Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. Evaluation of hematopoiesis and innate immune response mediated by macrophages in mice submitted to nutritional recovery after protein malnutrition
A desnutrição protéico-energética (DPE) afeta mais de 1 bilhão de pessoas no mundo, principalmente crianças, idosos e pacientes hospitalizados. Ela provoca alterações metabólicas e hormonais, além de afetar o tecido hemopoético. O comprometimento da hemopoese provoca anemia e leucopenia, modificando a resposta imune inata e adquirida do organismo. Dessa forma, é comum a associação entre desnutrição e infecção, levando ao comprometimento do tratamento e aumento da morbidade e mortalidade de indivíduos hospitalizados. Após a recuperação nutricional, é relatada a reversão das alterações bioquímicas e hormonais, bem como das alterações na hemopoese e na resposta imune. Porém, muitos resultados são controversos, existindo dúvidas quanto à reversibilidade das alterações. Assim, nos propusemos a avaliar os efeitos da recuperação nutricional nos parâmetros bioquímicos, hormonais, hematológicos e imunológicos em modelo murino de desnutrição. Os animais desnutridos apresentaram perda de peso significativa, redução de proteínas totais, albumina, glicose, insulina e IGF-1, bem como aumento de glutamina plasmática, glutamina sintetase muscular e corticosterona. Houve redução dos parâmetros hepáticos e musculares, bem como alteração na sensibilidade à insulina, evidenciada pelos testes de OGTT e ITT. Todas as alterações descritas caracterizam o quadro de desnutrição. Após a recuperação nutricional, alguns parâmetros foram normalizados, mas as concentrações de glicose, insulina e IGF-1 permaneceram reduzidas. Da mesma forma, as alterações na concentração de DNA hepático e na sensibilidade à insulina permaneceram nos animais renutridos. A pancitopenia periférica e hipocelularidade da medula óssea e do baço observadas nos animais desnutridos foram revertidas após a renutrição. A avaliação de macrófagos peritoniais mostrou reversão parcial do comprometimento da capacidade e adesão e espraiamento, bem como da atividade fungicida nos animais renutridos. A produção de peróxido de hidrogênio continuou baixa após a recuperação nutricional, enquanto a produção de óxido nítrico voltou a aumentar. O comprometimento da produção de citocinas pró-inflamatórias decorrente da desnutrição não foi completamente revertido, visto que, em camundongos Swiss Webster, somente a produção de TNF-α retornou ao normal, enquanto em camundongos C56BL/6J a produção de nenhuma citocina foi restabelecida. A avaliação da via de sinalização do fator de transcrição NFkB mostrou alteração na expressão de MyD88, TRAF-6, IkKβ e IkBα em animais desnutridos. Após a recuperação nutricional, algumas dessas proteínas não retornaram ao normal. Os animais desnutridos também apresentaram comprometimento da ativação de NFkB, que não foi normalizada após a recuperação nutricional. Sendo assim, é possível afirmar que o retorno a uma dieta normoprotéica não é suficiente para reverter todas as alterações causadas pela desnutrição.Protein-energy malnutrition (PEM) affects more than 1 billion people worldwide, mainly children, elderly and hospitalized patients. It causes metabolic and hormonal changes, besides affecting hematopoietic tissue. Impaired hematopoiesis causes anemia and leukopenia, modifying innate and acquired immune response of the organism. Thus, it is common the association between malnutrition and infection, leading to impaired treatment and increasing morbidity and mortality in hospitalized patients. After nutritional recovery, it is reported reversal of biochemical and hormonal changes, as well as, reversal of changes in hematopoiesis and immune response. However, many results are controversial, and there are doubts about the reversibility of the changes. Thus, we proposed to evaluate the effects of nutritional recovery biochemical, hormonal, haematological and immunological parameters in a murine model of malnutrition. The malnourished animals showed significant weight loss, reduction in total protein, albumin, glucose, insulin and IGF-1, as well as increased plasma glutamine, corticosterone and muscle glutamine synthetase. There was a reduction in muscle and liver parameters as well as change in insulin sensitivity, evidenced by the tests of OGTT and ITT. All modifications described characterize the malnutrition. After nutritional recovery, there was normalization of some parameters, but the concentrations of glucose, insulin and IGF-1 remained low. Likewise, changes in hepatic DNA concentration and insulin sensitivity remained in