    Cytogenetic findings on shoe workers exposed long-term to benzene.

    Cytogenetic analysis of peripheral blood lymphocytes was performed to detect cytogenetical alterations in 58 shoe workers (57 male and 1 female) who had been exposed to particular mutagenic or carcinogenic agents and in 20 subjects selected from the general population as a control group. Frequencies of damaged cells, including gaps, breaks, and rearrangements (acentric fragment, deletion, translocations) were scored for both groups. The incidence of chromosomal aberrations (particularly chromatid gaps and breaks) in the study group was significantly higher than in the control group. No effects of smoking were observed and breaks alone were found to be influenced by alcohol consumption. No significant correlation was detected between the working period in the group exposed to benzene and frequency of chromosomal aberrations. Benzene content was determined to be between 0 and 28.5% in eight kinds of glues studied by fractional distillation. Hexane content ranged between 0 and 68.35% using the same method. This study indicated that the content of benzene and hexane in the glues are above normal limits

    Investigation of APC mutations in a Turkish familial adenomatous polyposis family by heterodublex analysis

    PURPOSE: Familial adenomatous polyposis is an autosomal dominant disease characterized by the presence of 100 or more colorectal adenomatous polyps. Mutations in the adenomatous polyposis coli gene are primarily responsible for the development of this disease. This study was designed to investigation of adenomatous polyposis coli (APC) gene mutations in members of familial adenomatous polyposis family to identify individuals at risk of the disease. METHODS: We examined one patient with familial adenomatous polyposis and 21 family members including one affected person from familial adenomatous polyposis and 20 nonsymptomatic persons. We studied E, D, F, and G segments of exon 15 of the adenomatous polyposis coli gene by heteroduplex analysis. RESULTS: We used silver staining method for staining. We found a mutation for five persons at segment F of exon 15 of the adenomatous polyposis coli gene. Two of them were affected by colorectal cancer, one of whom was the proband, and the other three were non-symptomatic family members. The pathogenetic mutation was a T deletion at codon 1172, causing a frameshift in the adenomatous polyposis coli gene, as a result of the sequencing analysis of these cases. CONCLUSIONS: Investigation of adenomatous polyposis coli gene mutations is very important for the identification of genetic susceptibility to colorectal cancer and for the definition of tumor developing at an early stage. Furthermore, the identification of this mutation for the first time in a Turkish family will be useful to foster further studies on familial adenomatous polyposis in Turkey

