    Existence and stability analysis of spiky solutions for the Gierer-Meinhardt system with large reaction rates

    We study the Gierer-Meinhardt system in one dimension in the limit of large reaction rates. First we construct three types of solutions: (i) an interior spike; (ii) a boundary spike and (iii) two boundary spikes. Second we prove results on their stability. It is found that an interior spike is always unstable; a boundary spike is always stable. The two boundary spike configuration can be either stable or unstable, depending on the parameters. We fully classify the stability in this case. We characterise the destabilizing eigenfunctions in all cases. Numerical simulations are shown which are in full agreement with the analytical results

There are still many unknowns on the implication of cellular factors that regulate the entry process. Methodology/Principal Findings: We performed a yeast two-hybrid screen using as bait the carboxy-terminal endodomain of S, which faces the cytosol during and after opening of the fusion pore at early stages of the virus life cycle. Here we show that the ezrin membrane-actin linker interacts with S endodomain through the F1 lobe of its FERM domain and that both the eight carboxy-terminal amino-acids and a membrane-proximal cysteine cluster of S endodomain are important for this interaction in vitro. Interestingly, we found that ezrin is present at the site of entry of S-pseudotyped lentiviral particles in Vero E6 cells. Targeting ezrin function by small interfering RNA increased S-mediated entry of pseudotyped particles in epithelial cells. Furthermore, deletion of the eight carboxy-terminal amino acids of S enhanced S-pseudotyped particles infection. Expression of the ezrin dominant negative FERM domain enhanced cell susceptibility to infection by SARS-CoV and S pseudotyped particles and potentiated S-dependent membrane fusion. Conclusions/Significance: Ezrin interacts with SARS-CoV S endodomain and limits virus entry and fusion.     Determination of wheat spike and spikelet architecture and grain traits using X-ray Computed Tomography imaging

Background: Wheat spike architecture is a key determinant of multiple grain yield components and detailed examination of spike morphometric traits is beneficial to explain wheat grain yield and the effects of differing agronomy and genetics. However, quantification of spike morphometric traits has been very limited because it relies on time-consuming manual measurements. Results: In this study, using X-ray Computed Tomography imaging, we proposed a method to efficiently detect the 3D architecture of wheat spikes and component spikelets by clustering grains based on their Euclidean distance and relative positions. Morphometric characteristics of wheat spikelets and grains, e.g., number, size and spatial distribution along the spike can be determined. Two commercial wheat cultivars, one old, Maris Widgeon, and one modern, Siskin, were studied as examples. The average grain volume of Maris Widgeon and Siskin did not differ, but Siskin had more grains per spike and therefore greater total grain volume per spike. The spike length and spikelet number were not statistically different between the two cultivars. However, Siskin had a higher spikelet density (number of spikelets per unit spike length), with more grains and greater grain volume per spikelet than Maris Widgeon. Spatial distribution analysis revealed the number of grains, the average grain volume and the total grain volume of individual spikelets varied along the spike. Siskin had more grains and greater grain volumes per spikelet from spikelet 6, but not spikelet 1–5, compared with Maris Widgeon. The distribution of average grain volume along the spike was similar for the two wheat cultivars. Conclusion: The proposed method can efficiently extract spike, spikelet and grain morphometric traits of different wheat cultivars, which can contribute to a more detailed understanding of the sink of wheat grain yield

    One of the challenges of neuromorphic computing is efficiently routing spikes from neurons to their connected synapses. The aim of this thesis is to design a spike-routing architecture for flexible connections on single-chip neuromorphic systems. A model for estimating area, power consumption, memory, spike latency and link utilisation for neuromorphic spike-routing architecture is described. This model leads to the proposal for a new spike-routing architecture with a hybrid addressing scheme and a novel synaptic encoding scheme. The proposed architecture is implemented in a SystemC simulation tool with a supporting tool for encoding arbitrary SNN topologies for the synapse encoding scheme. Running the simulations with synthetic benchmarks and a handwriting recognition SNN shows that the proposed architecture is memory-efficient and provides low latency spike-routing with high synaptic activation concurrency.Electrical Engineering | Circuits and System

