Mecanismos de remodelado y calcificación cardiovascular: papel del receptor nuclear NOR-1 y la LOX
Programa de Doctorat en Biomedicina / Tesi realitzada a l'Institut d'Investigacions Biomèdiques de Barcelona (IIBB-CSIC)[spa] La calcificación cardiovascular (CCV) afecta a arterias y válvulas comprometiendo su normal funcionamiento y aumentando el riesgo de sufrir accidentes cardiovasculares y de desarrollar hipertrofia cardíaca e insuficiencia cardiaca, entre otros. Actualmente no existen tratamientos eficaces que prevengan o reduzcan la CCV. Nuestro grupo había descrito la sobreexpresión del receptor nuclear NOR-1 en lesiones ateroscleróticas humanas y su inducción en células endoteliales y células musculares lisas vasculares (CMLVs) por estímulos que promueven disfunción endotelial y potencian la migración y proliferación de ambos tipos celulares. La Lisil Oxidasa (LOX) es una enzima que cataliza el entrecruzamiento de las fibras de colágeno y elastina en el proceso de maduración de la matriz extracelular (MEX). Nuestro grupo había caracterizado la implicación de LOX en la disfunción endotelial y el remodelado vascular, y cómo la actividad LOX contribuye al estrés oxidativo y a la rigidez vascular. En el presente trabajo hemos profundizado en el estudio de los mecanismos celulares y moleculares por los que NOR-1 y LOX participan en la CCV. Detectamos aumento de los niveles de expresión de NOR-1 tanto en válvulas aórticas como en lesiones ateroscleróticas humanas calcificadas, en las que colocalizó con marcadores de calcificación como RUNX2 (Runt-related transcription factor 2) y osteopontina (OPN). La expresión endógena de NOR-1 aumentó al incubar células instersticiales de válvula aórtica (VICs) o CMLVs humanas con un medio con alto contenido en fosfato que indujo calcificación. La sobreexpresión de NOR-1 en VICs expuestas a éste medio limitó la inducción de los marcadores de diferenciación osteoblástica/calcificación y la mineralización de la MEX. Además, al exponer a medio osteogénico CMLVs o anillos aórticos de ratones que sobreexpresan NOR-1 en las CMLVs (TgNOR-1CMLV) la expresión de los marcadores de calcificación y la mineralización de la MEX fue menor que en células o anillos aórticos de animales control. Finalmente, al inducir hiperlipidemia y aterosclerosis experimental (mediante la administración de AAV-PCSK9D374Y y dieta aterogénica) observamos que la transgénesis de NOR-1 disminuye el desarrollo de aterosclerosis a nivel de la aorta y la calcificación de las lesiones ateroscleróticas. Las lesiones ateroscleróticas de animales TgNOR-1CMLV eran de menor tamaño y tenían menor infiltrado inflamatorio; además, estaban menos calcificadas y presentaban menor expresión de marcadores osteogénicos, menor grado de entrecruzamiento del colágeno y menor contenido de glucosaminoglucanos que las de los animales control. Por lo tanto, NOR-1 reduce la calcificación de células en cultivo y la calcificación de la íntima asociada a la aterosclerosis. En relación al impacto de LOX en la CCV, observamos una mayor expresión de esta enzima tanto en válvulas aórticas calcificadas como en placas ateroscleróticas humanas en las que colocalizaba con RUNX2. Al inducir la calcificación de VICs con un medio rico en fosfato observamos un aumento de la secreción de la LOX que promovió la maduración de la MEX y su mineralización de forma dependiente de su actividad catalítica. Al inducir aterosclerosis mediante la administración de AAV-PCSK9D374Y y dieta aterogénica, los ratones que sobreexpresan LOX en las CMLVs de la pared vascular (TgLOXCMLV) presentaron mayor desarrollo de aterosclerosis en la aorta y mayor calcificación de las lesiones. Las lesiones ateroscleróticas de ratones TgLOXCMLV tenían más infiltrado inflamatorio y una cubierta fibrosa más rica en CMLVs y más engrosada. El análisis de la calcificación evidenció que las placas de ratones TgLOXCMLV estaban más calcificadas y presentaban mayor expresión de marcadores osteogénicos, mayor grado de entrecruzamiento de colágeno y mayor contenido de glucosaminoglucanos que las de los animales control. Por lo tanto, nuestro estudio proporciona evidencias tanto in vitro como in vivo del papel clave de la LOX en la CCV.