    Dimer-dimer stacking interactions are important for nucleic acid binding by the archaeal chromatin protein Alba

    Archaea use a variety of small basic proteins to package their DNA. One of the most widespread and highly conserved is the Alba (Sso10b) protein. Alba interacts with both DNA and RNA in vitro, and we show in the present study that it binds more tightly to dsDNA (double-stranded DNA) than to either ssDNA (single-stranded DNA) or RNA. The Alba protein is dimeric in solution, and forms distinct ordered complexes with DNA that have been visualized by electron microscopy studies; these studies suggest that, on binding dsDNA, the protein forms extended helical protein fibres. An end-to-end association of consecutive Alba dimers is suggested by the presence of a dimer-dimer interface in crystal structures of Alba from several species, and by the strong conservation of the interface residues, centred on Are and Phe(60). In the present study we map perturbation of the polypeptide backbone of Alba upon binding to DNA and RNA by NMR, and demonstrate the central role of Phe(60) in forming the dimer dimer interface. Site-directed spin labelling and pulsed ESR are used to confirm that an end-to-end, dimer dimer interaction forms in the presence of dsDNA.Peer reviewe

    Anti-tyrosinase activity of Morus alba L. in vitro cell cultures

    reservedLa tirosinasi è un enzima multifunzionale contenente rame che è responsabile della sintesi della melanina umana. Per evitare la sovrapproduzione e l’accumulo di melanina nella pelle che porta all’iperpigmentazione, gli inibitori di tirosinasi sono ampiamente utilizzati sia in campo farmaceutico che cosmetico. Inoltre, la tirosinasi è coinvolta nell’imbrunimento enzimatico di molta frutta e verdura fresca, fenomeno che influenza non solo le qualità sensoriali dell’alimento, ma anche le caratteristiche nutrizionali con conseguente grande perdita economica per l’industria agricola e alimentare. Un gran numero di inibitori della tirosinasi sono riportati in letteratura, tuttavia alcuni di loro mostrano scarsa attività e/o scarso profilo di sicurezza per la salute umana in campo alimentare. Pertanto, è importante trovare inibitori della tirosinasi provenienti da fonti naturali, sicuri e efficaci. Tra gli inibitori della tirosinasi da piante, gli estratti di Morus alba L. (Moraceae) mostrano una notevole attività antitirosinasi. In questo studio sono stati valutati campioni di succo grezzo e frazionato di calli di Morus alba come nuovi inibitori della tirosinasi. I risultati hanno mostrato che tutti i campioni frazionati possiedono una minore attività inibitoria rispetto al succo grezzo. Pertanto, è stato ipotizzato che potrebbe esistere un effetto sinergico tra i componenti del succo grezzo che merita ulteriori ricerche

    Abstract Essay in Political Science, autumn 2010. ”ALBA – A regional integration project with good conditions?” Autor: Anna Sjögren. Course: Political Science, C-level, Tutor: Sten Berglund   An essential part of the political science research on regional integration has been about why states join economic unions and what forces drive the process of integration. This essay is a case study on the Latin American regional integration project called ALBA. The purpose is to, from the ideas of the swiss political scientist Walter Mattli, investigate if ALBA has the conditions to be a successful integration group. To reach the purpose two questions have been asked: 1) What does the demand for regional integration in Latin America looks like? 2) What does the supply of regional integration in Latin America looks like? My methodological approach has been to read scientific articles and news articles that deal with ALBA and to see what they say about the conditions that are crucial according to Mattli. The result shows that the market in the region is not so diversified but that the member states have been doing some trading with each other. A lot of the trade has involved oil and that has effected the cooperation. The global financial crisis has implyed challenges to ALBA, as the oil price has fallen. But it has also given the alliance more legitimacy when the economic system that ALBA criticize has showed its deficiencies. To become a long-term project ALBA should end its dependence on oil. In Hugo Chávez ALBA has a rather undisputed leader. That Cuba, since the start of ALBA, has changed its political leader can have a significance. Due to the political orientation of ALBA the deveopment in the member states is crucial for the future of the union. That Brazil has chosen not to be a member of ALBA has both advantages and disadvantages for the alliance. Chávez has avoided a putative struggle of power but ALBA would probably be stronger if the region's economic, demographic, and territorial giant was participating

    The Author/Translator Interactional Process. A Case Study

    See Naples and Kill (1988) is a lively and colourful novel by the con-temporary English writer, Gregory Dowling, translated into Italian in 2015. Following the tradition of translation studies (Venuti 2000, Bass-nett 2002, Cronin 2006), this paper analyses the rewriting process of literary translation, considering in particular the fruitful but sometimes tense and even conflictual relationship between writer and translator. The translation of the novel See Naples and Kill was an ongoing rewriting process entailing a constant dialogue between the writer and the translator. Therefore, the study aims at answering two main ques-tions: what happens if the rewriting process of translation is constant-ly questioned by the author? What happens if the author has a good mastery of the target language and s/he is her/himself a translator? By exploring the relationship between translation and re-creation, the research focuses on the differences and similarities between the primary creation (source text) and the secondary creation (target text), and aims to verify in which way the dialogic encounter of two different personalities and cultures does not make them merge but, by retaining their own uniqueness, leads eventually to their mutually en-riching each other. A comparative analysis of the source text and the different drafts of the translated version accompanied by the author’s comments will shed light on the tense author-translator relationship in the specific case under investigation and how both actors handle this tension in order to create a new work resulting from the (dis)agreement of the two parties