renourished animals. Peripheral pancytopenia and hypocellularity in bone marrow and spleen observed in malnourished animals were reversed after refeeding. The evaluation of peritoneal macrophages showed partial reversal of impairment of adhesion and spreading ability, as well as fungicidal activity in animals renourished. The hydrogen peroxide production remained low after nutritional recovery, while nitric oxide production increased again. Impaired production of proinflammatory cytokines due to malnutrition was not completely reversed, whereas in Swiss Webster mice, only the production of TNF-α returned to normal, whereas in C56BL/6J mice no cytokine production was restored. The assessment of the signalling pathway of transcription factor NFkB showed alterations in the expression of MyD88, TRAF-6 IkKβ and IkBα in malnourished animals. After nutritional recovery, some of these proteins didn\'t return to normal. Malnourished animals also showed impaired activation of NFkB, which wasn\'t normalized after nutritional recovery. Therefore, it is possible to say that the return to a normal diet is not enough to reverse all the changes caused by malnutrition
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Efeito da desnutrição protéica experimental na hemopoese medular
A desnutrição protéico-energética (DPE) é a forma de desnutrição mais encontrada em áreas subdesenvolvidas, atingindo principalmente recém-nascidos de baixo peso para sua idade gestacional, pacientes hospitalizados sob alimentação parenteral, idosos, crianças pré-escolares, indivíduos com distúrbios alimentares como na anorexia nervosa e bulimia e pacientes portadores de neoplasias, doenças crônicas e sob quimioterapia. No sistema hematopoético, a proliferação e diferenciação celular ocorrem de forma constante e equilibrada, sensível às demandas de linhagens celulares específicas. O comportamento medular da população de cada linhagem hematopoética é controlado pelo equilíbrio entre auto-renovação das células tronco, a diferenciação e a morte celular. Alterações estruturais e celulares em órgãos hematopoéticos têm sido relatadas na DPE, com redução da celularidade de medula óssea e da sua capacidade de resposta a estímulos inflamatórios e/ou infecciosos em indivíduos desnutridos. Neste trabalho estudamos o efeito da deficiência protéica experimental em diferentes linhagens hematopoéticas, através da realização do hemograma e do mielograma; avaliamos a capacidade proliferativa ex vivo por técnicas imunocitoquímica (detecção de regiões organizadoras de nucléolo marcadas pela prata - AgNOR) e de imunohistoquímica (detecção do antígeno de proliferação nuclear - PCNA) e, em ensaios de cultura líquida e clonogênicos em metilcelulose. Paralelamente, avaliamos o perfil imunofenotípico e estudos morfológicos de células da medula óssea em DPE. Células não-derentes foram cultivadas em meio líquido (CL), avaliando-se durante 1,3 e 5 dias, e células totais foram cultivadas em metilcelulose, avaliando-se durante 7, 14 e 21 dias, em ambos os ensaios, a proliferação celular em hemocitômetros e por imunofenotipagem as populações celulares presentes nesses ensaios, sendo ambos suplementados com IL3, EPO, GM-CSF e G-CSF. Os resultados mostraram que a hemopoese é afetada pela desnutrição protéica, pois encontramos hipoplasia medular, destacando-se depleção dos setores eritróides, grânulo-monocíticos e megacariocítico e bloqueio maturativo. A anemia e leucopenia demonstradas são decorrentes da hipoplasia medular e, esta parece não ser dependente da apoptose. Os resultados da análise imunofenotípica evidenciaram que as células da medula óssea de animais desnutridos, apresentam redução na expressão de CD 45, CD34, CD38, CD117, TER 119 e CD71. As populações linfoides também foram afetados pela desnutrição, destacando-se a depleção de células linfóides primitivas e maturas. Além disso, foi verificado que animais desnutridos apresentam menor produção de células sob ação e estímulo de fatores de crescimento externo, nos ensaios de CL e CSS. A análise morfológica revelou redução no número de células viáveis e de progenitores granulo-monocíticos e eritróide, sugerindo alterações intrínsecas nestas células. Os resultados obtidos tanto com PCNA e AgNOR em amostras da medula óssea foram similares: os animais desnutridos apresentaram menor número de células em atividade proliferativa. O presente trabalho sugere proliferação celular medular reduzida na desnutrição protéica, ocasionando uma hipoplasia, principalmente do setor mielóide, decorrente da diminuição da existência de células primitivas e de progenitores bem como de sua capacidade proliferativa, da