    Familial adenomatous polyposis coli (FAP) is an autosomal dominant disease characterised by the presence of 100 or more colorectal adenomatous polyps. Our proband case who have a retinal lesions (congenital hypertrophy of the retinal pigment epithelium,called CHRPE) and hundreds adenomatous polyps on all colon and rectum is a 36 years old woman. We isolated DNA from pheripheral blood samples of proband and her family members by proteinaz K incubation and phenol-chloroform extraction. We studied E,D, F,and G segments of exon 15 of APC gene by heterodublex analyses (HDA). For staining, we used non-radioactive silver staining method. We determined mutation in 5 person from this family in segmentF of exon 15 of APC. Two of them were patients with FAP (one is ourproband case) and another three persons were non semptomatic family members.     Giriş ve Amaç: SARS COV-2 virüsü, CD4+ ve CD8+ T hücrelerini ve B hücrelerini etkileyerek bağışıklığı aktive etmektedir. CD45RA T hücreleri, antijenleri hatırlamaması ile \"naive\" özelliklere sahipken CD45RO T hücreleri, antijenleri hatırlayıp çoğaldıkları için \"bellek\" T hücreleri olarak kabul edilir. Peptivator SARS-CoV-2 Select, SARS-CoV-2 spesifik CD4+ ve CD8+ T hücrelerini in vitro uyararak efektör sitokinlerin salgılanmasına sebep olup SARS-CoV-2 spesifik T hücrelerinin saptanmasına ve izolasyonuna olanak tanır. Bu çalışmadaki amacımız SARS-COV-2 hastalarının kanından elde edilen lenfositlerin peptivatör ile etkileşimi sonucu hafıza hücrelerinin incelenmesidir. Gereç ve Yöntem: Araştırmamız deneysel tiptedir. Marmara Üniversitesi Pendik Eğitim ve Araştırma Hastanesi Göğüs Hastalıkları polikliniğinde yatan Koronavirüs hastaları, araştırmanın evrenini oluşturmaktadır. T.C. Sağlık Bakanlığı Marmara Üniversitesi Pendik Eğitim ve Araştırma Hastanesi Göğüs Hastalıkları polikliniğinde Koronavirüs hastalığı sebebi ile yatan aşılı ve aşısız, PCR pozitif 18- 50 yaş arası 9 hastadan gönüllü olmaları kaydıyla kan örnekleri toplanmıştır. Hastalardan alınan periferik kanların bir kısmı ile tam kan immünfenotipleme analizi yapılmıştır. Hastalardan alınan periferik kanın bir kısmı ile de mononükleer hücre (PKMH) izolasyonu yapılıp uyarımsız (US), CDmix uyarımlı ve peptivatör ile uyarımı sonrasında akım sitometrisi kullanılarak analizi yapılmıştır. Bulgular: Flow sitometri tam kan immünfenotipleme analizlerinde CD45RO+ oranı aşısız olan grupta 93,6 (±6,76), aşılı grupta 94,2 (±6,07) olarak bulunmuştur. Hastalardan elde edilen PKMH’ler; US, CDmix ve Peptivatör ile kültür edildikten sonra flow sitometri analizlerinde US kültürde CD45RO+FoxP3 oranı aşısız grupta 8,29 (±4,62), aşılı grupta 3,5 (±2,19) olarak bulunmuştur. CDmix kültüründe CD45RO+FoxP3 oranı aşısız grupta 32,6 (±32,4), aşılı grupta 18,8 (±18,6) bulunmuştur. Peptivatör kültüründe CD45RO+Foxp3 oranı aşısız grupta 36,2 (±35,5), aşılı grupta ise 7,24 (±5,94) olarak bulunmuştur. Sonuç: COVID-19’da mRNA teknolojisi ile üretilen BioNTech/Pfizer aşısı enfeksiyon geçiren hastalarda hafıza hücre sayılarını aşısızlara göre arttırmaktadır. Peptivatör etkileşimi ise aşılı PCR+ grupta ve aşısız PCR+ grupta immün sistemi etkilediği ve hafıza hücrelerini arttırdığı gözlenmektedir.Introduction and Aim: SARS COV-2 virus activates immunity by affecting CD4+ and CD8+ T cells and B cells. Marmara University Pendik Training and Research Hospital Chest Diseases Coronavirus patients hospitalized in the outpatient clinic constitute the universe of the research. T.R. Ministry of Health Marmara University Pendik Vaccinated and unvaccinated, PCR positive 18-Blood samples were collected from 9 patients aged 50 years, provided that they were volunteers. Whole blood with some of the peripheral blood taken from the patients immunophenotyping analysis was performed. Mononuclear cell (PKMH) isolation was performed with some of the peripheral blood taken from the patients. Analysis was performed using flow cytometry after stimulation with no stimulation (US), CDmix stimulation and peptivator. Results: In flow cytometry whole blood immunophenotyping analysis, the CD45RO+ ratio was 93.6 (±6.76) in the unvaccinated group and 94.2 in the vaccinated group.(±6.07). PCMHs obtained from patients; Flow cytometry after culturing with US, CDmix and Peptivator In their analysis, the CD45RO+FoxP3 ratio in US culture was found to be 8.29 (±4.62) in the unvaccinated group and 3.5 (±2.19) in the vaccinated group.The CD45RO+FoxP3 ratio in the CDmix culture was 32.6 (±32.4) in the unvaccinated group and 18.8 (±18.6) in the vaccinated group. In peptivator culture The CD45RO+Foxp3 ratio was found to be 36.2 (±35.5) in the unvaccinated group and 7.24 (±5.94) in the vaccinated group.Conclusion: BioNTech/Pfizer vaccine produced with mRNA technology in COVID-19 decreased the number of memory cells in infected patients. increases compared to the unvaccinated. Peptivator interaction, on the other hand, affects the immune system in the vaccinated PCR+ group and the unvaccinated PCR+ group, and it affects the memory. observed to increase the number of cells

