Nerve function and regeneration in diabetic rats: effects of ZD-7155, an AT1 receptor antagonist
Effects of the angiotensin II AT1 receptor antagonist ZD-7155 on nerve function, blood flow, capillarization, oxygenation, and regenerative capacity after injury were studied in streptozocin-diabetic rats. Deficits in saphenous sensory and sciatic motor conduction velocity measured after 1 or 2 mo of diabetes in anesthetized rats were prevented and corrected by ZD-7155. Sciatic resistance to hypoxic conduction failure, which was increased by 71% by 2 mo of diabetes, was attenuated by 39% with ZD-7155. Endoneurial capillary density, which was unaffected by diabetes, was increased by 34% with 2 mo of ZD-7155 treatment. Sciatic nutritive endoneurial blood flow, which was reduced by 45% by 2 mo of diabetes, remained in the nondiabetic range with ZD-7155. Mean endoneurial O2 tension was reduced 38% by diabetes, which was attenuated by ZD-7155. Punctate freeze damage of sciatic nerve caused complete fiber degeneration. Fourteen days postlesion, there was a 26% deficit in myelinated fiber regeneration distance after 2 mo of diabetes, which was prevented by ZD-7155 treatment from diabetes induction. Thus alterations in the renin-angiotensin system contribute to the neurovascular etiology of nerve dysfunction in experimental diabetes. </jats:p
Comparative Cardiovascular Effects of the Angiotensin II Type 1 Receptor Antagonists ZD 7155 and Losartan in the Rat
Abstract
Binding experiments show that ZD 7155 is a potent angiotensin II type 1 receptor antagonist. In this study this novel substance was studied in normotensive and hypertensive rats.
The relative potency and duration of the antihypertensive effects of ZD 7155 were compared with those of the reference substance, losartan. The inhibitory effects of both compounds on angiotensin II-induced pressor actions were studied in the conscious normotensive Sprague-Dawley (SD) rat and in the conscious, spontaneously hypertensive rat (SHR). Arterial blood pressure and heart rate (HR) were obtained by direct intraarterial recording. Angiotensin II infusion was administered intravenously in the dose range 53.3 ng—12.8 μg kg−1 min−1 to the conscious rats. ZD 7155 was administered in a bolus dose of 1.082 μmol kg−1 (0.51 mg kg−1) and losartan in bolus doses of 2.165 and 6.495 μmol kg−1 (1.0 and 3.0 mg kg−1). In conscious SD rats, ZD 7155 and losartan behaved as competitive antagonists and the pressor response curve to angiotensin II was shifted to the right. Experiments in conscious SD rats also showed that ZD 7155 was approximately ten times as potent as losartan in suppressing the angiotensin II-induced pressor response (240 ng kg−1; 10 min infusion). In addition, experiments with conscious rats demonstrated that ZD 7155 could suppress the angiotensin II-induced pressor response for approximately 24 h when ZD 7155 was administered intravenously in a 1.082 μmol kg−1 bolus dose and angiotensin II was given at 240 ng kg−1 (in a 10−min infusion). Experiments in conscious SHRs using ZD 7155 (1.082 μmol kg−1) and losartan (6.495 μmol kg−1) as intravenous boluses indicated that both ZD 7155 and the reference compound losartan exhibited a significant antihypertensive effect. These results demonstrate that ZD 7155 is a potent angiotensin II-type 1 antagonist which is approximately ten times as potent as losartan in suppressing the angiotensin II-induced pressor response. Furthermore, ZD 7155 may suppress the angiotensin II-induced pressor response for 24 h and in the SHR ZD 7155 induces a pronounced and persistent antihypertensive effect.</jats:p
Tobramycin-Treated Pseudomonas aeruginosa PA14 Enhances Streptococcus constellatus 7155 Biofilm Formation in a Cystic Fibrosis Model System