disfunção da ação regulatória de fatores de crescimento, de alterações nos receptores transmembrana e os mecanismos transducionais, modificações histológicas da MO e modificações dos componentes do microambiente e da relação arquitetural desse microambiente.The protein-energy malnutrition (PEM) is the most disseminated kind of malnutrition in underdeveloped areas and the most common kind of malnutrition in newborns who are underweight according to their gestation age, hospitalised and receiving parenteral feeding patients, elderly people, individuals with nutrition problems such as anorexia nervosa and bulimia, patients who have neoplasia or chronic diseases, patients undergoing chemotherapy. In the haematopoietic system, the cellular proliferation and differentiation occur in a constant and balanced way, sensitive to specific cellular lineage demands. The stem cells auto-renewing, differentiation and cellular death control the population rate of each haematopoietic lineage. Structure and cellular alterations in haematopoietic organs have been mentioned in PEM. The reduction of the cell number of bone marrow and the incapacity to increase its production in answer to inflammatory and/or infectious stimulus may be important mechanisms responsible for the rising in the number of infections in individuals with malnutrition. The aim of this essay is to study the effect of the experimental protein deficiency in different haematopoietic lineages, as well as on the proliferation capacity ex vivo by immunocitochemical techniques (detection of silver stained organizer regions -AgNOR) and immunohistochemical (detection of proliferating cell nuclear antigen- PCNA), and in vitro cultivated in liquid culture (CL) and clonogenic assays (EC). Besides that, it evaluates the immunophenotyping profile and morphological studies in DPE. Cells with no adhesion were cultivated in liquid culture (CL), evaluating during 1, 3 and 5 days while the clonogenic assays (EC), evaluating during 7, 14 and 21 days the cellular proliferation, in analysis of cell number and by immunophenotyping the present cellular populations, being both the assays supplemented with IL3, EPO, GM-CSF, G-CS. The results showed that the haemopoiesis is affected by the protein malnutrition: marrow hypoplasia increasing was found depletion of the erythroid and granulo-monocytic components, and in more severe degrees of malnutrition, also the depletion of the megacaryocytic component and a blockage of maturation. The demonstrated anaemia and leucopoenia are due to the marrow hypoplasia at least, after the malnutrition process installation the hypoplasia is not a result of the apoptosis increase in medulla cells. The results made evident that the marrow cells of animals with malnutrition, showed reduction in the number and expression of CD45, CD34, CD117, TER 119 and CD71. Linphoid populations were also affected by malnutrition, standing out the depletion of primitive and mature linphoid cells. Moreover, it was found out that animals with malnutrition show cell production under action and stimulus of external growth factors in assays of the CL and CSS. The morphological analysis revealed reduction in the cells number and erythroid and granulo-monocytic progenitors, suggesting intrinsic changes in these cells. The obtained results, with PCNA and AgNOR in bone marrow were similar: the animals with malnutrition showed a low number of cells in proliferation activity. This present essay allowed the analysis revealed reduction in the cells number in DPE with marrow hypoplasia of the different hemopoietics populations, and the possible mechanisms may be: reduction in the existence of primitive cells and progenitors as well as their proliferation capacity; regulatory action of growth factors, of the transmembrane receptors and the signaling mechanisms and of the components of the microenvironment and of the architectural structure of this microenvironment
Evaluation of cell cycle by hematopoietic cells of malnourished mice