    Robust decision making for future train maintenance: Illustrating the abilities of decision making under deep uncertainty within the train maintenance context

    This research has illustrated the abilities of incorporating robust decision making within train maintenance. Both scheduled and unscheduled maintenance processes of the Nederlandse Spoorwegen (NS) have been examined, after which exploratory modeling and analysis enabled developing valuable insights for decision makers. Scenario analysis through simulation allowed for the quantification of the effect of deep uncertainty in relation to train maintenance. From the scenario analysis it is discovered what uncertainties are, according to the simulation model, of significant influence towards maintenance performance. The results presented allow decision makers to support their decisions when evaluating which type of policies to perform within the next decade. While the outcomes are specific for NS, the aim is to show that the approach held in this research is also applicable throughout other fields where decision making information quality should be improved.Engineering and Policy Analysi

Başlatıcının –OH fonksiyonalitesi ε-kaprplaktonun yaşayan karakterli halka açılmaso polimerizasyonunda kullanılmıştır. 115 0C’de SnOct2 katalizi kullanılarak (PDI<1.1) düşük molekül ağırlığı dağılımına sahip kontrollü moleküler ağırlıklı ve –OH son gruplu polimerler elde edilmiştir. Tempo fonksiyonalitesi içeren PCL stirenin radikalik olarak polimerleştirilmesinde makrobaşlatıcı olarak kullanılır. Bu reaksiyon poli(CL-b-St) dibloğu vermek üzere 125 0C’de gerçekleştirilir. Son olarak elde edilen diblok kopolimer , CuBr/PMDETA katalizi kullanılarak t-BA’ın ATRP’ da makrobaşlatıcı olarak kullanılır. Reaksiyon 100 0C’de gerçekleştirilmiştir veABC tipli poli(CL-b-St-b-tBA) miktoarm yıldız polimerleri düşük molekül ağırlığında (PDI<1.45) ve kontrollü molekül ağırlığında ROP-SFRP-ATRP sırası izlenerek sentezlenmiştir. Yeni elde edilen polimerler 1H-NMR ve jel geçirgenlik kromatografisiyle (GPC) karakterize edilmiştir.First of all a tri functional initiator 2- phenyl -2-[2,2,6,6-tetramethyl piperidinyl)oxy]ethyl-3-[(2-bromo-2-methyl propanoyl)oxy)-2-(hydroxy methyl)-2-methyl propanoate in a six-step reaction sequence. A hydroxyl functional core was used as an initiator for the “living” ring opening polymerization (ROP) of ε-caprolactone producing a hydroxyl terminatedpoolymer with controlled molecular weight and narrow polydispersity (PDI<1.1) using SnOct2 catalyst in bulk at 115 0C. Tempo-supported PCL behaved as a polymaric counter radical for the radival polymerization of styren , giving poly(CL-b-St) in quantitative efficiency in bulk at 125 0C. Finallly this obtained diblock copolymer used as a macroinitiator for ATRP of t-BA using CuBr/PMDETA as a catalyst at 100 0C to afford ABC type poly(CL-b-St-b-t-BA) mictoarm star polymers with controlled molecular weight and low polydispesity (Mw/Mn<1.45) via ROP-SFRP-ATRP sequence.     DAPSONE AS AN ALTERNATIVE THERAPY IN CHILDREN WITH FAMILIAL MEDITERRANEAN FEVER

