Secondary metabolites by chemical screening, 41 - Structure and biosynthesis of mutolide, a novel macrolide from a UV mutant of the fungus F-24 ' 707

    No full text
The 14-membered macrolide, mutolide (1), was discovered by chemical screening of the culture broth of the fungus F-24'707y, obtained after UV mutagenesis of the wild type strain, which normally produces the spirobisnaphthalene cladospirone bisepoxide (2). The structure of 1 was established by detailed spectroscopic analysis, X-ray analysis and derivatisation. The biogenetic origin of the carbon skeleton and the hydroxy groups was verified by feeding sodium [1-C-13]acetate and O-18(2) to growing cultures of the fungus. Macrolide 1 is generated from acetate/malonate only. The unexpected change of the normal metabolite pattern of this strain is discussed, and proves the value of the OSMAC method

Secondary metabolites by chemical screening, 42 - Cladospirones B to I from Sphaeropsidales sp F-24 ' 707 by variation of culture conditions

Variation of the culture conditions - static surface cultures in particular - of the fungus Sphaeropsidales sp. (strain F-24'707), which produces cladospirone bisepoxide (1), led to the isolation of eight new spirobisnaphthalenes - the cladospirones B to I (8-15) -together with seven known representatives of this class of secondary metabolites. Cladospirones C (9) and D (11) show antibiotic activity against bacteria and algae. The structures of cladospirone B (8) and E (12) were confirmed by X-ray structure analysis. Cladospirones C (9) and G to I (10, 14-15) represent new members of the spirobis-naphthalene family, thanks to their hydroxylation patterns. Moreover, they underline the extraordinary status of this interesting class of compounds as the most diverse secondary metabolites, allowing for their small number of carbon atoms, described to date. Almost all possible permutations of stereochemistry and oxygen substitution pattern on the C-10 skeleton are produced by different fungi

Adrenomedullin refines mortality prediction by the BODE index in COPD - The "BODE-A" index.

peer reviewedThe BODE index is well-validated for mortality prediction in COPD. Concentrations of plasma proadrenomedullin, a surrogate for mature adrenomedullin, independently predicted 2-year mortality among inpatients with COPD exacerbation. We compared accuracy of initial proadrenomedullin level, BODE, and BODE components, alone or combined, in predicting 1-year or 2-year all-cause mortality in a multicenter, multinational observational cohort with stable, moderate to very severe COPD.Proadrenomedullin was significantly associated (P<0.001) with 1-year mortality (4.7%) and 2-year mortality (7.8%), and comparably predictive to BODE regarding both (C statistics: 0.691 vs. 0.745, 0.635 vs. 0.679). Relative to using BODE alone, adding proadrenomedullin significantly improved 1-year and 2-year mortality prognostication (C statistics: 0.750, 0.818; both P<0.001). Proadrenomedullin plus BOD was more predictive than was the original BODE including 6-minute-walk distance. In multivariable analysis, proadrenomedullin (LR X2 13.0, P<0.001), body mass index (8.5, P=0.004), and 6-minute-walk distance (7.5, P=0.006), but not modified MMRC dyspnoea score (2.2, P=0.14) or FEV1 % predicted (0.3, P=0.60), independently foretold 2-year survival.Proadrenomedullin plus BODE better predicts mortality in COPD patients than does BODE alone; proadrenomedullin may substitute for 6-minute-walk distance in BODE when 6-minute-walk testing is unavailable

Correlation study of semi-quantitative CT emphysema score with the BODE index in COPD patients

    No full text
Background: Chronic obstructive pulmonary disease (COPD) is characterized by airflow limitation due to airway narrowing and loss of elastic recoil. Emphysema is the hallmark of elasticity loss and can be assessed by CT scan by measuring low attenuation areas (LAA). The BODE staging system, including FEV1 and other variables, helps to predict mortality and hospitalization for COPD. Aim: To assess any relationship between BODE index and CT extension of emphysema in mild-to-severe COPD patients. Patients and study design: Thirty-three out of 48 enrolled out-patients were included in the study (M/F: 24/9, mean age±SD: 67±8.9 yrs; smoking history: 34±19 pack/yrs). BODE index and semi-quantitative CT scoring of emphysema were calculated for each patient within two weeks from enrolment. Results: The mean BODE index and CT emphysema score were 4.3±3.2 and 1±1.2, respectively. No emphysema was recorded in 16 (48%) patients; the emphysema score was 1 (less than 25% of LAA) in seven cases, 2 (between 25 and 75%) in 6 cases and 3 (more than 75%) in 3 patients. Severe emphysema (score: 4) was reported in one patient. We found no correlation between the emphysema score and the BODE index (p= 0.348; r=0.168). Similarly, no correlation was found with FEV1, the level of dyspnoea (MMRC score) and the 6-minute walking distance (m). However, a weak inverse correlation was found with the body mass index (p 0.038; r -0.362). Conclusions: Our preliminary results suggest that no correlation exists between BODE index and semi-quantitative CT emphysema scoring. Further efforts better assessing emphysema and other COPD-related pathological abnormalities, i.e. airway remodelling, are needed in larger samples siz