As diferentes formas da desnutrição acometem milhões de pessoas em todo mundo, afetando geralmente crianças, recém-nascidos abaixo do peso para sua idade gestacional, idosos e pacientes sob nutrição parenteral. A desnutrição protéica (DP) induz atrofia em órgãos hemopoéticos, induzindo pancitopenia de sangue periférico, modifica a resposta imune específica e inespecífica, e desta forma, a associação entre doença e DP é freqüente. Alguns estudos indicam que a desnutrição produz vários efeitos celulares como redução da capacidade de proliferação, em vários órgãos. Como conseqüência da constante e elevada demanda de proteína pelo tecido hemopoético, este pode apresentar alterações qualitativas e quantitativas em condições de desnutrição protéica. Neste trabalho avaliamos os efeitos da DP sobre o ciclo celular de células de medula óssea (MO) de camundongos. Camundongos Swiss machos, adultos, foram alimentados ou com ração contendo 2% de proteína (grupo desnutrido, D) ou com ração com 20% de proteína (grupo controle, C). A fonte protéica utilizada foi caseína. Após perda de 25% de peso corpóreo do grupo D, colheu-se sangue e células da MO e do baço. As células do sangue foram quantificadas bem como as do baço e da MO. As células da medula óssea foram posteriormente coradas com laranja de acridina (AO) e Iodeto de propídeo (PI), avaliando-se, por citometria de fluxo, respectivamente DNA/RNA e DNA. Estes mesmos procedimentos foram executados após 10 e 15 dias da administração, em ambos os grupos, do quimioterápico 5-Fluoracil (que leva as células a estado de repouso -G0). Os animais do grupo desnutrido, assim como aqueles após 10 e 15 dias da administração do 5-Fluoracil, apresentaram anemia, além de redução significativa de número de hemácias, hematócrito e reticulócitos, e alterações quantitativas nas linhagens medulares e do baço. Os dados da citometria de fluxo (expressos em % de células da população analisada) mostram que em DP, uma maior quantidade de células da MO encontra-se fora do ciclo celular (G0), e uma menor população de células nas fases proliferativas do ciclo celular, S/G2/M, em populações de todas linhagens medulares, exceto eritróide e linfóide B, e todo os estágios maturativos, assim como células Sca-1, células tronco murina. Esta maior população celular em G0 também é observada após o efeito do 5-Fluoracil, o que corrobora a suposição de incapacidade de retorno ao ciclo celular por parte das células de medula dos animais desnutridos. Estes resultados podem explicar, em parte, a hipoplasia medular e pancitopenia sanguínea.The different forms of the malnutrition attacks millions of people in everybody generally, affecting children, just-born below of the weight for its gestacional age, aged and patient under parenteral nutrition. The protein malnutrition induces atrophy in haematopoietic organs, inducing pancitopenia of peripheral blood, modifies the specific and inespecífica immune reply, and of this form, the association between illness and DPE are frequent. Some studies indicate that the malnutrition produces some effect cellular as reduction of the proliferation capacity, in some organs. As consequence of the constant and raised protein demand for the bone marrow, that can present qualitative and quantitative alterations in conditions of protein malnutrition. In this work we evaluate the effect of the DP on the cellular cycle of cells of bone marrow of mice. Mice Swiss males, adults, had been fed or with ration I contend 2% of protein (unfed group, D) or with ration with 20% of protein (group has controlled, C). The used protein source was casein. After loss of 25% of corporeal weight of group D, harvested blood and cells of bone marrow and spleen. The cells of the blood had been quantified as well as the ones of spleen and bone marrow. The cells of the bone marrow later had been coradas with orange of acridine (AO) and propidium iodide of (PI), evaluating, for citometria of flow, respectively DNA/RNA and DNA. These same procedures had been executed after 10 and 15 days of the administration, in both the groups, of the 5-Fluoracil (that it takes the cells the state of rest - G0). The animals of the malnutrition group, as well as those after 10 and 15 days of the administration of the 5-Fluoracil, had presented anemia, beyond significant reduction of number of erythrocytis, hematocrit and reticulocytes, and quantitative alterations in the progenitors by bone marrow and of spleen. The data of the flow cytometric (express in % of cells of the analyzed population) show that in DP, a bigger amount of cells of outside meets bone marrow of the cellular cycle (G0), and a lesser population of cells in the proliferative phases of the cellular cycle, S/G2/M, in populations of all progenitors by bone marrow, except erythroids and lymphoids B, and all the maturative periods of training, as well as cells Sca-1, stem cells murina. This bigger cellular population in G0 also is observed after the effect of the 5-Fluoracil, what it corroborates the assumption of incapacity of return to the cellular cycle on the part of the cells of marrow of the malnutrition animals. These results can explain, in part, the hipoplasia to medular and sanguineous pancitopenia
