The trombone as portrayed in Portuguese iconography during the sixteenth and early seventeenth centuries
This article studies eleven sixteenth and seventeenth century iconographical sources depicting slide brass instruments, by both Portuguese and foreign artists active in Portugal, as well as foreign artists depicting Portuguese scenes. This study addresses questions concerning aspects of trombone technique that have not previously been considered and may have implications in the way the trombone was understood elsewhere in Europe. It focuses on aspects of technique depicted that may be representative of the trombone’s contemporary design and performance and therefore the manner in which the instrument was held and indeed played. Finally, this article suggests a transitional technical period when the way of holding the single-slide trumpet may have been used to play the trombone.Publisher PD

Interview to Rui Furtado, AFA Consult foundator
This research addresses a series of projects built by the engineering company "AFA Consult" through the testimony of one of its engineers, Rui Furtado, who is also a professor of structures at the Universidade Lusófona do Porto, where the author was a student of his. The chosen projects follow the storyline of being built using a mixed structure of steel and concrete. The research presents, first, a series of case studies with mixed structural solutions for different architectural spaces. The article then focuses on the testimony of Rui Furtado through an interview that delves into the figure of Paulo Mendes da Rocha, following the thread of the Car Museum in Lisbon, one of the case studies with a mixed structure chosen. In addition, the professional relationship between Paulo and Rui is explored through the exhibition currently on view at Casa da Arquitectura (Porto), "Paulo, beyond drawing", of which Rui Furtado is the curator. Thus, the path is traced towards reconciling conclusions between the worlds of architecture and engineering, with humanism as the protagonist.La presente investigación aborda una serie de proyectos construidos por la empresa de ingeniería “AFA Consult” a través del testimonio de uno de sus ingenieros, Rui Furtado, que además es profesor de estructuras en la Universidade Lusófona do Porto, donde el autor fue alumno suyo. Los proyectos escogidos siguien la linea argumental de estar construidos mediante una estructura mixta de acero y hormigón. La investigación presenta, primeramente, una serie de casos de estudio con soluciones estructurales mixtas para diferentes espacios arquitectónicos. El artículo, posteriormente, se centra en el testimonio de Rui Furtado mediante una entrevista que profundiza en la figura de Paulo Mendes da Rocha, siguiendo el hilo del Museo de los Coches en Lisboa, uno de los casos de estudio con estructura mixta escogidos. Memory, Conflict & Commerce in Early Modern Europe
This is a solid and compelling collection of articles dedicated to the themes of conflict, memory and language in early modern English theatre and poetry. Shakespeare is the main focus of most of the papers, but there are welcome deviations from this rule, either as complements to the discussion (usually other early modern dramatists, who are dealt with in varying degrees of detail) or as independent objects of investigation (John Donne’s poetry in Margaret Edson’s Wit).
In the two essays having more directly to do with memory, “Memory of Tragedy” and “This is my play’s last scene”, memory is illuminatingly exercised from different temporal positions, in one case as the early modern memory of tragedy and in the other as the contemporary moment’s memory of early modern poetry. However, in “Memory of Tragedy”, some additional bibliographical support would be welcome. The use of the concept of memory, which is central to the article, would benefit from explicit grounding in cultural or collective memory studies. The article could also be improved by a conclusion bringing together, under the theme of the memory of tragedy, the several variants of tragedy discussed throughout the paper. “This is my play’s last scene”, which also deals, though more implicitly, with forms of memory, is an engaging and compelling essay about the central role played by John Donne’s poetry in a contemporary play, Margaret Edson’s Wit. The close attention devoted to poetic detail, which is itself a theme in Wit, neatly brings out the relations between Donne’s poetry and Edson’s play, while linking this paper to the remaining ones.
Whereas “Memory of Tragedy” discusses issues of genre, “As You (Don’t) Like It” focuses on plotting. By means of a structural analysis of central aspects of the plots of As You Like It, King Lear and Hamlet, the author proposes a convincing account of the importance of heterotopic spaces in the movement towards the restitution of order in all three plays. The shift from As You Like It, where an effective restoration does occur, to the two tragedies, where this restoration is complicated and eventually cancelled by contingency, is itself historicised as a general transition from “‘cyclical-temporal’ myth-making into ‘discrete-linear’ plot-making” (Lotman).
