    Mutations in the nonstructural protein 3A confer resistance to the novel enterovirus replication inhibitor TTP-8307.

    Contains fulltext : 79778.pdf (Publisher’s version ) (Open Access)A novel compound, TTP-8307, was identified as a potent inhibitor of the replication of several rhino- and enteroviruses. TTP-8307 inhibits viral RNA synthesis in a dose-dependent manner, without affecting polyprotein synthesis and/or processing. Drug-resistant variants of coxsackievirus B3 were all shown to carry at least one amino acid mutation in the nonstructural protein 3A. In particular, three mutations located in a nonstructured region preceding the hydrophobic domain (V45A, I54F, and H57Y) appeared to contribute to the drug-resistant phenotype. This region has previously been identified as a hot sport for mutations that resulted in resistance to enviroxime, the sole 3A-targeting enterovirus inhibitor reported thus far. This was corroborated by the fact that TTP-8307 and enviroxime proved cross-resistant. It is hypothesized that TTP-8307 and enviroxime disrupt proper interactions of 3A(B) with other viral or cellular proteins that are required for efficient replication

    Heparin and a semisynthetic mucopolysaccharide (R01-8307) cause a fall in aldosterone production by both the human and rat adrenal, and narrowing of zona glomerulosa width in the rat. We have shown in studies of R01-8307-treated rats that the fall in aldosterone production precedes histological changes in the zona glomerulosa. Corticosterone production was not altered, suggesting that pituitary adrenocorticotropin production and the function of the inner cortical zones were not affected. Direct addition of R01-8307 to incubating rat adrenal quarters did not alter either aldosterone or corticosterone production. </jats:p

    Measuring the poverty impact of ACIAR projects: a broad framework

    This report sets out some broad ideas about how poverty evaluation could be conducted for ACIAR research projects. As with good benefit–cost analysis, there are good practices that need to be observed when undertaking poverty analysis. While poverty is a broad concept, and can be addressed through many means, these need to be grounded in some common understanding of the economics of poverty. This report is concerned mostly with quantitative evaluation, in the same sense that current ACIAR project evaluations are quantitative. That is, it is concerned with saying something about the order of magnitude of the effects of the project. Of course, qualitative analysis is important, and in most cases is a prelude to quantification — there is little point quantifying if you don’t understand what you are talking about. Quantification, however, provides a discipline and focus for qualitative speculation and provides an important extra dimension when comparing the effects of different projects. When quantifying, there are many sensible approaches that could be adopted. We will focus here on approaches that are broadly consistent with the current approaches to benefit–cost analysis and that could readily be used to augment those approaches. The report begins by reviewing some basic notions of poverty (Chapter 2) and then goes on (Chapter 3) to discuss in principle the ways that agricultural research could influence poverty.     Uncovering oxysterol-binding protein (OSBP) as a target of the anti-enteroviral compound TTP-8307

    The genus Enterovirus (e.g. poliovirus, coxsackievirus, rhinovirus) of the Picornaviridae family of positive-strand RNA viruses includes many important pathogens linked to a range of acute and chronic diseases for which no approved antiviral therapy is available. Targeting a step in the life cycle that is highly conserved provides an attractive strategy for developing broad-range inhibitors of enterovirus infection. A step that is currently explored as a target for the development of antivirals is the formation of replication organelles, which support replication of the viral genome. To build replication organelles, enteroviruses rewire cellular machinery and hijack lipid homeostasis pathways. For example, enteroviruses exploit the PI4KIIIβ-PI4P-OSBP pathway to direct cholesterol to replication organelles. Here, we uncover that TTP-8307, a known enterovirus replication inhibitor, acts through the PI4KIIIβ-PI4P-OSBP pathway by directly inhibiting OSBP activity. However, despite a shared mechanism of TTP-8307 with established OSBP inhibitors (itraconazole and OSW-1), we identify a number of notable differences between these compounds. The antiviral activity of TTP-8307 extends to other viruses that require OSBP, namely the picornavirus encephalomyocarditis virus and the flavivirus hepatitis C virus

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     In his article, “Vector Autoregressions as a Tool for Forecast Evaluation,” Roy H. Webb proposes that VAR forecasts be used as a standard of comparison for other forecasts. He begins by explaining how conventional forecasting models are constructed and used, and summarizes a few common objections to these models. He then describes the VAR methodology and compares forecasts from a simple VAR model with those from a consulting firm that uses a conventional model and with a series of consensus forecasts. The VAR model holds its own in this competition; in fact, only the VAR model is able to predict the 1981-1982 recession one year before its occurrence.Forecasting
