Binding of GadE, GadX and NsrR on various promoter regions.
<p>The strains EDL933 Δ<i>gadE</i>-pBADMycHisA::<i>gadE</i>, Δ<i>gadX</i>-pBADHisA::<i>gadX</i>, and Δ<i>nsrR</i>-pBADMycHisA::<i>nsrR</i> were grown in the absence (black bars) or in the presence (white bars) of NOR-4. ChIP assays followed by qPCR were performed to determine the relative enrichment in DNA molecules bound to GadE (A), GadX (B), NsrR (C). Values higher than 20 (twice the values obtained for the strain EDL933 containing the empty pBADmycHisA vector) indicate protein binding to the promoters of interest. Induction of NhaR, GadE, and GadW by YdeO in <i>E. coli</i>.
<p>YY5002 (<i>gadE</i>-<i>3xflag</i>) (a), YY5003 (<i>gadW</i>-<i>3xflag</i>) (b), and BW25113, (c) harboring pTrc99A (−) or pYdeO (+), were grown in LB medium until logarithmic phase. After centrifugation the lysate solution was prepared in lysis buffer containing 8 M urea by sonication. The lysates were subjected to western blotting as described in <a href="http://www.plosone.org/article/info:doi/10.1371/journal.pone.0111962#s2" target="_blank">Materials and Methods</a>. Anti-FLAG (SIGMA) and anti-NhaR (Lab preparation) were used for detection of GadE-3xFLAG/GadW-3xFLAG, and NhaR, respectively [A]. The amounts of GadE-3xFLAG, GadW-3xFLAg, and NhaR were represented as the ratio of level of RNA polymerase-α subunit, detected by anti-α (Neoclone) [B].</p
Acid stress response in Escherichia coli: mechanism of regulation of gadA transcription by RcsB and GadE
International audienceEscherichia coli can survive extreme acid stress for several hours. The most efficient acid resistance system is based on glutamate decarboxylation by the GadA and GadB decarboxylases and the import of glutamate via the GadC membrane protein. The expression of the corresponding genes is controlled by GadE, the central activator of glutamate-dependent acid resistance (GDAR). We have previously shown by genetic approaches that as well as GadE, the response regulator of the Rcs system, RcsB is absolutely required for control of gadA/BC transcription. In the presence of GadE, basal activity of RcsB stimulates the expression of gadA/BC, whereas activation of RcsB leads to general repression of the gad genes. We report here the results of various in vitro assays that show RcsB to regulate by direct binding to the gadA promoter region. Furthermore, activation of gadA transcription requires a GAD box and binding of an RcsB/GadE heterodimer. In addition, we have identified an RcsB box, which lies just upstream of the -10 element of gadA promoter and is involved in repression of this operon

Acoustical measurements on stages of nine U.S. concert halls
A measurement tour of nine U.S. concert halls included acoustical measurements on the stage of each hall. Two teams (from the National Research Council of Canada, and the Technical University of Denmark) made measurements of the acoustical quantities suggested by Gade: the ``support'' family of measures describing the acoustical conditions for the musicians on stage. This paper compares the results from the two measurement teams and discusses the influence of hall designs
Chiral, enantiopure aluminum(III) and titanium(IV) azatranes
Al-III and Ti-IV complexes of C-3-symmetric tetradentate trisamidoamme ligands with trigonal bipyramidal coordination geometry, containing chlorine or dialkylamido groups, or with a free coordination site in the apical position, have been synthesised by salt metathesis and amine elimination. Products with threefold symmetry were generally obtained for tetravalent titanium, whereas for the aluminium complexes either asymmetric structures with two of the three podand arms taking part in coordination to the metal or symmetric arrangements possessing the full threefold symmetry were formed depending on the steric properties of the ligands. Interaction between infectious diseases and personality traits: ACP1*C as a potential mediator
In geographical regions characterized by high pathogen prevalence, it has been shown that human populations tend to be characterized by lower levels of extraversion (E) and openness to experience (OtE). According to the "behavioral immune system" hypothesis, the reduction of extraversion and openness levels represents a behavioral defense against infections. Like the 'classical' immune system, the "behavioral immune system" could also be shaped by its underlying genetic background. Previous studies have shown that the *C allele of the ACP1 gene confers increased susceptibility to infectious/parasitic diseases. We hypothesized that carriers of the ACP1*C allele should likewise be associated with reduced E and OtE. We tested this hypothesis using two samples comprised of 153 students from Southern California (Group 1), and 162 female subjects recruited from an executive health program (Group 2), genotyped for ACP1 polymorphism and evaluated by the NEO Five-Factor Inventory (NEO-FFI). ACP1 was significantly associated with E: we found that carriers of ACP1*C showed reduced scores for E (Group 1: β= -4.263, P= 0.027; Group 2: β= -8.315, P= 0.003; Group 1. +. Group 2: β= -5.366, P= 0.001). Across groups, ACP1 was only marginally associated with OtE. In conclusion, the present study found that the ACP1*C allele, previously associated with an increased vulnerability to infectious/parasitic diseases may also be able to shape behavioral immune defenses by interaction with the level of E. © 2014 Elsevier B.V