Cystic fibrosis (CF) is a human genetic disorder which results in a lung environment that is highly conducive to chronic microbial infection. Over the past decade, deep-sequencing studies have demonstrated that the CF lung can harbor a highly diverse polymicrobial community. We expanded our existing in vitro model of Pseudomonas aeruginosa biofilm formation on CF-derived airway cells to include this broader set of CF airway colonizers to investigate their contributions to CF lung disease, particularly as they relate to the antibiotic response of the population. Using this system, we identified an interspecies interaction between P. aeruginosa, a bacterium associated with declining lung function and worsening disease, and Streptococcus constellatus, a bacterium correlated with the onset of pulmonary exacerbations in CF patients. The growth rate and cytotoxicity of S. constellatus 7155 and P. aeruginosa PA14 were unchanged when grown together as mixed biofilms in the absence of antibiotics. However, the addition of tobramycin, the frontline maintenance therapy antibiotic for individuals with CF, to a mixed biofilm of S. constellatus 7155 and P. aeruginosa PA14 resulted in enhanced S. constellatus biofilm formation. Through a candidate genetic approach, we showed that P. aeruginosa rhamnolipids were reduced upon tobramycin exposure, allowing for S. constellatus 7155 biofilm enhancement, and monorhamnolipids were sufficient to reduce S. constellatus 7155 biofilm viability in the absence of tobramycin. While the findings presented here are specific to a biofilm of S. constellatus 7155 and P. aeruginosa PA14, they highlight the potential of polymicrobial interactions to impact antibiotic tolerance in unanticipated ways
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Teknolojideki gelişmeler, toplumsal hayatı etkilediği kadar toplumsal ilişkileri düzenleyen kuralları da etkilemektedir. Bu kapsamda icra ve iflâs hukuku, teknolojik gelişmelerin etkisinin büyük ölçüde görüldüğü hukuk dalları arasında yer almaktadır. Özellikle adalet hizmetlerinin elektronik ortamda yürütülmesine yönelik bir sistem olan UYAP, bilişim teknolojisindeki imkanlar çerçevesinde adeta bir evrim geçirmektedir. Bu sürecin bir aşaması da 7155 sayılı Kanun düzenlemesidir. Kanuna göre icra takibi elektronik ortamda UYAP ile entegrasyonu sağlanan MTS üzerinden başlatılacak ve belirli bir aşamaya dek elektronik ortamda yürütülecektir. Bilişim teknolojilerinin imkanlarıyla UYAP, icra takiplerinde kullanıcılar arasında iletişimi sağlayan elektronik bir ortam olmanın ötesine geçerek kullanıcılara cevap veren bir otomasyonu ifade etmektedir. UYAP dönüşümünün bir sonraki aşamasında ise bilgi teknolojisinin yarattığı olanaklarla otonom yapay zekânın sisteme entegrasyonu mümkün hale gelebilir. 7155 sayılı Kanun'da öngörülen icra prosedürü, 2004 sayılı İcra ve İflâs Kanunu ile benzerlikler gösterse de kendine özgü düzenlemeler barındırmaktadır. Çalışmada 7155 sayılı Kanun kapsamındaki icra takipleri incelenmekte ve özellikle sistemin yapısı ele alınmaktadır. In this context, enforcement and bankruptcy law is among the branches of law where the impact of technological developments is seen to a great extent. UYAP, which is a system especially for the execution of justice services in an electronic environment, is almost undergoing an evolution within the framework of the possibilities in information technologies. One step of this process is the regulation of Law No. 7155. According to the law, enforcement proceedings will be initiated electronically over the system integrated with UYAP and will be carried out electronically until a certain stage. With the possibilities of information technologies, UYAP goes beyond being an electronic environment that provides communication between users in execution proceedings and expresses an automation that responds to users. In the next stage of UYAP's transformation, the integration of autonomous artificial intelligence into the system may become possible with the opportunities created by information technology. Although the enforcement procedure in Law No. 7155 shows similarities with the enforcement procedure regarding money receivables in the Execution and Bankruptcy Law No. 2004, it has a unique content. In the study, enforcement proceedings within the scope of Law No. 7155 are examined and especially the structure of the system is discussed. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases.
