    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Effects of fibroblast growth factor 8 and 18 on ovine ovarian granulosa cell function

    Fibroblast Growth factors (FGFs) are growth factors which have diverse biological activities including broad mitogenic and cell survival activities. FGFs constitute a large family of 22 distinct polypeptide growth factors varying in size from 17 to 34 kDa and have between 13 to 71% sequence homology. More specifically, FGF8 and FGF18 are both important modulator of granulosa cell functions. FGF8 and FGF18 are homologous factors which possess similar sequence homology, but we hypothesized they may interact to FGFRs differently leading to distinct effects, particularly on granulosa cell growth and induce proliferation following a short period of exposition. This study was performed to investigate the effects of FGF8 and FGF18 on ovine granulosa cells proteome. Ovine ovaries were obtained from adult sheep’s irrespective of stage of estrous cycle and were cultured using a standard protocol. Granulosa cells were harvested from follicles then, seeded and cultivated. After, they were exposed to 10 ng/mL of FGF8 or FGF18. Cell proteins were extracted, cysteine bonds were reduced and acetylated and proteins were digested with trypsin. Tryptic peptides were analyzed using a bottom-up proteomic approach, mass spectrometry and a label-free quantitation method. The results obtained revealed following treatment with FGF8 or FGF18 for 30 minutes, an important shift toward upregulation for the entire granulosa cell proteome was measured. Additionally, several proteins, including ATF1, STAT3, MAPK1, MAPK3, MAPK14, PLCG1, PLCG2, PKCA, PIK3CA, RAF1, GAB1 and BAG2 were significantly upregulated (> 1.5-fold; p 1.5 fois ; p < 0.01). Tacitement, ces résultats suggèrent une activation de la voie MAPK/ERK cohérente avec ce qui est connu dans la littérature et nos hypothèses. De plus l’expression de deux facteurs de transcription, STAT3 et ATF1 ont été fortement augmentés. Ils sont directement impliqués dans la croissance, la prolifération et les mécanismes de survie cellulaire. Cette augmentation très significative de STAT3 et ATF1 pourrait avoir des conséquences importantes sur la viabilité de l’ovocyte

    Identifying new pain relief targets : using Caenorhabditis elegans to study the endocannabinoid system with advanced proteomics

    Ce projet de recherche examine la relation entre les canaux TRP et le systeme endocannabinoide chez Caenorhabditis elegans, dans le but de developper de nouvelles approches pour reduire la douleur animale et ameliorer leur bien-etre. Bien que des recepteurs structuraux des cannabinoides comme NPR-19 et NPR-32 aient ete identifies chez C. elegans, une caracterisation approfondie de ces recepteurs reste a faire. Nous proposons que les endocannabinoides de C. elegans agissent a la fois sur les recepteurs orthologues vanilloides et cannabinoides, mais seuls les recepteurs vanilloides seraient essentiels pour le soulagement de la douleur. Notre etude a utilise une combinaison d'interventions pharmacologiques, de tests d'evitement thermique et de profilage proteomique thermique pour evaluer les effets de l'AEA, de la capsaicine et du THC sur les souches sauvages et mutantes de C. elegans. Nos resultats indiquent une interaction complexe entre la signalisation des endocannabinoides et les canaux TRP dans la perception de la douleur chez C. elegans. L'AEA (? 10 ?M) a supprime la reponse a la douleur face a la chaleur nocive chez C. elegans apres une exposition d'une heure et a montre des effets analgesiques chez tous les mutants mentionnes, suggerant une redondance des recepteurs ou des mecanismes compensatoires. L'analyse des donnees proteomiques et bio-informatiques a revele une augmentation des processus de transport transmembranaire des cations, une diminution des activites metaboliques liees au stress, ainsi qu'une amelioration des voies de formation des ribosomes, de synthese proteique et d'exportation des proteines. L'AEA a significativement influence diverses voies, y compris l'homeostasie ionique, la signalisation calcique, la signalisation mTOR, le metabolisme du glutathion et les processus lies a l'apoptose. Grace au profilage proteomique thermique, 46 cibles potentielles de l'AEA ont ete identifiees, englobant des orthologues des recepteurs cannabinoides (NPR-32, NPR-19), un orthologue des recepteurs vanilloides (OCR-2), ainsi que des proteines supplementaires associees a la regulation de la nociception et des reponses inflammatoires.This research examines the relationship between TRP channels and the endocannabinoid system in Caenorhabditis elegans, aiming to develop new approaches for reducing animal pain and improving their well-being. Although structural cannabinoid receptors like NPR-19 and NPR-32 have been identified in C. elegans, a thorough characterization of these-receptors is still pending. We propose that C. elegans endocannabinoids would function-through both vanilloid and cannabinoid ortholog receptors, but only vanilloid receptors-would be essential for pain relief. Our study employed a combination of pharmacological interventions, thermal avoidance tests, and thermal proteome profiling to evaluate the impacts of AEA, capsaicin, and THC on wild-type and mutant C. elegans strains. Our results indicate a complex interplay between endocannabinoid signaling and TRP channels in C. elegans pain perception. AEA (? 10 ?M) suppressed the pain response to harmful heat in C. elegans after 1 hour exposure and showed pain-relieving effects in all the mutants mentioned, suggesting receptor redundancy or compensatory mechanisms. Analysis of proteomics and bioinformatics data revealed an increase in transmembrane transport processes involving cations, a decrease in specific stress-related metabolic-activities, and enhanced pathways for ribosome formation, protein synthesis, and protein export. AEA was observed to significantly influence various pathways, including ion-homeostasis, calcium signaling, mTOR signaling, glutathione metabolism, and apoptosis related processes. Through Thermal Proteome Profiling, 46 potential AEA targets were identified, encompassing cannabinoid receptor orthologs (NPR-32, NPR-19), a vanilloid-receptor ortholog (OCR-2), and additional proteins associated with nociception regulation and inflammatory responses