[eng] Background: Cardiovascular calcification (CVC) is a global health concern whose underlying mechanisms are not fully understood and for which there are no effective therapies. We described NOR-1 expression in human atherosclerotic lesions and its induction by atherogenic stimuli; however, its contribution to CVC is unknown. We have also outlined the potential contribution of LOX, an enzyme involved in matrix remodeling, in biomineralization of vascular smooth muscle cells (VSMCs), but it is uncertain whether LOX could participate in valvular calcification and in atherosclerosis-related mineralization. Methods: Human samples of calcific aortic valves and atherosclerotic lesions, as well as valvular interstitial cells (VICs) and VSMCs exposed to high phosphate medium were analyzed. To induce atherosclerosis and calcification, transgenic mice overexpressing NOR-1 or LOX in VSMCs (TgNOR-1VSMC or TgLOXVSMC) were transduced with AAV-PCSK9D374Y and fed a high fat/high cholesterol diet. Results: Higher levels of NOR-1, as well as osteogenic markers (RUNX2 and osteopontin), were detected in human calcified aortic valves and in atherosclerotic lesions in which NOR-1 colocalized RUNX2. NOR-1 expression increased when calcification was induced in human cells (VICs and VSMCs) or mouse VSMCs. Gain- and loss-of-function approaches determined that NOR-1 modulates genes involved in osteoblastic transdifferentiation and extracellular matrix (ECM) mineralization. VSMCs and aortic rings from TgNOR-1VSMC mice exposed to osteogenic media exhibited reduced expression of osteogenic markers and decreased mineralization. NOR-1 transgenesis decreased atherosclerosis and intimal calcification. Lesions from transgenic mice had lower expression of osteogenic markers, lower degree of collagen crosslinking and lower content of glycosaminoglycans. We also observed increased LOX expression in both human calcified aortic valves and atherosclerotic plaques in which LOX colocalized with RUNX2. The induction of calcification in VICs increased LOX secretion which promoted ECM maturation and its mineralization in a manner dependent on its catalytic activity. LOX transgenesis promoted aortic atherosclerosis and intimal calcification. Lesions showed greater inflammatory content and thicker fibrous cap. Plaques from TgLOXVSMC also exhibited greater content of osteogenic markers, cross-linked collagen and glycosaminoglycans.

Estudio de posibles alteraciones a nivel hipotalámico en ratas alimentadas con una dieta desequilibrada rica en grasa
La ingesta de dietas ricas en grasa se asocia a la aparición de obesidad y a toda una serie de
complicaciones metabólicas relacionadas que afectan a la salud. Además, recientemente se ha
relacionado la ingesta de dietas hiperlipídicas y la obesidad con una mayor disfunción cognitiva y un
mayor riesgo a desarrollar enfermedades neurodegenerativas. La ingesta de dietas hiperlipídicas pero
controladas en calorías va ligada a la aparición del fenotipo “falso delgado” o “metabolically obese
normal-weight” (MONW). Estos individuos no presentan obesidad, pero sí un mayor acúmulo de
grasa visceral y alteraciones metabólicas características del estado obeso, como la deposición de grasa
en el hígado. Estudios previos del grupo de investigación donde se ha realizado este trabajo han
demostrado que el fenotipo falso delgado, en roedores, va asociado a alteraciones cognitivas que se
manifiestan a nivel de cambios comportamentales (mediante la realización de un test de memoria) y de
cambios en la expresión génica en el hipocampo, una región cerebral muy importante en el
procesamiento de la memoria. El hipotálamo es otra región cerebral que tiene un papel clave en la
aparición de alteraciones cognitivas. Por este motivo, en este trabajo se ha estudiado si el fenotipo
falso delgado asociado a la ingesta isocalórica de dietas hiperlipídicas también produce alteraciones a
