Understanding of the regulatory mechanism on the activity of chloroplast-type ATP synthase by X-ray crystallography
葉緑体型ATP合成酵素の部分複合体であるα3β3γに存在する挿入配列とε阻害の関係を構造生物学的側面から理解するために、α3β3γεのX線結晶構造解析に取り組んだ。これまで、結晶化に適した変異α3β3γεの作成に成功し、結晶を得ることができたが、高分解能のデータはまだ得られていない。
また、葉緑体型ATP合成酵素の活性制御に重要な挿入配列がどのように遠く離れた触媒部位であるβサブユニットに影響しているかを生化学的手法により検証し、γサブユニット上のN末端およびC末端のα-ヘリックスのコイルドコイル構造の相対的なズレが、挿入配列による活性制御効果をβサブユニットに伝達している可能性が示唆された。Inserted region on the gamma subunit of chloroplast type ATP synthase strongly affect to the inhibitory effect of endogenous epsilon subunit (epsilon inhibition). To understand the relationship between the inserted region and the epsilon subunit, we try to dissolve structure of sub-complex of chloroplast type ATP synthase (alpha3beta3gamma1epsilon1). Though we could not yet get crystal in high resolution, we succeeded to make stable sub-complex mutant for crystallization and could get some crystal.
We also investigated an affect of the inserted region on the gamma subunit to the ATPase activity on the beta subunit because these two parts exist far form each other. Regulatory mechanism of the activity of chloroplast ATP synthase by the ε subunit
本研究では葉緑体型ATP合成酵素の活性制御に重要なεサブユニットのC末端側α-ヘリックス部分およびγサブユニット上の調節領域部分に注目し、これらの重要性を生化学的、生理学的に調べた。その結果、これらの調節機構によるATP合成酵素のATP加水分解活性の抑制が、ATPが合成されない暗所下での細胞内ATP量の維持に必要であることを明らかにした。Physiological effects of C-terminal α-helices of the ε subunit and regulatory region of the γ subunit of chloroplast-type ATP synthase were investigated. Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
A novel morphological pattern of stress-dependent intracellular and extracellular vesicles
Since the antioxidant enzyme peroxiredochin 2 (Prx2) associates with negatively charged phospholipids to form vesicles, the principal investigator examined the role of Prx2 in the intracellular (outer) vesicular transport process by using of biochemical and cell biological methods. We investigated that the cellular localization relationship of Prx2 and intracellular and extracellular vesicles. According to the results, we revealed the possibility that Prx2 is actively involved in the formation and localization of intracellular and extracellular vesicles through its negatively charged phospholipid binding ability. Furthermore, high-speed atomic force microscopy observation revealed that Prx2 interacts with lipids by recognizing not only electrostatic interactions between positively charged amino acids on Prx2 and negatively charged phospholipids, but also the curvature of the lipid membrane.研究代表者らは、抗酸化酵素ペルオキシレドキン2(Prx2)が負電荷リン脂質と会合して小胞を形成することから、Prx2の細胞内(外)小胞輸送プロセスに対する役割を生化学的、細胞生物学的手法を用いて探索した。Prx2と細胞内外小胞の細胞内局在性を検討した結果、Prx2は、その負電荷リン脂質結合能によって細胞内外小胞の形成や局在に積極的に関与している可能性を明らかにすることができた。さらに、高速原子間力顕微鏡観察により、Prx2は、Prx2上の正電荷アミノ酸と負電荷リン脂質による静電的な相互作用だけでなく、脂質膜の曲率も認識して互いに相互作用することを明らかにした。研究課題/領域番号:19K06596, 研究期間(年度):2019-04-01 - 2022-03-31出典:研究課題「ストレス依存的な細胞内外小胞の新規形態様式」課題番号19K06596

Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more

We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