A common link between these papers is the attention devoted to language and form. This focus is attained not at the cost of history, but rather in close connection with it. In its attention to issues of poetic language, this collection of articles strikes a welcome note in a field which, in recent times, has frequently disparaged attention to form as mere formalism. The importance of analysing the language of these texts in detail is especially borne out in the remaining three articles of this collection. “NEW LANGUAGES” is a fascinating, and bibliographically supported, discussion of the transformations undergone in the use of rhetoric during the early modern period, as exemplified in and mapped out by a number of history plays from this time. The relations between rhetoric, truth, value and power are brought out through a persuasive discussion of specific moments from Shakespeare’s Second Tetralogy as well as from John Ford’s Perkin Warbeck. “The Insults of The Merchant of Venice”, on the other hand, makes a forceful and amply demonstrated case for understanding insults in The Merchant of Venice as profoundly and disturbingly different from other, more playful types of insults in the Shakespearean canon. This article needs some editing (especially because of incomplete references) and matters of style should occasionally be attended to, namely in the first paragraph (the phrase “I started a paper” is used several times without variation). In section 3, the paragraph containing the final sequence of words as well as their dictionary meanings may perhaps be tightened in terms of its structure. Although these words and their meanings are all motivated by the central connection which is made between insults and rating, their brief appearance in quick succession seems occasionally gratuitous. The final article in this collection, “With all my Heart”, also focuses on The Merchant of Venice. In what is an admirably pursued and amply footnoted source study, the author reconsiders the motif of the pound of flesh in The Merchant of Venice, and more specifically of Antonio’s heart, by bringing out its double-edged signification. This contextual approach moreover benefits from being carried out through a close analysis of emblem books and accounts of public executions in early modern England, while also paying attention to other, more canonically established narrative sources
As células T durante infeções micobacterianas: produção e activação
Tese de Doutoramento em Ciências da SaúdeInfections caused by bacteria from the Mycobacterium genus are among the major
health threats worldwide. Mycobacterium tuberculosis is the etiological agent for tuberculosis
(TB). In 2014, TB alone was responsible for 9.6 million people felling ill and 1.5 million deaths.
The incidence of other mycobacteriosis that used to affect mainly immune-compromised
patients (such as HIV/AIDS patients) is rising even in HIV-negative people. In several cases
these mycobacteriosis are caused by mycobacteria from the Mycobacterium avium complex
(e.g.: M. avium). It is crucial to better understand T cell response to mycobacterial infections
and how infection by these bacteria affects T cell differentiation and immunity.
Previous results from our lab showed that disseminated infection with a high virulence
strain of M. avium (25291), but not with a low virulence strain (2447), lead to a progressive
thymic atrophy. Here we uncover a complex mechanism responsible for M. avium-induced
premature thymic atrophy during which increased levels of corticosteroids synergize with nitric
oxide produced by interferon gamma (IFNγ) activated macrophages. Following these results,
we further show that IFNγ reduces the percentage of T cell progenitors in the bone marrow
(BM). Furthermore, IFNγ also diminishes the ability of BM precursors to reconstitute the
thymus of immune-compromised mice. Besides effects extrinsic to the thymus, we observed
that thymic stroma from M. avium 25291 infected mice is not able to sustain T cell
differentiation, even when BM precursors from uninfected mice are provided. Finally, M. avium
25291 infected mice present increased thymocyte death dependent on IFNγ and independent
of caspase 3. The role of nitric oxide is still unclear, further study is needed to uncover if this
mediator of premature thymic atrophy is acting directly or indirectly on the thymus or on the
BM.
IFNγ is the best-described T cell-derived cytokine known to be critical for host
resistance against M. tuberculosis infection. However, improved protection against TB was not
achieved when increased production of IFNγ was induced, indicating a role for additional
meditators yet to be uncovered. IL-21 is produced predominantly by activated CD4+ T cells and
has been shown to be very important during viral chronic infections and cancer. Here we show
that IL-21 signaling plays a crucial role in T cell responses during M. tuberculosis infection by augmenting CD8+ T cell priming, promoting T cell accumulation in the lungs, and enhancing T
cell cytokine production. In the absence of IL-21 signaling, more CD4+ and CD8+ T cells in
chronically infected mice express the T cell inhibitory molecules PD-1 and TIM-3. Finally we
correlate these immune defects with increased susceptibility of IL-21RKO mice, which have
increased lung bacterial burden and earlier mortality compared to WT mice.
A central function of CD4+ T cells is to regulate other components of the immune
system during the immune response to pathogens. Little is known about the need for CD4+ T
cell help during M. tuberculosis infection. As a cytokine mainly produced by CD4+ T cells and
shown to be essential for CD8+ T cell immunity to TB, IL-21 is a strong candidate as mediator
of CD4+ T cell help to CD8+ T cells during the immune response to M. tuberculosis infection.
Here we present the development of a new and efficient model to study CD4+ T cell help
during M. tuberculosis infection. Using this model we show that helped CD8+ T cells are more
protective than the helpless ones, by reducing bacterial burden and increasing survival of M.
tuberculosis infected immune-compromised mice. Finally this model allowed us to compare
the expression profile between helped and helpless CD8+ T cells and we present a possible
gene signature to identify when CD8+ T cells are or not being helped.
The findings presented in this thesis contribute to a better understanding of T cell
immunity to mycobacteria infections. They show the mechanisms responsible for M.aviuminduced
thymic atrophy and uncover new T cell mediated immune responses that are essential
for optimal immunity to TB. With these findings we provide new insights for the prevention and
treatment of mycobacterial infections.As infeções causadas por bactérias do género Mycobacterium são consideradas de
grande ameaça à saúde mundial. O Mycobacterium tuberculosis é responsável pela doença
denominada tuberculose (TB). Só em 2014, 9.6 milhões de pessoas ficaram doentes com TB
e 1.5 milhões morreram com esta doença. Além da TB, há outras micobacterioses que no
passado afectavam apenas pessoas com imunodeficiências (como por exemplo HIV/SIDA).