    Objective: Familial Mediterranan Fever is an hereditary autoinflammatory disease that presents with recurrent febrile attacks and poly serositis. Colchicine is the only known treatment in this diease. However, nearly 5-10% of patients are resistant to colchicines. There are many different modalities in colchicine resistant patients, biologic and immunosupressive drugs being the known ones. We studied the efficacy of Dapsone as an anti inflammatory drug in children with FMF who did not tolerate colchicine well. Methods: This is a case series study in 10 patients who had FMF on the base of Tel-Hashomer criteria and did not tolerate colchicine or did not respond to it well. Patients took 2mg/kg dapsone in single dose, during 6 months. Findings In four patients episodic attacks returned after 27 days, so the drug was discontinued. One patient refused to continue the study; in five patients dapsone was taken in average for 8 months and 6 days, at least for 6 months. These five patients had no episodes of attack during the following observation. Conclusion: Dapsone could control episodic attacks of FMF in 50% of cases. It might be considered as an alternative therapy in FMF cases not responding to colchicine

    Effects of laser ablated silver nanoparticles on Lemna minor

    The present study investigates and models the effect of laser ablated silver nanoparticles (AgNPs) on the development of the aquatic macrophyte Lemna minor. Toxic effects of five different AgNP concentrations (8, 16, 32, 96 and 128 mu g L-1) on L minor were recorded over seven days under simulated natural conditions. Biosorption of AgNPs by L minor was modeled using four sorption isotherms, and the sorption behavior was found to agree most closely with the Langmuir-Freundlich model (R-2 = 0.997). While toxic effects of AgNPs could be observed in all models and concentrations, the greatest increase in toxicity was in the 8-32 mu g L-1 range. Dry weight- and frond number-based inhibition experiments suggest that growth inhibition does not necessarily scale with AgNP concentration, and that slight fluctuations in inhibition rates exist over certain concentration ranges. Very close fits (R-2 = 0.999) were obtained for all removal models, suggesting that the fluctuations are not caused by experimental variation. In addition, L minor was found to be a successful bioremediation agent for AgNPs, and displayed higher removal rates for increasing AgNP doses. FT-IR spectroscopy suggests that carbonyl groups are involved in AgNP remediation. (C) 2014 Elsevier Ltd. All rights reserved

    Physicochemical characterization of poly(tert-butyl acrylate-b-methyl methacrylate) prepared with atom transfer radical polymerization by inverse gas chromatography

    Poly(tert-butyl acrylate) (PtBuA) was synthesized by atom transfer radical polymerization (ATRP) using methyl-2-bromo propionate (MBP) as an initiator in bulk at 80 degrees C. The successive ATRP of methyl methacrylate in diphenyl ether at 80 degrees C using previously obtained PtBuA as a macroinitiator led to formation of poly(tert-butyl acrylate-b-methyl methacrylate) (poly(tBuA-b-MMA)). The synthesized macroinitiator and block copolymer have controlled molecular weight and low polydispersity (M-w/M-n < 1.2). The block copolymer was characterized by gel permeation chromatography (GPC) and H-1 NMR. The retention diagrams of poly (tBuA-b-MMA) for some aliphatic esters and aromatic hydrocarbons were obtained using inverse gas chromatography (IGC) technique. The glass transition temperatures, T(g)s of poly(tBuA-b-MMA) were determined by both differential scanning calorimeter (DSC) and IGC. It was observed that the block copolymer represents three TO at 50, 75 and 100 degrees C by IGC although it represents only one T, at 71 degrees C by DSC. After the column was quenched from 180 to 0 degrees C, the T-g at 100 degrees C shifted to 105 degrees C however others did not change. Specific retention volumes, V-g(0) and the thermodynamical polymer-solvent interaction parameters such as Flory-Huggins, chi(infinity)(12), equation-of-state, chi(*)(12) and effective exchange energy, X-eff were found for all studied solvents. Partial molar heat of sorption, Delta(H) over bar (1,sorp), partial molar heat of mixing, Delta(H) over bar (infinity)(1) and molar heat of vaporization, Delta H-v, were determined. In addition, the solubility parameter of the corresponding block copolymer, 62 was determined as 11.0 (cal/cm(3))(1/2) at 25 degrees C. (c) 2005 Elsevier Ltd. All rights reserved
