    El conflicto colombiano sitúa una dimensión social, cuyo factor desencadenante ha sido la ocupación de territorio, es decir, en el pasado como en la actualidad el acceso a la tierra confronta a la inmensa minoría (terratenientes-latifundios) con la gran mayoría (campesinos e indígenas, particularmente) en torno al uso de la tierra. La tan anhelada reforma agraria en Colombia ha sido mediatizada a través de leyes, programas, proyectos que al final se resumen en paliativos agrarios; esta resistencia al llevar a cabo la reforma agraria, ha devenido en una muy significativa razón de lucha guerrillera, estrategias de colonización por paramilitares, apropiación indebida de baldíos por los feudales y finalmente agentes internacionales con la mirada puesta en la explotación de los recursos naturales. Los acuerdos de paz con los grupos ilegales han sido realizados de manera paralela al conflicto armado. En Colombia, los diálogos de paz se presentan en medio del sostenimiento del conflicto lo que ha ocasionado que tanto grupos guerrilleros y paramilitares tengan reservas al momento de la desmovilización, en especial por la vulnerabilidad a la que quedan expuestos los combatientes y en particular por las expresiones de violencia en contra de los excombatientes a cargo del grupo contrario, o inclusive enmarcadas en una política criminal de orden estatal.CONACy

    Production, characterization and evaluation of anti-diabetes peptides from proteins of improved common bean (Phaseolus vulgaris L.) cultivars using bioinformatic tools, enzymatic systems, and in vitro and in vivo models