    Efficient Spike-Coding with Multiplicative Adaptation in a Spike Response Model

    Neural adaptation underlies the ability of neurons to maximize encoded informa- tion over a wide dynamic range of input stimuli. While adaptation is an intrinsic feature of neuronal models like the Hodgkin-Huxley model, the challenge is to in- tegrate adaptation in models of neural computation. Recent computational models like the Adaptive Spike Response Model implement adaptation as spike-based addition of fixed-size fast spike-triggered threshold dynamics and slow spike- triggered currents. Such adaptation has been shown to accurately model neural spiking behavior over a limited dynamic range. Taking a cue from kinetic models of adaptation, we propose a multiplicative Adaptive Spike Response Model where the spike-triggered adaptation dynamics are scaled multiplicatively by the adap- tation state at the time of spiking. We show that unlike the additive adaptation model, the firing rate in the multiplicative adaptation model saturates to a maxi- mum spike-rate. When simulating variance switching experiments, the model also quantitatively fits the experimental data over a wide dynamic range. Furthermore, dynamic threshold models of adaptation suggest a straightforward interpretation of neural activity in terms of dynamic signal encoding with shifted and weighted exponential kernels. We show that when thus encoding rectified filtered stimulus signals, the multiplicative Adaptive Spike Response Model achieves a high coding efficiency and maintains this efficiency over changes in the dynamic signal range of several orders of magnitude, without changing model parameters

    Bivalent intra-spike binding provides durability against emergent Omicron lineages: Results from a global consortium

    The SARS-CoV-2 Omicron Variant of Concern (VoC) and its sublineages contain 31-36 mutations in spike and escape neutralization by most therapeutic antibodies. In a pseudovirus neutralization assay, 66 of the nearly 400 candidate therapeutics in the Coronavirus Immunotherapeutic Consortium (CoVIC) panel neutralize Omicron and multiple Omicron sublineages. Among natural IgGs, especially those in the receptor binding domain (RBD)-2 epitope community, nearly all Omicron neutralizers recognize spike bivalently, with both antigen binding fragments (Fabs) simultaneously engaging adjacent RBDs on the same spike. Most IgGs that do not neutralize Omicron bind either entirely monovalently or have some (22-50%) monovalent occupancy. Cleavage of bivalent-binding IgGs to Fabs abolishes neutralization and binding affinity, with disproportionate loss of activity against Omicron pseudovirus and spike. These results suggest that VoC-resistant antibodies overcome mutagenic substitution via avidity. Hence, vaccine strategies targeting future SARS-CoV-2 variants should consider epitope display with spacing and organization identical to trimeric spike

    Spike discharge prediction based on neuro-fuzzy system

    This paper presents the development and evaluation of different versions of neuro-fuzzy model for prediction of spike discharge patterns. The author aims to predict the spike discharge variation using first spike latency and frequency-following interval. In order to study the spike discharge dynamics, the author analyzed the cerebral cortex data of the cat. Adaptive neuro-fuzzy inference systems (ANFIS), Wang and Mendel, dynamic evolving neural-fuzzy inference system, hybrid neural fuzzy inference system, genetic for lateral tuning and rule selection of linguistic fuzzy system (GFS.LT.RS) and subtractive clustering and fuzzy c-means algorithms are applied for data. Among these algorithms, ANFIS and GFS.LT.RS models have better performance. On the other hand, ANFIS and GFS.LT.RS algorithms can be used to predict the spike discharge dynamics as a function of first spike latency and frequency with a higher accuracy compared to other algorithms

    Describing the stochastic dynamics of neurons using Hamilton’s equations of classical mechanics

    We consider the most likely behaviour of neuron models by formulating them in terms of Hamilton’s equations. Starting from a Lagrangian for a stochastic system, we describe how Hamilton’s equations of classical mechanics can be used to derive an equivalent description in terms of canonical co-ordinates and momenta. We give physical meaning to these generalized momenta; specifically they are linear combinations of the noise terms in the stochastic model. Pseudo-kinetic energy and potential energy terms are also derived. The conjugate momenta can be considered as growing modes, and by implication the most likely noise input to a system will grow exponentially at large times; this surprising prediction agrees with existing experimental work on a single neuron. For many-neuron models, multiple growing modes will exist, and the numerical analysis of these is more complicated; however, the approach may still provide insight on the more detailed dynamics of these systems
