    Mice as Early Biomarkers of Alport Syndrome

    The efficiency of the inhibition of the angiotensin converting enzyme, the most widely used therapy for the Alport syndrome, depends on the onset of the therapy-the earlier the better. Hence, early progressive biomarkers are urgently required to allow for preclinical diagnosis, an early start of possible therapy as well as the monitoring of this therapy. In the present study, an improved comprehensive and precise proteomic approach has been applied to the serum of juvenile Alport-mice, nontreated and treated, and wild-type controls of various ages to search for biomarkers. With a total of 2542 stringently altered proteins, the serum composition clearly shows a dependency on age, that is, stage, and therapy. Initially, the serum constituents indicate an enhanced extracellular matrix remodeling, cell damage, and the production of particular acute phase proteins. A panel of IS potential biomarker candidates has been identified. In later stages, renal filtration failure and systemic acute phase reaction determine the composition of the serum; an effect that is well-known for manifested human Alport syndrome. With a small number of mouse urine samples, for example, the proteomic results for gelsolin could be verified using ELISA. Once verified in man, these early biomarkers would allow for a sensitive and specific diagnosis of the Alport syndrome in children as well as facilitate the monitoring of a possible therapy

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Das Alport Syndrom (AS) ist eine genetisch bedingte Nierenerkrankung, die durch Mutationen in den Genen COL4 α3, α4 oder α5 hervorgerufen wird. Diese kodieren für die Ketten α3, α4 bzw. α5 von Kollagen IV, und sind damit wesentlich am Aufbau der adulten glomerulären Basalmembran der Niere beteiligt. Im Verlauf der Erkrankung kommt es oft schon im Kindesalter zu einer voranschreitenden Beeinträchtigung der Nierenfunktion bis zum terminalen Nierenversagen. Inzwischen ist es möglich, den Fortgang der Krankheit durch den Einsatz von ACE-Hemmern (ACE – Angiotensin-converting enzyme), wie zum Beispiel Ramipril, zu verlangsamen und die Zeit bis zur Dialyse- oder Transplantationspflichtigkeit entscheidend zu verzögern. Diese Therapie ist umso effizienter, je eher damit begonnen wird. Doch aufgrund des symptomlosen Beginns wird die Erkrankung bisher meist zu spät oder gar nicht diagnostiziert. Daher werden präklinische Krankheitsmarker zum frühzeitigen Nachweis dringend benötigt, um einen schnelleren Therapiebeginn und damit eine Verbesserung des zu erwartenden Therapieerfolges zu erzielen. Zudem sind noch immer viele Abläufe der Pathogenesekette des AS ungeklärt. Im Rahmen dieser Dissertation sollten wenig-invasive präklinische Biomarker für das AS gesucht werden. Dies erfolgte mittels massenspektrometrischer Analyse von Serum-Proben von COL(IV)A3 -/--Mäusen im Vergleich zu gesunden Kontrolltieren. Außerdem sollte untersucht werden, ob diese Biomarker durch die Ramipril-Behandlung beeinflusst werden. Untersucht und verglichen wurden Proben von gesunden Kontroll-, unbehandelten-, und mit Ramipril behandelten COL(IV)A3 -/- -Mäusen, jeweils in den drei juvenilen Altersgruppen von 4,5, 6 und 7,5 Wochen. Dazu erfolgte zunächst eine zweidimensionale Fraktionierung der Serum-Proben mittels Größenausschluss- und Anionenaustauschchromatographie. Anschließend wurde die massenspektrometrische Analyse der Fraktionen und die softwaregestützte Auswertung der Daten vorgenommen. Es wurden beim unbehandelten AS 115 Proteinketten mit ansteigender, 41 mit abfallender Konzentration und 18 mit sowohl ansteigenden als auch abfallenden Proteoformen gefunden. Zahlreiche dieser Veränderungen sind unter Ramipriltherapie reversibel. Daraus wurden 25 Biomarkerkandidaten abgeleitet, die sich durch eindeutige Unterschiede zu Vergleichsproben bereits in der präklinischen Krankheitsphase auszeichnen. Diese sind das Retinol-bindende Protein 4, Serum-Amyloid P, HMW Kininogen II, Transferrin, LCAT-Phosphatidylcholin-Sterol-Acyltransferase (LCAT), Clusterin (Apolipoprotein J), Betaine-Homocysteine-S-Methyltransferase 1 (BHSM1), Isoform 2 der Kollagen-α1(I)-Kette (COLα1), Hämopexin, Haptoglobin, Angiotensinogen, Apolipoprotein (Apo) AI, Apo A IV, Apo CIII, Beta-2-Glykoprotein 1 (2GP1), Komplementfaktor H, Fibronektin, Isoform 3 der Sulfhydryl-Oxidase 1 (QSOX1), Leucin-reiches-HEV-Glykoprotein (LRHEV), der Inter-alpha-Trypsin-Inhibitor-(IATI)-Komplex aus dem alpha-1-microglobulin/bikunin precursor Protein (AMBP), die schweren Ketten H1 (HCH1) und H2 (HCH2), Vomeronasal 2 Rezeptor 67 (Vom), Zinkfingerprotein 758 und die Superoxid-Dismutase. Es ist weiterführend in humanen Proben zu prüfen, ob diese Markerkandidaten eine ausreichende diagnostische Sensitivität und Spezifität haben und ob sie zum Therapiemonitoring und bei der Entwicklung neuer Therapieansätze Verwendung finden können. bzw. 5 von Kollagen IV, und sind damit wesentlich am Aufbau der adulten glomerulären Basalmembran der Niere beteiligt.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