    Diabetes is an important metabolic disease expected to increase worldwide 55% by 2035; with an estimated healthcare cost of $245 billion per year in the US alone. The use of alternative plant based treatments has the advantage of no side effects and could reduce treatment cost. Common beans health benefits are attributed to bioactive compounds; including resistant starch and oligosaccharides that contribute to dietary fiber; phenolic compounds and bioactive peptides released from proteins during enzymatic digestion. Biological potential of bean peptides has been related to antihypertensive, anticancer, anti-inflammatory, antioxidant and antifungal; however, the anti-diabetes potential of common bean protein fractions and pure peptides has not been studied. In that sense, the central hypothesis of this research was that common bean protein hydrolyzed fraction contains bioactive peptides that could modulate molecular targets of type 2 diabetes. To fill the gap in the knowledge, our long-term goal was to understand the mechanism by which common bean peptides contribute to the management of type 2 diabetes. To achieve this aim, we generated anti-diabetes peptides from common bean protein and determined their efficacy using bioinformatics tools, enzymatic systems, in vitro and in vivo models. Our research started with the selection and characterization of proteins, peptide profile, protein inhibitors, α-amylase inhibitors, phenolic compounds and anthocyanins of improved common bean cultivars from Mexico and Brazil (Aim 1). The peptides and phenolic compounds found presented outstanding anti-diabetes potential from the biochemical and in silico studies. Furthermore, we evaluated the peptides anti-diabetes potential after simulated gastrointestinal digestion and characterized their bioactive peptides, angiotensin converting enzyme, dipeptidyl peptidase IV, and α-glucosidase inhibition (Aim 2). Negro Otomi cultivar presented the lowest IC50 to inhibit DPP-IV (0.14±0.01 mg DW/ mL). The effect of processing was evaluated in Aim 3, where the goal was to determine the bioactive properties of the released peptides from commercially available precooked common beans (Phaseolus vulgaris). Bioactive properties and peptide profiles were evaluated in protein hydrolysates of raw and commercially precooked common beans. Hydrolysates from Navy beans were the most potent inhibitors of DPP-IV with no statistical differences between precooked and raw (IC50 = 0.093 and 0.095 mg protein/mL, respectively). In general, we observed that processing did not affect the bioactive properties of released peptides from precooked beans. With the objective to increase the anti-diabetes potential of common bean protein fractions, the fourth Aim was to optimize the production of anti-diabetes protein fractions from black bean (Phaseolus vulgaris L.) protein isolates. The results suggested that the best combination of conditions to generate anti-diabetes peptides were with alcalase for two h and E/S of 1:20, with high inhibitions values for dipeptidyl peptidase IV (DPP-IV, 96.7%), α-amylase (53.4%) and α-glucosidase (66.1%). The protein fractions were characterized using LC-ESI-MS/MS and 33 sequences were identified. Finally, we evaluated the anti-diabetes mechanism of action of common bean protein fractions and pure peptides using in silico, in vitro, and in vivo models. Sequenced peptides showed outstanding potential to bind and blocking intestinal glucose transporters SGLT1 and GLUT2. Besides, using Caco-2 cells monolayer insert we observed that protein fractions reduce glucose absorption by 6.5 and 24.5 after 30 min and 24 h, respectively. It could be due to protein fractions were also able to decrease protein expression and translocation to the membrane of GLUT2 and SGLT1. Furthermore, protein fractions were able to lower postprandial glucose by 24.5% in an oral glucose tolerance test in rats after receiving 50 mg/kg BW compared to untreated control. Also, a hyperglycemic rat model showed that protein fractions at 100 mg of PF/ Kg BW twice daily significantly reduced postprandial glucose levels by 48% compared to the non-treated diabetic group, without having consequences in body weight or other biochemical parameters.     Este proyecto se basó en la práctica pedagógica e incongruencias con los lineamientos curriculares del ministerio de educación nacional. Área de ciencias naturales. Para ello, se realizó una investigación tipo cualitativa, pues los instrumentos que se utilizaron permitieron establecer un análisis descriptivo sobre los sucesos o hechos en la investigación. La información se obtuvo mediante una revisión documental, una entrevista semiestructurada y observación directa. La población y muestra correspondió a los docentes miembros del equipo de área de Ciencias Naturales. Se logró revisar la articulación entre la estructura nacional de formación curricular y la propuesta formativa institucional. Posteriormente, se establecieron las incongruencias entre los lineamientos curriculares.     La zona minera del centro del Cesar tiene la presencia de diversas empresas mineras y diferentes operaciones. Para vigilar la calidad de aire de la zona se tienen implementado por parte de la autoridad ambiental (CORPOCESAR) una moderna red de monitoreo acreditada por el IDEAM, cuyas estaciones se ubican en las poblaciones del área de influencia de las operaciones mineras y registran todas las fuentes de emisiones, no solo las provenientes de la zona minera, sino también las emisiones generadas al interior de las poblaciones por quema de basuras, tráfico de vehículos, entre otros. Conscientes de la gran variedad de fuentes de emisión existentes en la zona, las Empresas Mineras han implementado una serie de medidas tanto al interior de los proyectos mineros, como en las poblaciones vecinas y alrededores, con el objetivo de mejorar las condiciones de calidad aire de la zona. Dichas medidas se han realizado en el marco de la gestión ambiental efectiva realizada por las empresas en coordinación con los diferentes actores en la zona minera del departamento del Cesar. La implementación de estas medidas ha requerido desarrollar una estrategia que va desde la realización de diferentes estudios técnicos a nivel local y regional para identificar y evaluar objetivamente las fuentes de emisión fuentes más representativas de la zona, hasta el trabajo coordinado del sector público y privado para la ejecución de medidas puntuales para el control de emisiones. Se presentará el esquema de gestión de la calidad de aire implementado en la zona minera del Cesar y la descripción de los sistemas y tecnologías usadas actualmente al interior de los proyectos mineros para la mitigación de material particulado, la articulación de estas medidas a modernos sistemas de medición en tiempo real para su control, así como la descripción de las medidas externas y su impacto en el mejoramiento de la calidad de aire de las poblaciones

    Effect of metallic waste addition on the electrical, thermophysical and microwave crack-healing properties of asphalt mixtures

    This paper aims to evaluate the effect of metallic waste addition on the electrical, thermophysical and microwave crack-healing properties of asphalt mixtures. With this purpose, asphalt mixtures with two different types of metallic waste, steel wool fibres and steel shavings, added in four different contents, were tested. Electrical and thermophysical properties of asphalt mixture specimens with, and without, metallic waste were measured. The spatial distribution of the metallic waste inside the asphalt mixture samples was evaluated by using X-ray computed tomography. In addition, crack-healing properties of asphalt samples using microwave radiation heating were assessed at two different healing times, 30 s and 40 s. To quantify the efficiency of the healing process, five healing cycles were carried out for each asphalt sample. The main results showed that asphalt mixtures with shavings presented lower air void contents than mixtures with fibres. Moreover, fibres produced an increase in the electrical conductivity of the mixtures because long fibres in the mixtures form electrically conductive channels. In contrast, shavings did not have significant effect on the electrical properties of the mixtures. Likewise, it was proven that metallic waste reduced the thermal conductivity and the specific heat capacity of asphalt mixtures. Conversely, shavings decreased the thermal diffusivity of asphalt mixtures regardless of their content. Overall, it was found that the healing level reached by the asphalt mixtures tested by microwave radiation depends on the healing time and the type and content of metallic waste used.
