    The difficulty in treating mood and anxiety disorders has brought about renewed clinical interest in alternative treatments, such as transcranial direct current stimulation (tDCS) of the dorsolateral prefrontal cortex (DLPFC). However, the optimal parameters for stimulation and underlying mechanisms of action are unclear. Psychiatric treatments have acute effects on emotional processing which predict later therapeutic action. Such effects have been proposed as cognitive biomarkers for screening novel treatments for depression and anxiety disorders. In this thesis the proposed cognitive, behavioural and neural underpinnings of mood and anxiety disorders are explored using tDCS of the DLPFC in humans. This is investigated with a series of studies which measure the acute effects of tDCS on emotional processing relevant to depression and trait anxiety or anxiety disorders. An initial investigation in healthy volunteers revealed an anxiolytic like effect (reduced threat vigilance) of a single session of tDCS on a behavioural test of proven clinical relevance. In addition, stimulation parameters were refined for future studies. The subsequent two behavioural investigations combined tDCS with an attentional bias modification (ABM) training paradigm. However, no bias modification was achieved in these studies and predictably, no subsequent effects on attentional bias, mood/ trait anxiety or worry were observed. The first of the ABM investigations included a measure of cortisol awakening response (CAR) and it was found that a single session of tDCS (regardless of ABM condition) reduced subsequent CAR in healthy volunteers, also suggesting an anxiolytic effect. The final study, in a sample of trait anxious females, used functional imaging to reveal that tDCS of the DLPFC increased frontal activation and reduced amygdala response to fearful face distractors during low cognitive effort. This provides the first causal evidence that modulating activity directly in the DLPFC inhibits amygdala response to threat in humans, and provides a potential neural mechanism for the previous reduction in vigilance to threat, and also for the efficacy of tDCS in the treatment of depression and anxiety disorders. The evidence from this thesis puts forward an anxiolytic-like effect of frontal tDCS on cognitive, neural and adrenal biomarkers relevant to clinical anxiety disorders, and indicates a potential cognitive mechanism (reduced fear vigilance) and an underlying neural mechanism (increased top-down control of amygdala response) that may partially mediate the reported clinical efficacy of prefrontal tDCS in the literature

    Polarity-dependent effects of transcranial direct current stimulation in obsessive-compulsive disorder

    About one third of patients with obsessive-compulsive disorder (OCD) fail to experience significant clinical benefit from currently available treatments. Hyperactivity of the presupplementary motor area (pre-SMA) has been detected in OCD patients, but it is not clear whether it is the primary cause or a secondary compensatory mechanism in OCD pathophysiology. Transcranial direct current stimulation (tDCS) is a noninvasive brain stimulation technique with polarity-dependent effects on motor cortical excitability. A 33-year-old woman with treatment-resistant OCD received 20 daily consecutive 2 mA/20 min tDCS sessions with the active electrode placed on the pre-SMA, according to the 10-20 EEG system, and the reference electrode on the right deltoid. The first 10 sessions were anodal, while the last 10 were cathodal. Symptoms severity was assessed using the Yale-Brown Obsessive Compulsive Scale (Y-BOCS) severity score. In the end of anodal stimulation, OCD symptoms had worsened. Subsequent cathodal stimulation induced a dramatic clinical improvement, which led to an overall 30% reduction in baseline symptoms severity score on the Y-BOCS. Our study supports the hypothesis that pre-SMA hyperfunction might be responsible for OCD symptoms and shows that cathodal inhibitory tDCS over this area might be an option when dealing with treatment-resistant OCD

    BACKGROUND: Presupplementary motor area (pre-SMA) hyperactivity has been detected in obsessive-compulsive disorder (OCD) patients. However, it is not understood whether this is a putative primary cause or a compensatory mechanism in OCD pathophysiology. Considering the polarity-dependent effects on cortical excitability of transcranial direct current stimulation (tDCS), we applied cathodal and/or anodal tDCS to the pre-SMA of OCD patients to test which current polarity might better improve symptoms. METHODS: Twelve OCD patients received initially 10 anodal (n = 6) or cathodal (n = 6) daily consecutive 2 mA/20 min tDCS sessions with the active electrode placed bilaterally on the pre-SMA. In case of improvement or no change in symptoms severity, the subjects were maintained on the same current polarity for 10 more sessions. In case of symptoms worsening after the first 10 sessions they were switched to the other polarity for 10 more sessions to test the hypothesis of a polarity-dependent effect. Therefore, each subject received 20 tDCS sessions. The Yale-Brown Obsessive-Compulsive Scale (Y-BOCS) and the Sheehan Disability Scale (SDS) were administered biweekly to assess changes in symptoms severity. RESULTS: After 10 sessions, 50% of patients who initially received anodal stimulation were switched to cathodal, while 100% of patients initially assigned to cathodal stimulation continued on the same polarity. At the end of the study, a statistically significant decrease was observed in the mean Y-BOCS scores of those patients who underwent cathodal tDCS. No pre-post difference was found in the scores of patients following anodal tDCS. CONCLUSIONS: Cathodal but not anodal tDCS over the pre-SMA significantly improved OCD symptoms

    Because it is generally known that Andre Gide is one of France's most influential contemporary writers, there is no need to justify a study based on his works. Desirous of obtaining the opinion of an influential white author concerning Negroid people and learning his activities in their behalf, the writer of this thesis undertook the study, Andre Gide end the Negro. The value of such a study to the American Negro lies primarily in (1) a better acquaintance with and appreciation for one whose interest in darker people has resulted in a tangible contribution; (2) a knowledge of the condition of a people, who, though distant in territory, are kindred in race, and (3) a challenge for scholastic accomplishment. The method of procedure was a careful analysis of Voyage au Congo and Retour du Tchad supplemented by collateral readings. To secure information concerning Mr. Gide's official investigation in 1938 of the natives' educational facilities in Senegal, a letter was sent him. Despite the anxiety which must be his because of the present war in which his country is involved, the eminent author found time to respond. His reply is quoted on page thirty-one of this study. Grateful acknowledgment is hereby given both Mr. Gide for his amicable letter and Professor Cook, who made such a contact possible. The various factors for the evolution of Gide's interest in Negroid people are shown in Chapter I. Chapter II contains a discussion of the colonial abuses existing in French Equatorial Africa exposed by Gide and his efforts to eradicate these. Through Chapter III one learns the author's personal impression of the morale and intelligence of Negroid people. In Chapter IV an interpretation of Gide's literary art in Voyage au Gang and Retour du Tchad is presented. The results are summarized in the conclusion

    Andre Gide: a study of the relationship between the author and his characters

    Andre Paul Guillame Gide was a controversial French writer. Some critics claimed he never wrote about anyone but himself; others denied that he used himself as a model for any but his first works. This paper is the result of a study to determine to what extent the characters of Andre Gide are autobiographical. The novel and novelle of Gide were chosen as a basis for this study because their characters are definite and their situations relatively clear. Each major character in these works was analyzed and his characteristics and actions noted in detail. Then a study was made of the author by reading his own autobiographical works and many biographies and studies of Gide. The characteristics and actions of Andre Gide were analyzed and noted. With the personalities of the author and his characters broken down into habits, characteristics and actions and organized, each character was again observed and his traits and attitudes matched with those of the author whenever this was possible.