Evaluation aspects of the inflammatory response unchained by LPS in experimental protein malnutrition. Quantification the LPS\' receptors (CD14/TLR4) and the transcription factor NFκB
Sabe-se que a desnutrição modifica a resposta imune específica e inespecífica do organismo frente a agentes infecciosos comprometendo a produção e a função de células linfo-hemopoéticas, estando associada a modificações da resposta imune, resultando em maior suscetibilidade à infecções, porém os mecanismos exatos que comprometem o sistema imune em estados de desnutrição ainda estão para serem esclarecidos. A literatura relata que aproximadamente 60% das infecções que evoluem para sepse são adquiridas no ambiente hospitalar, envolvendo geralmente bactérias Gram negativas e incidindo especialmente em indivíduos com nutrição inadequada. Considerando tais aspectos e em função da complexidade da interação do estado nutricional e resposta do organismo frente a agentes patogênicos, envolvendo controles celulares e moleculares múltiplos ainda pouco conhecidos, propusemo-nos a estudar alguns aspectos da resposta inflamatória em desnutrição. Camundongos Swiss machos adultos, submetidos a desnutrição protéica-energética, após perda de aproximadamente 25% do peso corpóreo, foram inoculados com lipopolissacarideo de Escherichia coli: Hemograma, mielograma, esplenograma e dosagens sistêmicas de citocinas, foram realizadas. Células coletadas da cavidade peritonial de animais que não foram estimulados com LPS foram utilizadas para as determinações de citocinas e NO in vitro, bem como para quantificação dos receptores CD14 e TLR-4/MD-2 e do fator de transcrição NFκB. Células da cavidade peritonial foram usadas, também, para realização dos testes de espraiamento, fagocitose e atividade cida com Candida albicans. Animais desnutridos apresentaram anemia, leucopenia; severa redução na celularidade da medula óssea, do baço e da cavidade peritonial. A capacidade de espraiamento, fagocitose, atividade cida e síntese de citocinas e NO foram significativamente menores nos animais dos grupo desnutrido. O número de receptores de CD14 e TLR-4/MD-2 e do fator de transrição NFκB, também foram significativamente menores nos animais desnutridos. Estes achados sugerem que. animais desnutridos apresentam resposta deficiente frente ao LPS. A menor expressão de receptores CD14 e TLR-4/MD-2 pode ser responsável, em parte pela imunodepressão observada. Os dados nos levam a inferir que o estado nutricional interfere no estado de ativação de macrófagos e na capacidade de resposta dos animais.Malnutrition modifies the specific and non-specific immune response of the organism to infectious agents, hampering the production and function of Iympho-hemopoietic cells leading to a higher susceptibility of the orgonism to infections. However, the exact mechanisms by which the immune system is undermined has not yet been fully elucidated. Approximately 60% of infections that evolve to bacteremia are nosocomial, and usually involve Gram negative bacteria in individuais that have inadequate nutrition. Taking this into consideration, and in view of the complexity of the interaction between nutritional state and the organism\' s response to infection, which involves poorly known multiple cellular and molecular controls, we proposed to study a few aspects of the inflammatory response in protein malnutrition. Male, outbred Swiss mice were sumbitted to protein malnutrition and after the loss of about 25% of total body weight they were inoculad whith lipopolissacharide of Escherichia coli: Hemogram, mielogram, splenogram and the determination of systemic production of cytokines were used to evaluate. Cells from the peritoneal cavity of animais who were not inoculated were collected for determination of cytokines and NO production in vitro and to evaluate the expression of NFκB and CD14 and TLR-4/MD-2 receptors. Cells from the peritoneal cavity were used too for the spreading, phagocytosis and killing tests with Candida albicans. Malnourished animais presented anemia, leucopenia a severe reduction on bone marrow, spleen and peritoneal cavity cellularity. The spreading, phagocytosis, killing and the production of cytokines and NO was significantly lower in malnourished animais. The number of CD14 and TLR-4/MD-2 receptors and NFκB was found to be significantly lower in malnourished animais. These findings suggest that malnourished mice present a deficient response to LPS. The smaller expression of CD14 and TLR-4/MD-2 receptors may be partly responsible for the immunodeficiency observed. These data lead us to infer that the nutritional state interferes in the activation of macrophages and in the immune response capacity
