Liberalism, the Novel, & the Self: Lionel Trilling on the Political Functions of Literature
Lionel Trilling was a literary critic, not a political theorist, but he was very much concerned with the political effects of novels. This article explores Trilling\u27s understanding of politics and the novel and, in particular, his view that novels can strengthen liberal society by presenting conflicting ideas in a way that encourages critical thinking and a morally stronger individual. Focusing on the political movements of the 1950s and 1960s, this book argues that the arts can strengthen democracy by politically educating citizens. Previous research suggests metabolic rate as a potential proximate mechanism to explain behavioural covariation. We measured the resting metabolic rate (RMR), boldness and exploration in western stutter-trilling crickets, Gryllus integer, selected differentially for short and fast development over two generations. After applying mixed-effects models to reveal the sign of the covariation, we applied structural equation models to an individual-level covariance matrix to examine whether the RMR generates covariation between the measured behaviours. All traits showed among-individual variation and covariation: RMR and boldness were positively correlated, RMR and exploration were negatively correlated, and boldness and exploration were negatively correlated. However, the RMR was not a causal factor generating covariation between boldness and exploration. Instead, the covariation between all three traits was explained by another, unmeasured mechanism. The selection lines differed from each other in all measured traits and significantly affected the covariance matrix structure between the traits, suggesting that there is a genetic component in the trait integration. A comparative molecular analysis of viral DDB1-binding proteins in the context of their immune evasive properties
Für Viren ist es von entscheidender Bedeutung, den Restriktionsfaktoren der Wirtszelle entgegenzuwirken, um eine effiziente Replikation zu ermöglichen. Zahlreiche Viren kodieren für akzessorische Proteine, mit denen sie das zelluläre DNA damage-binding Protein 1 (DDB1) zweckentfremden und assoziierte Cullin Ubiquitin-Ligase-Komplexe zum proteasomalen Abbau von antiviralen Restriktionsfaktoren immunevasiv ausnutzen. Das Ziel dieser Arbeit war es, die Funktionen verschiedener viraler DDB1-interagierender Proteine, deren Zielproteine sowie die zweckentfremdeten Komplexe zu untersuchen. Dazu wurden Immunpräzipitationsstudien durchgeführt und Interaktionspartner mittels Massenspektrometrie identifiziert und vergleichend quantifiziert. Im Fokus der Untersuchung standen dabei das MCMV-kodierte Protein pM27, dessen homologe Proteine pE27, pR27 (beide RCMV) und pUL27 (HCMV) sowie die viralen Proteine Vpr (HIV-2), Vpx (HIV-2), das HBV-kodierte Protein HBx und dessen Woodchuck Hepatitisvirus-Homolog WHx. Als Negativkontrollen bzw. Außengruppe dienten EGFP und das HCMV-kodierte Protein pUL42, welches mit einer anderen Ubiquitin-Ligase (Itch) interagiert. Die Analyse des pM27-Interaktionsspektrums bekräftigte das gegenwärtige immunevasive Funktionsmodel von pM27, da sowohl DDB1, Cullin 4A und STAT2 vorzufinden waren. Eine weiterführende Analyse der Interaktion zwischen pM27 und verschiedenen N- und C-terminalen Verkürzungsmutanten von STAT2 zeigte, dass sowohl N- als auch C-terminale Bereiche von STAT2 mit pM27 kopräzipitieren. Des Weiteren wurde auch Xpo7 in dem pM27- Interaktionsspektrum gefunden und Cul4B und Ncoa5 als neue Interaktionspartner identifiziert. Ncoa5 und Xpo7 wurden unter Infektionsbedingungen validiert. Zusätzlich konnten zahlreiche beschriebene Interaktionen bestätigt und teilweise auf homologe und/oder paraloge Proteine übertragen werden. So wurde die bekannte Interaktion von HBx mit Spindlin-1 auf die viralen Homologe WHx7 und WHx8 und die zellulären Paraloge Spindlin-2, und -3 ausgeweitet. Eine vermutete, aber nicht experimentell bestätigte Interaktion von Vpr mit Bax konnte dokumentiert werden. Der Vergleich der Interaktionsspektren enthüllte überdies große Unterschiede sowohl in der generellen Interaktion als auch der Interaktionsstärke der viralen Proteine mit DDB1 und Cullin 4A/B. Zwar korrelierten die Interaktionsspektren in hohem Maße mit der phylogenetischen Verwandtschaft, jedoch zeichnete sich auch eine Überschneidung der DDB1-interagierenden Proteine verschiedener Virusfamilien ab. Für das HIV-2-kodierte Protein Vpx wurde der NF-κB-Transkriptionsfaktor p65/RelA als neuer Interaktionspartner identifiziert und diese Interaktion auf ihre funktionelle Relevanz hin untersucht. Dabei zeigte sich eine potente Inhibition des NF-κB-Signalwegs durch Vpx. Des Weiteren zeigte sich, dass eine Mutation von Vpx, welche die DDB1 Interaktion destabilisiert, diesen NF-κB-Antagonismus beeinträchtigte.
