Abstract: The macroanatomy of the tentacular head of Phascolosoma arcuatum belongs to phylum Sipuncula has been studied under light microscopic methods and computer aided graphics analysis. The tentacular head structure has been reconstructed using computer aided graphics analysis using series histological images on computer aided software platform. The P. arcuatum has only five, semitransparent, pigmented, lobed tentacles in tentacular crown with large tentacular coelomic connections. We found that the tentacles are lined by ciliated columnar epithelium with large number of mucosecretory cells, indicating their deposit feeding habit. Fewer number of tentacles, body wall and there burrowing habit supports there integumentary breathing habit. A thick striated collar encircles the tentacles forming the ventral wall of the mouth channel and protects tentacles from mechanical damage when retracted. These structural peculiarities revealed from macroanatomical analysis are strongly suggestive about the important role of the collar region of the tentacular crown of the head of P. arcuatum in relation to its burrowing habit to its concern habitat. Abstract The research objectives of this study are to analyze the effectiveness of committee and mechanism of corporate governance to prevent financial crime. Financial Crime often occur inside the company, especially in Indonesia. Financial crime covers fraud, money laundering, bribe, corruption, terrorist financing and so on. At 2018, there’s case where PT. Garuda Indonesia committed revenue recognition fraud by recognizing revenue amounted to USD 239 million from Mahata Aero Teknologi which is yet to be received by the end of 2018. The company’s management and external audit are fined for the negligent in preventing the fraud. Therefore, the effectiveness of company’s organizational structure played a big role to supervise and prevent the occurrence of financial crimes. In this discussion, the author investigates the effectiveness of audit committee, board of commissioners, stand-alone risk committee, external audit, effect of managerial ownership and company’s performance toward financial crime. Data for the research were collected from secondary data found in annual report of 426 companies and analyzed using logistic regression method using SPSS 25. The study findings prove that stand-alone risk committee, external audit, and company’s performance have a significant negative effect toward financial crime, while size of audit committee have a significant positive effect toward financial crime. Research show that meeting frequency of audit committee, board of commissioners, and managerial ownership has no significant effect toward financial crim

    No full text
Ross syndrome is a rare partial dysautonomic syndrome of unknown aetiology, characterized by segmental hypo/ anhidrosis associated with Holmes-Adie syndrome (tonic pupil and hypo/areflexia). The hypohydrosis or anhydrosis is patchy initially, later it becomes segmental or diffuse. This is due to affection of postganglionic cholinergic parasympathetic and sympathetic fibers involvement. There are a very few cases (approximately 50) have been reported in the literature since its original description. Author report a 22 years old male with classical features of Ross syndrome

Programmable human histone phosphorylation and gene activation using a CRISPR/Cas9-based chromatin kinase

    No full text
Histone phosphorylation is a ubiquitous post-translational modification that allows eukaryotic cells to rapidly respond to environmental stimuli. Despite correlative evidence linking histone phosphorylation to changes in gene expression, establishing the causal role of this key epigenomic modification at diverse loci within native chromatin has been hampered by a lack of technologies enabling robust, locus-specific deposition of endogenous histone phosphorylation. To address this technological gap, here we build a programmable chromatin kinase, called dCas9-dMSK1, by directly fusing nuclease-null CRISPR/Cas9 to a hyperactive, truncated variant of the human MSK1 histone kinase. Targeting dCas9-dMSK1 to human promoters results in increased target histone phosphorylation and gene activation and demonstrates that hyperphosphorylation of histone H3 serine 28 (H3S28ph) in particular plays a causal role in the transactivation of human promoters. In addition, we uncover mediators of resistance to the BRAF V600E inhibitor PLX-4720 in human melanoma cells using genome-scale screening with dCas9-dMSK1. Collectively, our findings enable a facile way to reshape human chromatin using CRISPR/Cas9-based epigenome editing and further define the causal link between histone phosphorylation and human gene activation.Fondren Library Open Access Author Fun

Common Charge-Shift Mutation Glu65Lys in K+ Channel beta(1)-Subunit KCNMB1: Pleiotropic Consequences for Glomerular Filtration Rate and Progressive Renal Disease

    No full text
Background: Glomerular filtration rate (GFR) is a heritable trait, and hyperfiltration (GFR increment in remnant nephrons) may accelerate renal functional decline in chronic kidney disease (CKD). Mesangial and vascular smooth myocytes control GFR by contraction, dependent on voltage-gated Ca2+ influx, which is controlled by the regulatory beta(1)-subunit (KCNMB1) of large-conductance heteromeric K+ ('BK') channels. KCNMB1 gain-of-function variant Glu65Lys results in generalized vasorelaxation and thus protection against systemic hypertension. Here we asked whether the Glu65Lys variant influences GFR, in the basal state or during progressive renal decline. Methods: We explored Glu65Lys effects on GFR in three populations spanning two ethnicities and two diseases (hypertension and nephrosclerosis). GFR was either estimated (eGFR from serum creatinine) or directly measured (iothalamate clearance). Results: The 65Lys variant was relatively common, occurring on similar to 5-10% of chromosomes in different biogeographic ancestry groups, and 65Lys carriers exhibited higher eGFR in two primary care populations: extreme BP values in Kaiser clinics (p = 0.029, accounting for similar to 0.2% of trait variance), or treated hypertensives in VA clinics (p = 0.017, accounting for similar to 0.9% of trait variance). In blacks with progressive renal disease (NIDDK AASK), 65Lys carriers displayed a steeper slope in GFR chronic decline (p = 0.030, accounting for similar to 0.4% of trait variance), and Glu65Lys genotype also predicted time of onset of renal failure (log rank p = 0.019). Conclusions: Common KCNMB1 gain-of-function variant Glu65Lys influences GFR, and 65Lys carriers exhibit not only elevated baseline GFR, but also more rapid GFR decline (and consequent development of renal failure) in CKD. The results suggest that profiling patients at Glu65Lys can assist in gauging renal prognosis as well as selection of rational therapy in hypertension with progressive renal disease.
