    Ferritin Gene Expression among Pennate and Centric Diatoms

    The ability to store iron through the use of iron storage proteins, ferritins, has evolved as an advantageous strategy for numerous diatom species. Iron is a micronutrient in the ocean that typically exists at low concentrations throughout much of the marine system. Therefore, iron deficiency often plays a central role in controlling primary productivity, and thus, diatom populations within the ocean (Martin, 1990). This project investigates the expression of an iron storage gene, ferritin, in three pennate diatoms and four centric diatoms. The strains were cultured at Bigelow Laboratory for Ocean Sciences and the University of North Carolina at Chapel Hill (UNC) and grown as triplicate cultures under various iron conditions. This experiment allowed for the comparison of ferritin gene expression along with iron storage capabilities, in contrasting iron environments among phylogenetically distinct diatoms. This was achieved by utilizing the ΔCt method. Our results suggest that pennates may upregulate ferritin levels in high iron environments. Marchetti et al (2009) similarly demonstrated that pennates such as Pseudo-nitzschia granii store iron during temporary pulses, which allows them to continue to proliferate once iron limited conditions return. Two of the examined centrics, however, demonstrated greatest ferritin gene expression under low iron conditions. This result is counterintuitive, as ferritin is a storage gene and high expression during low iron levels appears inefficient. However, this finding supports the theory that centrics may be using their ferritin as an iron buffer and thus eludes to physiological differences in the way these two diatom lineages are storing iron and utilizing their ferritin genes. There is also evidence of a biogeographical distinction among ferritin use in certain species. Diatoms in open ocean regions, where iron is often limited, may be utilizing their ferritin differently than those persisting in high-iron coastal areas.Bachelor of Scienc

    The impact of renewable energy systems on local sustainability

    The purpose of the paper is to discuss and assess the impact of renewable energy, RE, generation on sustainable communities, with a specific focus on employment impact. In recent years, local authorities have been involved in the energy planning process due to the site-specific nature of renewable production and thanks to their nearer connection to citizens. Four small and medium-size sustainable communities belonging to Marche region, in central Italy, have been analysed. The work forecasts the employment generated by the RE capacity defined by two different energy scenarios: 1) the 'business as usual' scenario with a 20% CO2 emission reduction target by 2020, where all the energy policies suggested by Sustainable Energy Action Plans have been considered; 2) the 'potential' scenario with a 50% CO2 emission reduction target by 2050. Results show that the spreading of renewable energy, as expected, can increase local jobs, although the high investments required and problems connected to the impact on the grid have to be considered in the analysis of the global economic effect

    Intact polar diacylglycerol (IP-DAG) lipids measured in cultures of four Antarctic diatom isolates

    Diatoms were isolated by Adrian Marchetti (http://orcid.org/0000-0003-4608-4775) from waters of the Bellingshausen Sea and grown on replete media. Cells were collected onto 0.7 µm GF/F filters. Extraction was performed using a modified Bligh and Dyer (Bligh and Dyer, 1959; https://doi.org/10.1139/o59-099) method described in Popendorf et al. (2013; https://doi.org/10.1007/s11745-012-3748-0). Lipid extracts were analyzed by HPLC-ESI-MS with data dependent-MS2 acquisition on a high-resolution, accurate mass Thermo Q Exactive Hybrid Quadrupole-Orbitrap mass spectrometer (ThermoFisher Scientific, Waltham, MA, USA) coupled to an Agilent 1200 HPLC system (Agilent, Santa Clara, CA, USA). The HPLC-ESI-MS method is described in Collins et al., 2016 (https://doi.org/10.1021/acs.analchem.6b01260). The LOBSTAHS lipidomics discovery software (Collins et al., 2016; https://doi.org/10.1021/acs.analchem.6b01260) was used to putatively identify HPLC-MS features in the data. We confirmed each LOBSTAHS identification using two additional means: (1) via comparison of data-dependent MS2 spectra with those from authentic standards or published reference spectra and (2) by requiring the presence of the same compound identity in data acquired in the opposite HPLC-ESI-MS ionization mode. We confirmed all LOBSTAHS identities at the lipid class level (e.g., PC versus PE, or MGDG versus TAG) using a new, experimental LOBSTAHS feature which automatically detects diagnostic product ion fragments and constant neutral losses (as given in Popendorf et al., 2013; https://doi.org/10.1007/s11745-012-3748-0) in the available data-dependent MS2 spectra for each sample. After identification, quantification of analytes was performed using a series of standard curves, followed by normalization to concentration of an internal standard. Lipid identities are resolved only to the level of bulk fatty acid composition (i.e., the sum of the properties of the substituents at both the sn-1 and sn-2 positions). As a result, in some cases (e.g., PC 40:8), several different molecules having the same bulk fatty acid composition have been identified. These can be distinguished by retention time, which is given in the "Comment" field for each entry

    Migration and Domestic Work

    This open access short reader offers a systematic overview of the scholarly debate on the experiences of migrant domestic workers at a global level, in the past as well as in present time. It tackles the nexus between migration and domestic work with a multi-layered approach. The book looks into the issue of (paid) domestic work in migratory contexts by investigating the feminization of migration, thereby considering the larger framework within which this specific phenomenon takes place. The author explains notions such as the “international division of reproductive labor” or “global care chains” which emphasize the inequality in the way care and domestic tasks are distributed today between middle-class women in receiving nations and migrant domestic workers. Moreover, the book shows how women migrating to work in the domestic work and private care sector are facing a complex landscape of migration and labor regulations that are extremely difficult to navigate. At the same time, this issue also addresses employers’ households who cannot find appropriate or affordable care among declining welfare states and national workers reluctant to take the job, whilst legal regulations make difficult to hire a domestic worker who is a third country national. As such this book offers an interesting read to academics, policy makers and all those working in the field
