    Estudo da atividade antiinflamatória dos ligantes do receptor periférico dos benzodiazepínicos: Pk11195 e Ro5-4864

No primeiro modelo o efeito antiinflamatório em de pré-tratamento com os ligantes PBR, PK11195 1-(2-chlorophenyl)-N-methyl-N(1-methylpropyl)-3-isoquinoline carboxamide e Ro5-4864 7-chloro-5-(4-Chlorophenyl)-1,3-dihydro-1-methyl-2-H-1,4-benzodiazepin-2, mostraram que a formação do edema em resposta a carragenina (300 mg/pata) foi inibida por PK11195 e Ro5-4864, em diferentes intervalos de tempo. O estudo do tempo resposta mostrou que a melhor resposta com ambos ligantes foi ótima em animais injetados no tempo de 24h, por esta razão este tempo de pré-tratamento foi escolhido para estudo da dose resposta. A formação do edema de pata em resposta a carragenina (300 mg/pata) foi reduzida por PK11195 e Ro5-4864 em diferentes doses (0,00001-10mg/kg, i.p.) produzindo uma inibição dose dependente na formação do edema de pata em camundongos induzida pela carragenina. PK11195 e Ro5-4864 (0.1 mg/kg, i.p.) quando administrado 24 h antes da indução do edema, inibiram de forma significativa o edema de pata de camundongos induzido por vários mediadores inflamatórios. No segundo modelo o pré-tratamento com os ligantes (0.1 mg/kg, i.p.), 24 h antes da indução da pleurisia induzida pela carragenina, mostrou uma significante inibição no recrutamento das células totais e diferenciais, principalmente a custas de neutrófilos, inibindo também a liberação de Interleucina-13 (IL-13) e Interleucina-6 (IL-6) no líquido pleura

    Ro5-4864 and PK11195, prototypical synthetic ligands of translocator protein 18 kDa (TSPO), have shown anti-inflammatory effects in several models of inflammatory diseases; however, their biochemical mechanisms remain poorly understood. Nod-like receptor family, pyrin domain containing 3 (NLRP3) inflammasome activation as a part of the innate immune system, has been implicated in a variety of inflammatory diseases. Here, we demonstrate for the first time that TSPO ligands, especially Ro5-4864, potently suppressed ATP-induced NLRP3 inflammasome activation in THP-1 and BMDM cells. Detailed action mechanism was further investigated in THP-1 cells. Ro5-4864 efficiently attenuated NLRP3 translocation to mitochondria, inflammasome assembly/oligomerization, activation of caspase-1, and subsequent secretion of the mature forms of interleukin-1β and -18. Ro5-4864 also reduced the production of mitochondrial superoxide and preserved the mitochondrial membrane potential in ATP-treated cells, suggesting that Ro5-4864 may act on mitochondria or more upstream targets in NLRP3 inflammasome signaling. We also observed the distinct effects of the TSPO ligands between THP-1 monocytes and macrophages, which suggested different NLRP3 inflammasome signaling depending on cell type. Collectively, our novel findings demonstrate that Ro5-4864 effectively inhibited ATP-induced NLRP3 inflammasome activation through the prevention of mitochondrial perturbation. Our results indicate Ro5-4864 as a promising candidate for the treatment of NLRP3 inflammasome-related diseases. © 2016 Elsevier Inc. All rights reserved

    Pro- and anti-convulsant drug effects in combination with the convulsant benzodiazepine Ro 5–4864

