A laboratory bioassay for evaluating pathogenicity of Macrophomina phaseolina and Rhizoctonia solani isolates to strawberry stolons

    No full text
Macrophomina phaseolina and Rhizoctonia spp. are the most important soilborne pathogens of the strawberry crop that cause seriously reduced yields. Various methods are being used to determine pathogenicity of these fungi isolated from strawberry; however, they take a long time to grow strawberry plants. They also encounter some problems to provide a large number of healthy plants for the pathogenicity tests. Therefore, the aim of this study was to develop a faster and more reliable pathogenicity test for soilborne fungal pathogens of strawberry by using the stolons of healthy strawberry plants. The stolon inoculation method was demonstrated to be easy and rapid for properly distinguishing soilborne fungal species of strawberry, whether pathogenic or non-pathogenic, and also for assessing the isolates for virulence

Effects of soil amendments combined with solarization on the soil microbial community in strawberry cultivation using quantitative real-time PCR

Experiments were conducted in commercial fields of strawberry-growing areas of the Aydin Province of Turkey during two cropping seasons: 2010-2011 and 2011-2012. Each year, eight separate treatments were used: (1) untreated control (C), (2) solarization alone (S), (3) solarization + broccoli (SBr), (4) solarization + fava bean (SFB), (5) solarization + dry olive-mill wastewater (SDOMW), (6) solarization + rice hulls (SRH), (7) solarization + sulfur powder (SSu), and (8) solarization + vermicast (SVe). DNA was extracted from bulk soil samples before and after solarization. The populations of the major taxonomic groups of bacteria and soil-borne fungal pathogens of strawberry were quantitatively calculated by quantitative real-time PCR (qPCR) with specific primer pairs using standard curves. The marketable fruit yield was recorded in the experimental plots. After the 6-week solarization period, there were significant reductions for total bacteria and for alpha-Proteobacteria in all of the experimental plots, except for SDOMW. However, the abundance of beta-Proteobacteria significantly increased in all of the experimental plots (except for C and SFB in 2011). Significant increases in Firmicutes and Actinobacteria were also recorded in all of the treated plots. The highest significant increase was noticed with SDOMW treatments for total bacteria, alpha-Proteobacteria, beta-Proteobacteria, Firmicutes and Actinobacteria in both years. The target level of Verticillium spp. Rhizoctonia solani and Fusarium oxysporum decreased significantly in all of the treated plots after solarization in both years. The reductions of M. phaseolina were significant in S, SVe, and SSu and were the highest in SDOMW in 2011; these reductions were significant in S, SSu and SDOMW in 2012. The abundance of Trichoderma spp. decreased insignificantly in all of the experimental plots. The highest significant yields were obtained from the combination of SDOMW, SRH and SSu compared to solarization alone in both seasons

Beyond the genetic code in leaf senescence

    No full text
Leaf senescence is not only genetically programmed but also induced by exogenous stress to ensure completion of the plant life cycle, successful reproduction and environmental adaptability. Genetic reprogramming is a major aspect of leaf senescence, and the senescence signaling that follows is controlled by a complex regulatory network. Recent studies suggest that the activity of transcription factors together with epigenetic mechanisms ensures the robustness of this network, with the latter including chromatin remodeling, DNA modification, and RNA-mediated control of transcription factors and other senescence-associated genes. In this review, we provide an overview of the relevant epigenetic mechanisms and summarize recent findings of epigenetic regulators of plant leaf senescence involved in DNA methylation and histone modification along with the functions of small RNAs in this process © The Author(s) 2017. A general view of poverty in Turkey as an issue for social work in the light of behavioral finance and game theory

#nofulltext# --- Cesur Kılıçaslan, Seher (Arel Author)In this chapter, the authors define poverty in general terms before including statistics for a detailed, Turkey-Specific discussion. Once the authors elaborate on the causes of poverty, they introduce behavioural economics and game theory, the fundamental aim of the chapter being to examine how these two theories affect perceptions of poverty and the struggle against poverty. Another issue that enters into the scope of this chapter is to what extent the poor themselves are responsible for their own poverty. On this question, game theory and behavioural economics can potentially be marshalled against the poor. However, we also argue that, by using a different approach, both theories can be interpreted in the poor's favour. We examine the Double-Sided nature of these two theories in detail and stress how important it is in the study of poverty to consider the disadvantageous position in which the poor find themselves. © 2015, IGI Global. Reklamın tüketimi arttırmaya yönelik etkisine maruz kalan birey, kendisinde ve yaşam koşullarında sürekli eksiklik hisseder; içinde bulunduğu durumla olmak istediği durum arasındaki çelişkiyi her gün yeniden yaşar.

