are able to constrain generalized NFW profiles with an inner slope ≳1.3 . When considering DM spikes, the adiabatic spike is completely ruled out.
