Evidence for immune system alterations in peripheral biological fluids in Parkinson's disease
No full textImmune-related alterations in Parkinson’s disease (PD) can be monitored by assessing peripheral biological fluids
that show that specific inflammatory pathways contribute to a chronic pro-inflammatory status. This pro-
inflammatory activity is hypothesized to be already present in the prodromal stages of PD. These pathways
maintain and reinforce chronic neurodegeneration by stimulating cell activation and proliferation what triggers
the pro-inflammatory status as well. The gut microbiome possibly contributes to inflammatory pathways and
shows specific differences in fecal samples from PD compared to healthy controls. In PD, Bacteroides abundance
correlates with inflammatory markers in blood and motor impairment. Increased pro-inflammatory and
decreased anti-inflammatory bacterial colonization can lead to changes in the metabolic pathways of amino
acids, inducing increased membrane permeability, described as a leaky gut, enabling advanced contact between
immune cells and gut microbiome and potentially a spreading of neuroinflammation through the body via the
blood. Increased cytokine blood levels in PD are correlated with disease severity, motor symptoms, and clinical
phenotypes.
α-synuclein is a central player in PD-associated inflammation, inducing specific T-cell activity and triggering
microglial activation in the central nervous system (CNS). Misfolded α-synuclein propagation possibly results in
the spreading of aggregated α-synuclein from neuron to neuron leading to a sustained neuroinflammation. This is
supported by age-dependent defects of protein uptake in microglia and monocytes, so-called “inflammaging”,
including α-synuclein oligomers, as the key pathological protein in PD.
Genetic risk markers and inherited forms of PD are also associated with inflammation, which is highly relevant
for potential therapeutical targets.
The documented associations of inflammatory markers and clinical phenotypes indicate a pro-inflammatory
concept of specific PD pathophysiology here. An in-depth understanding of inflammatory mechanisms in PD
from bottom (gut) to top (CNS) and vice versa is needed to design novel immunomodulatory approaches to delay
or even stop PD. Future studies focusing on structured protocols in large patient cohorts with appropriate control
groups and comparative analysis among studies will aid the discovery of novel candidate biomarkersOpen-Access-Publikationsfonds 202

Automatic vocalisation-based detection of fragile X syndrome and Rett syndrome
No full textFragile X syndrome (FXS) and Rett syndrome (RTT) are developmental disorders currently not diagnosed before toddlerhood. Even though speech-language deficits are among the key symptoms of both conditions, little is known about infant vocalisation acoustics for an automatic earlier identification of affected individuals. To bridge this gap, we applied intelligent audio analysis methodology to a compact dataset of 4454 home-recorded vocalisations of 3 individuals with FXS and 3 individuals with RTT aged 6 to 11 months, as well as 6 age- and gender-matched typically developing controls (TD). On the basis of a standardised set of 88 acoustic features, we trained linear kernel support vector machines to evaluate the feasibility of automatic classification of (a) FXS vs TD, (b) RTT vs TD, (c) atypical development (FXS+RTT) vs TD, and (d) FXS vs RTT vs TD. In paradigms (a)–(c), all infants were correctly classified; in paradigm (d), 9 of 12 were so. Spectral/cepstral and energy-related features were most relevant for classification across all paradigms. Despite the small sample size, this study reveals new insights into early vocalisation characteristics in FXS and RTT, and provides technical underpinnings for a future earlier identification of affected individuals, enabling earlier intervention and family counselling

The reality of fiction: the ECO by Mathias Goeritz

This article covers the full biography of a building, the Experimental Museum El Eco, designed by Germanborn and Mexican émigré artist and architect Mathias Goeritz. It provides an approach intersecting the biography of the author, the history of the building, and prominent individuals of the two cultural traditions, German and Mexican, who participated in the creation of a very special and unique building: El Eco. On the one hand, the ethics of Expressionism, the interest in non-European art, the cult of primitivism and the aesthetic system of the pair of concepts abstraction-empathy, all stemming from German culture. On the other, the pantheistic religiosity of landscape, zoomorphism and anthropomorphism, the interest in masks, and the aesthetics of monumental scale, stemming from pre-Cortesian Mexican culture. Taking the stance of intertwining Mathias Goeritz parcours with those of individuals and issues from his German past and his Mexican future – highlighting the figures of Wilhelm Worringer, Paul Westheim, Luis Barragán, Edmundo O'Gorman, and Ida Rodríguez Prampolini – this article proposes a return trip from fiction to reality, following in the footsteps of the author and comparing them with the pathway of the very building El Eco
Oswald Mathias Ungers and Sol LeWitt: Variations
No full textOne of the fundamentals of Oswald Mathias Ungers work is the definition of a new unity between art and architecture. What this investigation is focused on is the identification of the intersection point between art and architecture, considering the project for the Entrance to the Kaiserthermen in Trier (20032007) as case study, revealing the system of theoretical, methodological and compositional correspondences. Plastic art is Ungers' main interlocutor: one of the author's most important references is Sol LeWitt (1928-2007), an artist he admired and collected. Setting the work of Oswald Mathias Unger and Sol LeWitt side by side, offers the opportunity to underline the shared interpretation that the two authors have of the world of forms postulated by Henri Focillon (1881-1943) in Life of Forms, according to which form, autonomous with respect to time and place, has its own universe, governed by its own laws, space and matter. According to these premises, we propose to read Trier's building through the instruments of art, following the hypothesis that in this project art embodies a creative and positive component in the definition of architecture, a cultural stimulus that guides the author in the design process
