Analysis of novel transcription regulation mechanism of glyoxalase 1
これまでの研究により、glyoxalase 1(GLO1)遺伝子プロモーター領域近傍に新規遺伝子多型が存在することを新たに見出した。この新規遺伝子多型が、GLO1の転写を著しく亢進させることが明らかとなり、遺伝子多型の存在とGLO1酵素活性とは正の相関をしていた。また、この遺伝子多型DNAと特異的に結合するたんぱくタンパク質の同定にも成功し、当該タンパク質の存在が遺伝子多型の示す転写亢進に不可欠であることも明らかとなった。当該タンパク質の存在によってヒストンがアセチル化されることも明らかとなり、遺伝子多型による転写亢進にはエピゲノム調節が重要な働きをしていることが示唆された。A gene polymorphism in human glyoxalase 1 (GLO1) promoter region was newly identified and was found to significantly induce transcriptional expression of the GLO1 gene. The gene polymorphism was also positively correlated with the GLO1 enzyme activity. Furthermore, we identified associated proteins with the GLO1 gene polymorphism in the nuclear extracts, which is essential for transcriptional upregulation of GLO1 gene. A novel polymorphism-driven transcriptional regulation of human glyoxalase 1 gene
研究代表者が初めて見出したGlyoxalase 1 (GLO1)のプロモーター領域内の複数の遺伝子多型の中に、ヒトGLO1の転写活性を著しく亢進させる遺伝子多型が存在していた。この遺伝子多型の転写調節機構を解明すると、エピジェネティックな変化が関与していた。また、当該遺伝子多型と1型糖尿病患者の糖尿病血管合併症の発症進展の程度には相関があることも明らかとなり、当該遺伝子多型は糖尿病合併症のリスク予測因子や予防・治療の新規ターゲットになりうる。また、当該遺伝子多型の出現は環境因子に影響を受けることも初めて明らかとなった。他の遺伝子にも同様な調節機構が働いているのかどうか、今後の研究に期待したい。Detoxifying enzyme glyoxalase 1 (GLO1) is known to be involved in the development of diabetes and its complications. However, the transcriptional regulation of GLO1 is not yet fully understood. In this study, we have identified novel polymorphisms in the human GLO1 promoter region. One of the polymorphisms was found to significantly upregulate the transcription of GLO1. We next addressed to reveal molecular mechanisms under the upregulation of the GLO1 transcription. As a result, the recruitment of histone proteins and the epigenetic modifications were involved in the transcriptional regulation. Cell culture conditions such as high glucose concentration or normoxia could change the frequency of the polymorphisms of the GLO1 gene. The polymorphisms were also found to link to the development of advanced diabetic vascular complications. Understanding of pathophysiological mechanisms and development of the treatment of schizophrenia based on novel gene polymorphisms of glyoxalase 1.
Glyoxalase 1(GLO1)はこれまで統合失調症との関連が示唆されていたが、詳細は不明であった。本研究では、研究代表者らが新たに見出したGLO1プロモーター領域内に存在する遺伝子多型が統合失調症の病態形成に関与していることが、ヒト臨床検体だけでなく疾患iPS細胞を用いた検討でも示唆された。加えて、統合失調症の病態形成への関与が示唆される遺伝子変異をGLO1プロモーター領域内で新たに見出すことに成功した。この遺伝子変異は統合失調症特異的であり、この遺伝子変異に着目することで、統合失調症の病態形成の全貌を明らかにできる可能性がある。Glyoxalase 1 (GLO1) has been known to be associated with schizophrenia. However, the mechanism remains unknown. In this study, we revealed that novel gene polymorphisms in the human GLO1 promoter were involved in the pathogenesis of schizophrenia using human samples as well as iPS cells derived from the patients. In addition, we identified a novel gene mutation in human GLO1 promoter region in the patients with schizophrenia. The gene mutation was found to be specific to schizophrenia. Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