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Ce projet de recherche porte sur l’étude de la régulation des systèmes cannabinoïdes et vanilloïdes chez Caenorhabditis elegans (C. elegans), dans le but d’évaluer les effets antinociceptifs du tétrahydrocannabinol (THC) et du cannabidiol (CBD). C. elegans est un modèle largement utilisé pour étudier la nociception, visant principalement à caractériser les réponses nociceptives induites par le THC et le CBD, ainsi qu’à identifier les mécanismes et les cibles moléculaires impliqués. Les résultats des études sur l’utilisation du cannabis dans le traitement de la douleur chronique chez les mammifères sont controversés. Cette recherche vise à étudier l’effet du CBD et du THC sur la réponse nociceptive chez C. elegans et à approfondir la compréhension des mécanismes pharmacologiques sous-jacents. La méthodologie consiste à quantifier l’effet antinociceptif du CBD et du THC chez C. elegans par la méthode de la thermotaxie. Les nématodes sauvages (N2) étaient exposés à des concentrations croissantes de phytocannabinoïdes pour évaluer la relation concentration-effet. D’autres tests étaient effectués sur des souches mutantes exprimant des récepteurs cannabinoïdes et vanilloïdes afin d’identifier préalablement leurs cibles. Enfin, les analyses protéomiques et bioinformatiques seront effectuées pour identifier les voies de signalisation et les processus biologiques induits par l’interaction entre les phytocannabinoïdes et leurs cibles. Cette étude démontre l’activité antinociceptive du CBD et du THC chez C. elegans avec des effets rémanents pour THC, en ciblant respectivement le vanilloïde pour le CBD et le cannabinoïde pour les systèmes THC. Les analyses protéomiques et bio-informatiques mettent en évidence des différences significatives dans leurs voies de signalisation et leurs processus biologiques.The objective of this research project was to focus on studying the regulation of cannabinoid and vanilloid systems in Caenorhabditis elegans (C. elegans) to evaluate the anti-nociceptive effects of tetrahydrocannabinol (THC) and cannabidiol (CBD). C. elegans is a widely used model for studying nociception, with the main objective being to characterize nociceptive responses induced by THC and CBD, as well as identify the underlying molecular mechanisms and targets involved. Recent studies on the use of cannabis for the treatment of chronic pain in mammals have shown controversial results. This research aims to investigate the effect of CBD and THC on the nociceptive response in C. elegans and understand the underlying pharmacological mechanisms. The methodology consisted in quantifying the antinociceptive effect of CBD and THC in C. elegans using the thermotaxis method. WT(N2) were exposed to decreasing concentrations of phytocannabinoids to evaluate the dose and effect relationship. Further tests performed on mutant expressing cannabinoid and vanilloid receptors allowed preliminarily identification of their targets. Finally, proteomic and bioinformatics analyses were used to identify the signaling pathways and biological processes induced by these phytocannabinoids. The result of this study confirmed the antinociceptive effect of CBD and THC in C. elegans, with a remanent effect of THC. This effect is mediated by the vanilloid system for CBD and the cannabinoid system for THC, respectively. Also, proteomics and bioinformatics analyses revealed significant differences in signaling pathways and biological processes

    A systematic in vitro metabolism study of opioids using rat liver S9 fractions and mass spectrometry revealed CYP2D metabolism is impaired with age