Zusammengenommen konnte in dieser Arbeit die Interaktionsspektren von neun verschiedenen viralen akzessorischen Proteinen identifiziert und verglichen werden, wobei zahlreiche bekannte Interaktionen die Validität dieser Analyse unterstrichen und die Bestimmung der Funktion der Interaktion zwischen Vpx und p65/RelA die Relevanz der neu gefundenen Interaktionspartner verdeutlicht.To allow efficient viral replication, viruses have to overcome cellular restriction factors. Several viruses encode accessory proteins which exploit DDB1-containing Cullin ubiquitin ligase complexes as essential co-factors to mediate proteasomal degradation of antiviral restriction factors. To establish a comprehensive understanding of the function of different viral DDB1-binding proteins, their targets and the complexes assembled by these viral proteins, immuno-affinity precipitations (IP) were performed and the retrieved proteins were identified and quantified by mass spectrometry. The interactomes of the MCMV-encoded protein pM27 and its homologous proteins pE27, pR27 (both RCMV-encoded) and pUL27 (HCMV-encoded) as well as the viral proteins Vpr (HIV-2-encoded), and Vpx (HIV-2-encoded), HBx (HBV-encoded) and its woodchuck hepatitis virus homolog WHx were identified, analysed and compared. Empty plasmids and the HCMV-encoded protein pUL42, which binds the unrelated ubiquitin ligase Itch, served as negative controls and outgroup, respectively. The analysis of the pM27 interactome underlined the current model of pM27 function, since DDB1, Cullin 4A, as well as STAT2 were co-purified with pM27. The interaction between pM27 and STAT2 was further investigated by using a panel of N- and C-terminal STAT2 truncation mutants. The analysis revealed that pM27 specifically co-precipitates both N- and C-terminal fragments of STAT2. Additionally, also Xpo7 was retrieved with pM27 and Cul4B and Ncoa5 were identified as new interaction partners of pM27. The interactions with Xpo7 and Ncoa5 were validated under authentic infectious conditions. The analysis of the viral interactomes further confirmed previously described interactions and extended several known interactions to homologous and paralogous proteins. For example, the known interaction between HBx and Spindlin-1 was extended to the viral homologous proteins WHx7 and WHx8 and to the cellular paralogous proteins Spindlin-2 and -3. An anticipated interaction between Vpr and Bax, which was experimentally not proven so far, could be documented for the first time. The comparison revealed considerable differences concerning the interaction with DDB1 and Cullin 4A/B in terms of the general ability as well as the strength of interaction. On a global level, the correlation of the interactomes fitted well with the phylogenetic relationship of the proteins but also highlighted a considerable overlap between the DDB1 interactors of non-related viruses. As a proof of principle, the biological relevance of one newly identified interaction was further investigated. The NF-κB transcription factor p65/RelA co-precipitated with the HIV-2-encoded protein Vpx. This interaction resulted in an impaired NF-κB signaling. Additionally, a Vpx mutant which lacks the ability to interact with DDB1 was impaired in its ability to antagonize the NF-κB signaling.
Taken together, the interactions of nine different viral accessory proteins were identified and compared. The confirmation of numerous previously described interactions underlined the validity of this analysis and the determination of the biological function of the interaction between Vpx and p65/RelA highlighted the relevance of the newly found interaction partners
