    Surface immunoglobulins as a target for antibody therapy

    This thesis relates to the therapeutic use of antibody in the treatment of chronic lymphocytic leukaemia. The antibody was raised in sheep and directed against idiotypic determinants (Id) on the surface immunoglobulins of the neoplastic lymphocytes. Three patients received the therapy. The author's contributions to this work were to characterise Fab fragments prepared from the surface immunoglobulin, to investigate the catabolism of infused xenogeneic antibody, and to assist in the clinical trials.The work is divided into three sections. The first section deals with the proteolytic cleavage of surface and secreted immunoglobulins, the second with the catabolic rates of immunoglobulins in xenogeneic animals, the third with the clinical trials. In the first section, papain was used to cleave surface immunoglobulins on the neoplastic lymphocytes from chronic lymphocytic leukaemia patients. The immunoglobulins were cleaved in situ, releasing Fab fragments that carry idiotypic determinants specific to the particular neoplastic clone of cells. Sheep were immunised with the Fabµ fragments to produce anti-Id antibodies. The cleavage of surface IgM with papain released Fabµ fragments and another molecule which we termed fragment X. Fragment X contains Fabµ determinants which are not present on Fabµ prepared from serum IgM. These non-Fabµ determinants were highly immunogenic in sheep, and antibodies produced against them are of no use in antibody therapy. Fragment X probably results from the cleavage of membrane-bound IgM at a site C-terminal to the Cµ3 domain. Fragment X was detected in small amounts in the papain digest of serum IgM. The fragments released by the cleavage of surface IgD with papain were also studied. In the second section, the catabolic rates of rabbit IgG and its fragments in strain two guinea pigs were studied. This was to determine the feasibility of using the monovalent fragment Fab/c and whole IgG in antibody therapy. Whole IgG and Fab/c had similar survival times in guinea pigs, with a half life of around 8 days. Faby fragments were catabolised very rapidly with a half life of a few hours. In the third section, three patients with chronic lymphocytic leukaemia were treated with sheep antibodies directed against the Id on the surface immunoglobulin of their neoplastic cells. In each instance there was partial destruction of the tumour, the extent of which varied from one patient to the other. The effectiveness of they antibody therapy probably depends on many factors including the tumour load, antigenic modulation and the levels of circulating immunoglobulins that carry the tumour Id. The survival time of sheep IgG was determined in each patient.</p

    Dewasa ini orang banyak memperdebatkan kehujjahan hadis  do’if, apakah boleh dijadikan landasan beramal atau malah sebaliknya. Mereka melupakan hadis  ahad yang jauh pada abad kedua sudah menjadi perdebatan diantara ulama Mu’tazilah dan Ahlusunnah. Al-nadham sebagai ulama dari kelompok Mu’tazilah menganggap bahwa hadis  ahad bersifat dhan sehingga tidak dapat dijadikan hujjah dalam hal aqidah, mereka menolak adanya penggunaan hadis  ahad. Padahal periwayatan secara ahad tidak bisa terelakkan karena periwayatan hadis  pada masa Nabi dengan cepat tersebar ke masyarakat. Para sahabat sangat antusias dalam menerima dan menyampaikan secara bebas hadis  yang diterima dari Rasulullah saw. tanpa melihat berapa jumlah jalur orang yang menyampaikannya. Periwayatan secara ahad ataupun mutawatir tidak menjadi perhatian. Untuk mengkaji masalah tersebut maka penulis menggunakan pemikiran Ali Mustafa Yaqub dalam membahas “Dikotomi hadis  ahad terdapat bukunya yang berjudul Kritik Hadis. Penting untuk memahami perdebatan tentang hadis  ahad tersebut guna menambah wawasan. Menurut analisis penulis hadis  ahad tidak bisa ditolak dan tidak dijadikan hujjah dalam masalah akidah, ketika ini dilakukan konsekuensinya banyak hadis  yang akan ditolak, seperti tentang syurga dan beraka, dajjal, siksa kubur, mizan di akhirat dll.     With the rapid penetration of technology in varied application domains, the existing cities are getting connected more seamlessly. Cities becomes smart by inducing ICT in the classical city infrastructure for its management. According to McKenzie Report, about 68% of the world population will migrate towards urban settlements in near future. This migration is largely because of the improved Quality of Life (QoL) and livelihood in urban settlements. In the light of urbanization, climate change, democratic flaws, and rising urban welfare expenditures, smart cities have emerged as an important approach for society’s future development. Smart cities have achieved enhanced QoL by giving smart information to people regarding healthcare, transportation, smart parking, smart traffic structure, smart home, smart agronomy, community security etc. Typically, in smart cities data is sensed by the sensor devices and provided to end users for further use. The sensitive data is transferred with the help of internet creating higher chances for the adversaries to breach the data. Considering the privacy and security as the area of prime focus, this book covers the most prominent security vulnerabilities associated with varied application areas like healthcare, manufacturing, transportation, education and agriculture etc. Furthermore, the massive amount of data being generated through ubiquitous sensors placed across the smart cities needs to be handled in an effective, efficient, secured and privacy preserved manner. Since a typical smart city ecosystem is data driven, it is imperative to manage this data in an optimal manner. Enabling technologies like Internet of Things (IoT), Natural Language Processing (NLP), Blockchain Technology, Deep Learning, Machine Learning, Computer vision, Big Data Analytics, Next Generation Networks and Software Defined Networks (SDN) provide exemplary benefits if they are integrated in the classical city ecosystem in an effective manner. The application of Artificial Intelligence (AI) is expanding across many domains in the smart city, such as infrastructure, transportation, environmental protection, power and energy, privacy and security, governance, data management, healthcare, and more. AI has the potential to improve human health, prosperity, and happiness by reducing our reliance on manual labor and accelerating our progress in the sciences and technologies. NLP is an extensive domain of AI and is used in collaboration with machine learning and deep learning algorithms for clinical informatics and data processing. In modern smart cities, blockchain provides a complete framework that controls the city operations and ensures that they are managed as effectively as possible.