    La codéine et l'oxycodone sont des opioïdes utilisés pour soulager la douleur. Le succès du traitement dépend du métabolisme par les enzymes CYP450. Selon les médicaments utilisés, l’altération de métabolisme peut entraîner de graves conséquences, y compris des modifications dans leurs profils d'efficacité, de sécurité et de toxicité. L'objectif de cette étude consistait à évaluer par HPLC-MS / MS (trappe ionique linéaire) l'effet de l'âge sur le métabolisme de l'enzyme CYP2D chez les rats. La codéine et l'oxycodone ainsi que leurs métabolites spécifiques, morphine (CYP2D), norcodeine (CYP3A), oxymorphone (CYP2D) et noroxycodone (CYP3A), ont été quantifiés en utilisant la méthode de dilution isotopique. Les fractions S9 de foie de rats (Sprague-Dawley) de sexe masculin âgés de 3, 6, 12 et 18 mois ont été préparées et les paramètres de Michaelis-Menten ont été déterminés pour les principales voies métaboliques. La vitesse maximale de la réaction enzymatique (Vmax) a montré une saturation rapide des sites actifs de CYP2D dans la fraction s9 de foie chez les rats âgés de 18 mois. Les valeurs de Vmax observées pour la codéine chez les rats âgés de 3, 6 et 12 mois étaient respectivement de 0,327 ± 0,027, 0,355 ± 0,021 et 0,273 ± 0,028. Concernant l'oxycodone, les valeurs étaient de 0,690 ± 0,096, 0,499 ± 0,087, 0,153 ± 0,01 respectivement. Cependant, la valeur de Vmax chez les rats âgés de 18 mois était de 0,092 ± 0,003 pour la codéine et de 0,153 ± 0,01 pour l’oxycodone. For 18 month-old rats, Vmax value was 0.092 ± 0.003 for codeine compared to 0.153 ± 0.01 for oxycodone. The Vmax value affected significantly (P ≤ 0.05) the enzyme-mediated clearance of these drugs (CLuint = Vmax / Km). The results of this study suggest that CYP2D enzyme undergoes a conformational change with age, particularly in 18 month-old rats considered to be geriatric in age, thus leading to a significant decrease in the rate of the formation of active opioid metabolites and a decrease in enzymatic clearance of these drugs. This study suggests that there is an impairment of CYP2D and CYP3A in aging rats, which affects their drug metabolism

    The role of protein convertases in bigdynorphin and dynorphin A metabolic pathway

    Les dynorphines sont des neuropeptides importants avec un rôle central dans la nociception et l’atténuation de la douleur. De nombreux mécanismes régulent les concentrations de dynorphine endogènes, y compris la protéolyse. Les Proprotéines convertases (PC) sont largement exprimées dans le système nerveux central et clivent spécifiquement le C-terminale de couple acides aminés basiques, ou un résidu basique unique. Le contrôle protéolytique des concentrations endogènes de Big Dynorphine (BDyn) et dynorphine A (Dyn A) a un effet important sur la perception de la douleur et le rôle de PC reste à être déterminée. L'objectif de cette étude était de décrypter le rôle de PC1 et PC2 dans le contrôle protéolytique de BDyn et Dyn A avec l'aide de fractions cellulaires de la moelle épinière de type sauvage (WT), PC1 -/+ et PC2 -/+ de souris et par la spectrométrie de masse. Nos résultats démontrent clairement que PC1 et PC2 sont impliquées dans la protéolyse de BDyn et Dyn A avec un rôle plus significatif pour PC1. Le traitement en C-terminal de BDyn génère des fragments peptidiques spécifiques incluant dynorphine 1-19, dynorphine 1-13, dynorphine 1-11 et dynorphine 1-7 et Dyn A génère les fragments dynorphine 1-13, dynorphine 1-11 et dynorphine 1-7. Ils sont tous des fragments de peptides associés à PC1 ou PC2. En plus, la protéolyse de BDyn conduit à la formation de Dyn A et Leu-Enk, deux peptides opioïdes importants. La vitesse de formation des deux est réduite de manière significative dans les fractions cellulaires de la moelle épinière de souris mutantes. En conséquence, l'inhibition même partielle de PC1 ou PC2 peut altérer le système opioïde endogène.Dynorphins are important neuropeptides with a central role in nociception and pain alleviation. Many mechanisms regulate endogenous dynorphin concentrations, including proteolysis. Proprotein convertases (PCs) are widely expressed in the central nervous system and specifically cleave at C-terminal of either a pair of basic amino acids, or a single basic residue. The proteolysis control of endogenous Big Dynorphin (BDyn) and Dynorphin A (Dyn A) levels has a profound impact on pain perception and the role of PCs remain unclear. The objective of this study was to decipher the role of PC1 and PC2 in the proteolysis control of BDyn and Dyn A levels using cellular fractions of spinal cords from wild type (WT), PC1-/+ and PC2-/+ animals and mass spectrometry. Our results clearly demonstrate that both PC1 and PC2 are involved in the proteolysis regulation of BDyn and Dyn A with a more important role for PC1. C-terminal processing of BDyn generates specific peptide fragments Dynorphin 1-19, Dynorphin 1-13, Dynorphin 1-11 and Dynorphin 1-7 and C-terminal processing of Dyn A generates Dynorphin 1-13, Dynorphin 1-11 and Dynorphin 1-7, all these peptide fragments are associated with PC1 or PC2 processing. Moreover, proteolysis of BDyn leads to the formation of Dyn A and Leu-Enk, two important opioid peptides. The rate of formation of both is significantly reduced in cellular fractions of spinal cord mutant mice. As a consequence, even partial inhibition of PC1 or PC2 may impair the endogenous opioid system