nivel hipotalámico, centrándonos en aquellas que pudieran estar relacionadas con la alteración
cognitiva. Para ello se utilizaron muestras de hipotálamo de animales alimentados con dietas
hiperlipídicas (con un 45% -HF45- o un 60% -HF60- de la energía en forma de grasa), administradas
en condiciones isocalóricas a un grupo control. En primer lugar, se contribuyó a la caracterización de
los animales mediante la cuantificación de la grasa hepática, y se pudo observar como los animales
alimentados con dieta hiperlipídica, pese a ser normopeso, presentaban una mayor acumulación de
grasa hepática, lo cual es característico del fenotipo MONW. Por otra parte, se llevó a cabo una
búsqueda bibliográfica para seleccionar genes clave relacionados con la inflamación, la cognición y el
aprendizaje, y se estudió su expresión en hipotálamo de los animales control, HF45 y HF60. A
diferencia de lo que se había observado previamente en el hipocampo de los mismos animales, no se
observaron alteraciones en la expresión de los genes inflamatorios estudiados (Tnf-α e Il-1β) que
pudieran indicar un estado de inflamación tal como el que se ha descrito en hipotálamo cuando las
dietas grasas se administran “ad libitum” y que, a su vez, se relaciona con la alteración cognitiva. Sin
embargo sí que se observó alteración en la expresión de algunos genes relacionados con la cognición.
El resultado más destacable fue una disminución en la expresión de Bdnf, que codifica para un
protector neuronal (factor neurotrófico), en hipotálamo de animales MONW. Por otra parte, se observó
una tendencia a una menor expresión de los genes Psen1 y Psen2 y a una mayor expresión de Gfap, se
trata de genes clave en el desarrollo neuronal y en el mantenimiento de la homeostasis de células
cerebrales, por lo que este patrón de expresión génica se puede relacionar con la mayor susceptibilidad
a la disfunción cognitiva que presentan estos animale
Upregulation of NOR-1 in calcified human vascular tissues: impact on osteogenic differentiation and calcification
Cardiovascular calcification is a significant public health issue whose pathophysiology is not fully understood. NOR-1 regulates critical processes in cardiovascular remodeling, but its contribution to ectopic calcification is unknown. NOR-1 was overexpressed in human calcific aortic valves and calcified atherosclerotic lesions colocalizing with RUNX2, a factor essential for osteochondrogenic differentiation and calcification. NOR-1 and osteogenic markers were upregulated in calcifying human valvular interstitial cells (VICs) and human vascular smooth muscle cells (VSMCs). Gain- and loss-of-function approaches demonstrated that NOR-1 negatively modulates the expression of osteogenic genes relevant for the osteogenic transdifferentiation (RUNX2, IL-6, BMP2, and ALPL) and calcification of VICs. VSMCs from transgenic mice overexpressing NOR-1 in these cells (TgNOR-1VSMC) expressed lower basal levels of osteogenic genes (IL-6, BMP2, ALPL, OPN) than cells from WT littermates, and their upregulation by a high-phosphate osteogenic medium (OM) was completely prevented by NOR-1 transgenesis. Consistently, this was associated with a dramatic reduction in the calcification of both transgenic VSMCs and aortic rings from TgNOR-1VSMC mice exposed to OM. Atherosclerosis and calcification were induce in mice by the administration of AAV-PCSK9D374Y and a high-fat/high-cholesterol diet. Challenged-TgNOR-1VSMC mice exhibited decreased vascular expression of osteogenic markers, and both less atherosclerotic burden (assessed in whole aorta and lesion size in aortic arch and brachiocephalic artery) and less vascular calcification (assessed either by near-infrared fluorescence imaging or histological analysis) than WT mice. Our data indicate that NOR-1 negatively modulates the expression of genes critically involved in the osteogenic differentiation of VICs and VSMCs, thereby restraining ectopic cardiovascular calcification.This work was supported by the Spanish Ministerio de Ciencia e Innovación (

Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases.