Recentemente, a incidência destas micobacterioses tem vindo a aumentar mesmo em
pessoas sem imunodeficiência reconhecida. Estas micobacterioses são causadas na maioria
dos casos por bactérias pertencentes ao Complexo Mycobacterium avium como por exemplo
M. avium. O estudo da resposta imunitária mediada por células T, bem como o estudo de
alterações na diferenciação de células T, é de grande relevância para melhor percebermos
como evitar e resolver infeções causadas por micobactérias.
A infeção disseminada de ratinhos com a estirpe 25291 de M. avium provoca atrofia
tímica, mas este fenómeno não é observado após infeção com a estirpe 2447. Nesta tese nós
mostramos que a atrofia tímica resultante da infeção com M. avium é causada pela sinergia
entre glucocorticoides circulantes com óxido nítrico produzido por macrófagos ativados por
IFNγ. Na continuação destes resultados nós demonstramos que a ativação de macrófagos por
IFNγ diminui a percentagem de precursores de células T na medula óssea (MO) e que o IFNγ
diminui a capacidade dos precursores da MO de reconstituir o timo de ratinhos imunodeficientes.
Além de alterações na MO, nós observamos que o estroma tímico de ratinhos
infectados com M. avium 25291 apresenta uma capacidade diminuída de sustentar a
diferenciação de células T, mesmo na presença de precursores oriundos de MO de animais
não infectados. Por último, observamos um aumento de morte de timócidos em ratinhos
infectados com M. avium 25291, a qual é dependente de IFNγ e independente de caspase-3.
O mecanismo pelo qual o óxido nítrico é responsável pela atrofia tímica ainda está por
desvendar. É necessário novos estudos para perceber se este mediador da atrofia tímica atua
diretamente no timo ou na MO.
O IFNγ é uma citoquina produzida por células T essencial para uma resposta
imunitária eficaz à infeção por M. tuberculosis. Contudo, há pacientes que mesmo produzindo IFNγ, progridem para doença ativa ou permanecem latentes e não eliminam a bactéria. Além
disso, estudos com o intuito de aumentar a produção de IFNγ não resultaram em aumento de
proteção. Este facto indica a existência de outros mediadores ainda não conhecidos que
desempenham papéis importantes na imunidade na TB. A IL-21 é uma citoquina produzida
predominantemente por células T CD4+, e que é essencial durante a resposta imunitária
contra doenças virais crónicas e cancro. Nesta tese nós mostramos que a sinalização via IL-21
é crucial para a resposta imunitária contra infeção por M. tuberculosis, aumentando o priming
de células T CD8+, a sua acumulação no pulmão e a produção de citoquinas. Além disso, na
ausência do receptor para IL-21, mais células T CD4+ e CD8+ expressam moléculas de
inibição, nomeadamente PD-1 e TIM3, durante a fase crónica da infeção. Por último, ratinhos
sem o receptor para IL-21 infectados com M. tuberculosis apresentam carga bacteriana mais
alta e sobrevivem menos tempo que os ratinhos wild type.
A função principal das células T CD4+ é auxiliar a resposta de outros componente do
sistema imunitário. Contudo, ainda não é claro se as células T CD4+ auxiliam as células T
CD8+ durante a resposta imunitária contra M. tuberculosis. O facto da IL-21 ser produzida por
células T CD4+, e ser crucial para a resposta imunitária mediada por células T CD8+ durante a
infeção por M. tuberculosis, faz desta citoquina um potencial candidato a mediador do auxilio
provido por células T CD4+ às células T CD8+. Nesta tese nós apresentamos o
desenvolvimento de um método novo para estudar o auxilio promovido pelas células T CD4+
durante a infeção por M. tuberculosis. Usando este novo método nós mostramos que as
células T CD8+ auxiliadas protegem melhor que as não auxiliadas. Por fim, este método
também permitiu a comparação do expressão transcripcional entre células T CD8+ auxiliadas e
não auxiliadas e apresentamos aqui uma lista de genes que poderão ser usados para
identificar se as células T CD8+ estão a ser auxiliadas ou não.
Os resultados apresentados nesta tese contribuem para o melhor conhecimento da
imunidade mediada por células T contra infeção por micobactérias assim como dos
mecanismos responsáveis pela atrofia tímica na infeção por M. avium. Através da descrição de
novos mecanismos de resposta imunitária mediada por células T contra infeção por M.
tuberculosis abrimos novas linhas de estudo de prevenção e tratamento contra infeções
micobacterianas. Pelo melhor conhecimento dos mecanismos responsáveis pela atrofia tímica
em infeções crónicas podemos sugerir métodos de a prevenir