    Abstract The effects of several compounds believed to act at the GABA-benzodiazepine receptor complex and which have anticonvulsant or proconvulsant properties when administered in combination with picrotoxin and pentetrazol (leptazol, pentylenetetrazole) were investigated in combination with the convulsant benzodiazepine Ro 5–4864. Tracazolate (25–100 mg kg−1) failed to affect convulsions induced by Ro 5–4864; however, they were prevented by treatment with CL 218, 872 (20 mg kg−1). Compounds having proconvulsant activity in combination with a subthreshold dose of Ro 5–4864 were: CL 218,872 (5 mg kg−1), and CGS 8216 (20 mg kg−1) and FG 7142 (40 mg kg−1), two compounds characterized as ‘inverse agonists’; at benzodiazepine receptors. The phenylquinolines PK 8165 and PK 9084, originally believed to have anxiolytic properties, had no significant effect in combination with Ro 5–4864 (25–100 mg kg−1). The convulsant profile of Ro 5–4864 is compared with that of picrotoxin and pentetrazol.</jats:p

    Efeito antiinflamatório dos ligantes do receptor benzodiazepínico periférico (PK 11195 e Ro5-4864) no modelo de pleurisia induzida pela carragenina, em camundongos

Programa de Pós-Graduação em Farmácia.Estudos têm demonstrado o efeito antiinflamatório do PK11195 e do Ro5-4864 em modelos experimentais (TORRES et al., 1999; 2000). Estes dois fármacos são denominados ligantes do receptor benzodiazepínico periférico (RBP) estão localizados em diversos tecidos, em nível periférico. Neste estudo foi utilizado o modelo de pleurisia induzida pela carragenina (Cg, 1%/cav.), em camundongos. Este é um modelo pelo qual apresenta uma resposta inflamatória bifásica (4 e 48 h) importante para avaliação de novos fármacos com propriedades antiinflamatórias. Objetivos: 1) Avaliar o efeito do PK11195 e do Ro5-4864 sobre a resposta inflamatória induzida pela Cg na cavidade pleural de camundongos. Os parâmetros inflamatórios (leucócitos, exsudação, concentrações da mieloperoxidase (MPO), adenosina-deaminase (ADA), óxido nítrico (ONx) e permeabilidade vascular) foram estudados 4 e 48 h após a indução da pleurisia, 2) Avaliar se o possível efeito antiinflamatório dos fármacos estudados ocorre devido a síntese esteroidal. Materiais e métodos: Neste protocolo experimental, foram utilizados camundongos albinos suíços de ambos os sexos, pesando entre 18-22g. Todos os animais foram tratados previamente com solução de azul de Evans (25 mg/kg, i.v.) para análise indireta do exsudato, exceto para àqueles experimentos pelo qual, estudou-se as enzimas MPO e ADA, ONx e análise histológica. Inicialmente avaliou-se a curva dose, tempo e resposta sobre os parâmetros inflamatórios (leucócitos e exsudação). Após o estabelecimento da melhor dose e o melhor tempo de tratamento prévio dos animais com os fármacos em estudo, avaliou-se o efeito destes sobre as concentrações de MPO, ADA, ONx), tanto na primeira (4 h) como na segunda fase (48 h) da resposta inflamatória induzida pela carragenina na cavidade pleural de camundongos. Análise estatística: Os resultados obtidos com a contagem de leucócitos, exsudação, MPO, ADA, ONx e permeabilidade vascular foram expressos através da média ± e.p.m. A análise estatística para estes parâmetros foi realizada por meio do teste paramétrico ANOVA (Dunnett) complementado, quando necessário pelo teste t de student. A análise histológica foi expressa através de scores (0 = ausente; 1 = discreto; 2 = moderado e 3 = abundante). Para este último parâmetro a análise estatística utilizada foi o teste não paramétrico de Man-Whitney. Valores de P < 0,05 foram considerados significativos. Resultados: Na primeira fase (4 h) da resposta inflamatória induzida pela carragenina, o PK11195 e o Ro5-4864 inibiram significativamente o influxo de leucócitos (P < 0,01), às custas de neutrófilos (P < 0,01), a exsudação (P < 0,01), os níveis de ONx (P < 0,01) no lavado pleural e a permeabilidade vascular no pulmão (P < 0,01). Além disso, somente o Ro5-4864 foi capaz de inibir significativamente as concentrações da enzima MPO no lavado da cavidade pleural 4 h após (P < 0,05). Nesta fase, ambos os fármacos também inibiram os leucócitos no sangue dos animais inflamados (P < 0,01). A análise histológica revelou que o PK11195 e o Ro5-4864 inibiram de forma significativa a migração de neutrófilos, mononucleares e edema pulmonar (P < 0,01). No baço observou-se também a diminuição de leucócitos, neutrófilos na polpa vermelha, bem como diminuição da hiperplasia da polpa branca (P < 0,01). Neste último órgão, os fármacos estudados também aumentaram a apoptose (P < 0,01). Já no fígado, observou-se que o PK11195 e o Ro5-4864, diminuíram os neutrófilos e as células de Kuppfer (P < 0,01). Na segunda fase (48 h) da pleurisia induzida pela carragenina, o PK11195 e o Ro5-4864 inibiram de forma significativa os leucócitos (P < 0,01), às custas de mononucleares (P < 0,01), e os níveis de ADA no lavado pleural de xv camundongos inflamados (P < 0,01). A analise histológica revelou que ambos os fármacos inibiram os neutrófilos (P < 0,01) e os mononucleares, no pulmão (P < 0,01). No baço, tanto o PK11195 como o Ro5-4864 inibiram os neutrófilos e os mononucleares (P < 0,01), no entanto, somente o Ro5-4864 diminuiu de forma significativa o centro germinativo, neste órgão (P < 0,01). Já no fígado, observou-se que os dois fármacos diminuíram a hiperplasia de células de Küppfer (P < 0,01). Finalmente para avaliarmos o possível mecanismo de ação antiinflamatório do PK11195 e do Ro5-4864 utilizamos a DL-aminoglutatimida neste modelo experimental. Os resultados revelaram que este último fármaco reverteu o efeito antiinflamatório do PK11195 em relação a: exsudação, neutrófilos, mononucleares e níveis de ONx no lavado pleural (P < 0,01), na pleurisia induzida pela Cg 4 h. Nesta fase, este efeito ocorreu também em relação ao fármaco Ro5-4864 em relação a: neutrófilos, mononucleares, MPO e ONx (P < 0,01). Na pleurisia induzida pela Cg 48 h, a DL-aminoglutatimida reverteu os efeitos antiinflamatórios tanto do PK11195 como do Ro5-4864 em relação a: leucócitos, mononucleares, neutrófilos e exsudação na cavidade pleural de camundongos (P < 0,01). Além disso, a DL-aminoglutatimida reverteu também o efeito antiinflamatório do Ro5-4864 em relação aos níveis da ADA (P < 0,01). Conclusão: Estes resultados revelam que o efeito antiinflamatório tanto do PK11195 como do Ro5-4864 ocorre principalmente sobre o influxo de leucócitos ativados, bem como a inibição de enzimas pró-inflamatórios como MPO e ADA, além do mediador óxido nítrico. Além disso, parece que este efeito antiinflamatório é mediado pela síntese esteroidal. Estudos posteriores devem ser realizados para melhor elucidar o mecanismo de ação antiinflamatória dos ligantes do PBR

