    O impacto visual é elemento de decisão na escolha de um alimento e a indústria alimentícia utiliza-se de corantes para deixar os alimentos potencialmente mais atrativos. Porém, como grande parte dos corantes alimentares é derivada da indústria petroquímica e possuem na maioria das vezes efeitos indesejáveis, segue a formulação de leis no mundo que regulamentam o uso destes corantes e que protejem a saúde do consumidor. Além da função original de nutrição, alguns alimentos ajudam na prevenção e tratamento de doenças, sendo denominados de alimentos funcionais ou nutracêuticos. Seguindo a nova tendência do mercado alimentício em consumir corantes naturais e com componentes biologicamente ativos, buscou-se estudar neste trabalho alguns metabólitos secundários do fungo filamentoso Monascus ruber CCT 3802, todos oriundos da via de biossíntese dos policetídeos. Este microrganismo é tradicionalmente utilizado em países da União Européia, Estados Unidos, China e Japão por produzirem biopigmentos como o Monascus, através de fermentação do arroz; sendo este usado em carnes, peixes, bebidas, sorvetes e confeitaria. Entre outras moléculas bioativas produzidas por espécies de Monascus estáa enzima amilase, largamente utilizada nas indústrias de alimentos, farmacêuticas e de cosméticos, e a mevinolina (lovastatina), que vem sendo utilizada como complemento dietético no tratamento da hipercolesterolemia por inibir a biossíntese do colesterol. O presente trabalho busca enunciar as características metabólicas do fungo filamentoso Monascus ruber CCT 3802 àquelas relacionadas à produção de pigmentos vermelhos, amilase e lovastatina em diferentes substratos: glicose, amido, farelo de arroz, milhocina em cultivo submerso.As maiores concentrações de pigmento Monascus e biomassa foram obtidas com o amido e glutamato monossódico como fonte de carbono e nitrogênio respectivamente ficando na ordem de 4,98 UDO480 e 5,24 g/L enquanto que nos meios de cultivo estudados e técnicas utilizadas a lovastatina não foi detectada. Ainda no estudo, o Monascus produziu concentrações elevadas de amilase (35,22 UI).Abstract : The visual impact is element of decision in the choice of a food and the food industry used of colorants to leave the foods potentially more attractives. However, as great part of the alimentary colors they are derived of the industry petrochemical and they possess undesirable effects most of the time, it follows the formulation of laws in the world that regulate the use of these coloring and that protect the consumer's health. Besides the original function of nutrition, some foods help in the prevention and treatment of diseases, being denominated of functional foods or nutraceuticals. Following the new tendency of the nutritious market in consuming coloring natural and with components actives biologically , it was looked for to study in this work some secondary metabolites of the filamentous fungus Monascus ruber CCT 3802, all originating from the polyketides pathway. This microorganism is used traditionally at countries of the European Union, United States, China and Japan for they produce biopigments as Monascus, through fermentation of the rice; being this used in meats, fish, drunk, ice creams and sweet shop. Among other molecules bioactives produced by species of Monascus they are the enzyme amylase, largely used in the industries of foods, pharmaceuticals and of cosmetics, and the mevinolina (lovastatin), that has been used as dietary complement in the treatment of the hypercolesterolemia for inhibiting the biosynthesis of the cholesterol. The present work search to enunciate the metabolics characteristics of the filamentous fungus Monascus ruber CCT 3802 the those related to the production of red pigments, amylase and lovastatin in different substrates: glucose, starch, rice powder and corn steep liquor in cultivation submerged. The largest pigment concentrations Monascus and biomass were obtained with the starch and monosodium glutamate as source of carbon and nitrogen respectively being in the order of 4,98 UDO480 and 5,24 g/L while in the cultivation means studied and used techniques the lovastatin was not detected. Still in the study, Monascus produced high concentrations of amylase (35,22 UI)

    Exploring the Binding of Anticancer Drug BI-3802 with DNA Using Optical Tweezers

    BI-3802 is a relatively new anticancer drug known to bind to the transcription repressor BCL6 which can lead to the regression of the most common form of cancer in the lymphatic system, B-cell lymphoma. Based on the structure of BI-3802 we believe this small molecule drug can bind to DNA through intercalation. Intercalators are a special type of molecule which have a flat planar section that can stack between DNA base pairs. In this study, we use Optical tweezers to trap a single DNA molecule in order to explore the binding of BI-3802 with DNA. Once we trap a single DNA molecule, we stretch the DNA and measure the tension in the DNA as a function of extension. During this process, at a certain force, the DNA undergoes aphase transition from double stranded DNA (dsDNA) to single stranded DNA (ssDNA). DNA stretching is repeated in the presence of various concentrations of BI-3802 in order to study the interaction between the drug and DNA at the molecular level. Our results suggest that the force where the phase transition from dsDNA to ssDNA occurs is decreased as the concentration of BI-3802 is increased, indicating this drug destabilizes the naturally occurring double stranded structure of DNA at micromolar concentrations. To our knowledge this is the first study that explores the interactions of BI-3802 with DNA at the single molecule level. The outcome of these experiments may help us better understand drug interactions at the molecular level and help develop new therapeutics

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