Farklı yaşam dönemlerinde karşılaştıkları psiko-sosyal sorunları çözme noktasında ağırlıklı olarak enformel ağlardan faydalandıkları bilinen LGBTİ'lere karşı devletin sosyal hizmet/destek kapsamındaki yükümlülükleri belirlenmeye çalışılmış ve yaşanan güçlükleri aşmaya yönelik önerilerde bulunulmuşturThis study aims to define the requirements of effective provision of social services to LGBTIs (lesbian, gay, bisexual, transgender and intersex) in Turkey. Similar to other vulnerable groups/minorities, LGBTIs are also faced with challenges in meeting their basic necessities, most of which has an economic base such as housing, education, health, employment, social security, personal security, and need support to overcome these problems. However, as a result of discrimination based on sexual orientation or gender identity originating from the heteropatriarchal structure of the society, LGBTIs are excluded from the mechanisms where they can receive institutional support. In Turkey, it is seen that the Ministry of Family and Social Policies, the primary institution responsible for providing social services to those who are vulnerable/need support, does not put the problems of LGBTIs on its agenda and leaves them marginalized vis-à-vis economic and social problems. In the present study, considering the fact that LGBTIs have the right to enjoy social services as anyone else, the necessity of social policy decision makers and social service providers to take them into account is taken as a point of departure. Furthermore, it is aimed to determine the government's liabilities within the context of providing social services/support to LGBTIs, who are known to resort to informal networks mainly to solve their psycho-social problems at different phases of their lives, and several suggestions to overcome the current challenges are mad

Crowdshipping: The level of trust towards crowdshipping from the user’s perspective: A stated preference experiment

    No full text
Thanks to growing online shopping, last mile logistics is becoming a more relevant problem for cities due to its negative impacts, such as congestion and environmental problems. In this research, one of the urban freight transport services aiming to tackle these externalities is analysed: crowdshipping. Crowdshipping is a service where the package is delivered via a traveller who is already making an unrelated trip. Trust is a key concept affecting the adoption of crowdshipping yet to be explicitly investigated . Thereby, this research aims to explore the effect of trust and examine how users' adoption can be achieved. To analyse the stated gap, a stated choice experiment is conducted to test the effect of travel time, travel cost, track and trace, insurance, damage and reputation. A Mediation Choice Model is applied to explore how the relevant attributes would impact trust towards the service adoption. Based on the findings, the direct effects of all the selected attributes were found significant, except for tracking and tracing. Regarding the indirect effects, all the main attributes were statistically significant, meaning that trust has a mediating effect on the adoption of the service. Additionally, the heterogeneity in preferences is explored through a Latent Class Choice Model resulting in a two-class latent model, crowdshipping sceptics and crowdshipping enthusiasts, the latter being more likely to be composed of younger women. Although enthusiasts consider more features while opting for crowdshipping, the delivery company's reputation and delivery cost were the most important factors in crowdshipping service adoption.Transport, Infrastructure and Logistic

Gene transfer in the liver using recombinant adeno-associated virus

First author Seemin Seher Ahmed is a doctoral student in the Interdisciplinary Graduate Program in the Graduate School of Biomedical Sciences (GSBS) at UMass Medical School.Liver-directed gene transfer and gene therapy are rapidly gaining attention primarily because the liver is centrally involved in a variety of metabolic functions that are affected in various inherited disorders. Recombinant adeno-associated virus (rAAV) is a popular gene delivery vehicle for gene therapy, and intravenous delivery of some rAAV serotypes results in very efficient transduction in the liver. rAAV-mediated gene transfer to the liver can be used to create somatic transgenic animals or disease models for studying the function of various genes and miRNAs. The liver is the target tissue for gene therapy of many inborn metabolic diseases and may also be exploited as a "biofactory" for production of coagulation factors, insulin, growth hormones, and other non-hepatic proteins. Hence, efficient delivery of transgenes and small RNAs to the liver by rAAV vectors has been of long-standing interest to research scientists and clinicians alike. This unit describes methods for delivery of rAAV vectors by several injection routes, followed by a range of analytical methods for assessing the expression, activity, and effects of the transgene and its product. Globally invading populations of the fungal plant pathogen Verticillium dahliae are dominated by multiple divergent lineages

The spread of aggressive fungal pathogens into previously non-endemic regions is a major threat to plant health and food security. Analyses of the spatial and genetic structure of plant pathogens offer valuable insights into their origin, dispersal mechanisms and evolution, and have been useful to develop successful disease management strategies. Here, we elucidated the genetic diversity, population structure and demographic history of worldwide invasion of the ascomycete Verticillium dahliae, a soil-borne pathogen, using a global collection of 1100 isolates from multiple plant hosts and countries. Seven well-differentiated genetic clusters were revealed through discriminant analysis of principal components (DAPC), but no strong associations between these clusters and host/geographic origin of isolates were found. Analyses of clonal evolutionary relationships among multilocus genotypes with the eBURST algorithm and analyses of genetic distances revealed that genetic clusters represented several ancient evolutionary lineages with broad geographic distribution and wide host range. Comparison of different scenarios of demographic history using approximate Bayesian computations revealed the branching order among the different genetic clusters and lineages. The different lineages may represent incipient species, and this raises questions with respect to their evolutionary origin and the factors allowing their maintenance in the same areas and same hosts without evidence of admixture between them. Based on the above findings and the biology of V. dahliae, we conclude that anthropogenic movement has played an important role in spreading V. dahliae lineages. Our findings have implications for the development of management strategies such as quarantine measures and crop resistance breeding