    Acute lung injury (ALI) is a syndrome of inflammation and increased permeability of the blood-gas barrier. It is associated with high morbidity and mortality. Despite intensive research, treatments remain limited. The aim of the present study was to investigate the protective efficacy of a specific peripheral benzodiazepine receptor ligand, Ro5-4864, in experimental models of ALI in rats.ALI was generated by four different methods: (1) intravenous (tail vein) injection of Escherichia coli (0111:B4) lipopolysaccaride (LPS), (2) cecal ligation and puncture (CLP), (3) mesenteric ischemia/reperfusion, and (4) intraperitoneal injection of α-naphthylthiourea (ANTU). Ro5-4864 was administered to rats intraperitoneally 30 min before ANTU and LPS administration or intravenously 15 min before reperfusion and CLP. The levels of pulmonary edema (lung weight/body weight ratio) and pleural effusion were measured, and the severity of ALI was scored (0-3).Ro5-4864 showed a dose-dependent and significant prophylactic effect on the ANTU-induced lung weight/body weight and pleural effusion/body weight ratios and histopathologic scores. Ro5-4864 also showed significant prophylactic effects against the LPS-induced lung weight/body weight ratio and histopathologic scores. Ro5-4864 significantly decreased the intra-alveolar edema and perialveolar hemorrhage scores in the CLP group. However, we found no prophylactic effect of Ro5-4864 on mesenteric ischemia/reperfusion-induced ALI at the dose used (2 mg/kg intraperitoneally).These results have demonstrated, for the first time, a protective effect of Ro5-4864 on experimental ALI induced by ANTU, LPS, and CLP. Ro5-4864 might be a useful therapeutic agent for lung diseases, including ALI, in intensive care patients

