Abstract 5746: Circulating cytokine levels, Epstein-Barr viremia and risk of AIDS-related non-Hodgkin lymphoma
Abstract
BACKGROUND: Human immunodeficiency virus (HIV) infection may lead to cytokine dysregulation and decontrol of Epstein-Barr virus (EBV) latency as potential mechanisms for acquired immunodeficiency syndrome (AIDS)-related non-Hodgkin lymphoma (NHL). We therefore examined pre-diagnosis blood levels in AIDS-related NHL cases from three prospective cohort studies of HIV infection. METHODS: Plasma or serum were obtained from cases (n=66) 0.1 - 2.0 (median 1.0) years pre-NHL and from up to three HIV-positive lymphoma-free controls (n=186) sampled at comparable times, matched for cohort, age, race, sex and CD4 count at time of diagnosis. Cytokine measurements used Millipore 30-plex Luminex assays with values normalized by log-transformation; coefficients of variation for blinded replicate samples ranged from 10% - 46%. EBV free viremia was detected by real-time polymerase chain reaction (PCR) assays for the BNRF p143 gene; plasma samples originally collected with heparin anticoagulation (n=110) could not be assessed because of PCR inhibition. Statistical analyses were performed by conditional logistic regression for matched sets. RESULTS: Fifty-seven cases had at least one matched control, with mean (median) CD4 counts of 172 (86)/mm3 for cases and 174 (85)/mm3 for controls. Cases had significantly higher cytokine levels for nine of the 30 measured cytokines, with p<.01 for granulocyte-monocyte colony-stimulating factor (GM-CSF) and interleukin (IL)-15, and p<.05 for IL-1a, IL-4, IL-6, IL-8, IL-12 p40, IL-12 p70 and IL-13. Stepwise selection of a multivariable conditional model identified GM-CSF, IL-12p70 and IL-15 as the most predictive markers, with matched odds ratios (95% confidence intervals) of 1.5 (0.9 - 2.4), 1.5 (0.9 - 2.5) and 1.3 (0.9 - 1.9), respectively. EBV PCR-positivity had a matched odds ratio of 1.6 (0.5-5.0), based on serum and acid-citrate-dextrose plasma samples only. CONCLUSIONS: We found elevations of T-helper type-(Th)1, Th2 and pro-inflammatory cytokines and growth factors up to two years before AIDS-NHL; EBV viremia was not significantly predictive but statistical power was limited. Cytokine hyperstimulation of B-cell proliferation may be an important etiologic pathway to AIDS-related NHL.

Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Cross-protection of hSCARB2-Tg mice against the 5746 challenge by pre-immunization with live E59.
<p>(<b>A</b>) Surviving 1-d-old hSCARB2-Tg mice were primed with 1×10<sup>7</sup> pfu of live E59 s.c. (○) or medium (mock) (•) and then challenged with 3×10<sup>4</sup> pfu of 5746 s.c. after 7 d. Twelve mice in the live E59 pre-immunized group and 10 mice in the mock-treated group were examined. Logrank test was used for statistical analysis. (<b>B</b>) Group of transgenic animals received immunization and/or challenge of EV71 was illustrated. One-day-old hSCARB2-Tg mice were immunized with 10<sup>7</sup> pfu of live E59 or mock and then challenged on Day 21 of birth with 3×10<sup>4</sup> pfu 5746 or mock. In another group, 7-d-old hSCARB2-Tg mice immunized with E59 and challenged on Day 21 of birth with 5746 was included. (<b>C</b>) We harvested 5×10<sup>5</sup> splenocytes from individual mice on Day 28 of birth and subjected them to the IFN-γ and IL-4 ELISPOT assays that were described in the Materials and Methods. Data are representative of the results derived from 2 independent experiments, each with 4 mice per group.