    [3H]Ro5-4864 benzodiazepine binding in the kainate lesioned striatum and Huntington's diseased basal ganglia

    [3H]Ro5-4864-labeled peripheral-type benzodiazepine binding sites in the brain were studied after kainic acid lesions of the rat striatum. Following intrastrial kainate injections [3H]Ro5-4864 binding increased to approximately 1000% of control over a period of 1 week and was maintained at this level for up to 6 weeks. Two weeks after lesioning the number of binding sites (Bmax) was selectively increased while the dissociation constant (Kd) was only minimally affected. [3H]Ro5-4864 binding in the Huntington's diseased (HD) basal ganglia was not changed as compared to non-neurological control in the caudate nucleus and globus pallidus. A highly significant 51% increase was found in the HD putamen. It is concluded that the peripheral-type, Ro5-4864-sensitive benzodiazepine receptor in the brain may be predominantly localized on glial element

    The postnatal development of susceptibility to the convulsant effects of Ro5-4864 (4\u27-chlorodiazepam) was characterized in two inbred mouse strains (DBA/2J and BALB/c ByJ) which as adults differ markedly in their response to this convulsant. Onset of susceptibility to a dose of Ro5-4864 which caused a high frequency of clonic seizures in adults was observed at 10 days of age in DBA/2 mice, but not until 35 days in BALB/c By mice. At 14 days of age an abrupt increase in susceptibility to Ro5-4864-induced tonic seizures was found in DBA/2 but not BALB/c By mice. Both the peak of tonic seizure susceptibility (21 days) and the time course of its subsequent age-dependent decline closely paralleled the change in audiogenic seizure susceptibility in the DBA/2 strain. PK11195 (40 mg/kg) blocked Ro5-4864 (25 mg/kg)-induced, age-dependent tonic seizures but had no effect on clonic seizure induction in the same mice. These observations establish that both the susceptibility to Ro5-4864 in adult mice and the postnatal time course for development of susceptibility to this convulsant are inherently different in these two strains of mice. The lack of coincidence between the developmental onset of susceptibility to Ro5-4864-induced seizures and the onset of supersensitivity to Ro5-4864-induced tonic seizures during the period of peak audiogenic seizure susceptibility in DBA/2 mice implies that more than one neurochemical mechanism is involved in the ability of Ro5-4864 to induce seizures in this strain. However, the blockade of Ro5-4864-induced tonic seizures by PK11195 suggests that peripheral type benzodiazepine receptors may mediate this effect

    Diazepam and its Anomalous p-Chloro-derivative Ro 5-4864: Comparative effects on mouse neurons in cell culture

    The actions of diazepam and its p-chloro-derivative Ro 5-4864 were compared on mouse spinal cord and dorsal root ganglion neurons in cell culture. Diazepam enhanced but Ro 5-4864 reduced iontophoretic GABA responses in a concentration-dependent manner. Both diazepam and Ro 5-4864 limited sustained, high frequency repetitive firing of spinal cord neurons but diazepam was more potent. Ro 5-4864 was, however, more potent than diazepam in inhibiting spontaneous neuronal activity of spinal cord neurons and reducing the duration of calcium-dependent action potentials of dorsal root ganglion neurons.
